Chi onomidae axonomy. Using de ailed mo phome ic analysis (head capsule a ios,
mandibula pec en leng h, and s iae coun s) in conjunc ion wi h mi ochond ial COI
ba coding, we es ablished la al–adul associa ions and di e en ia ed hese species om
hei congene s. Mic o endipes baishanzuensis is cha ac e ized by a b ownish head capsule
wi h dis inc ly pale median ee h compa ed o he la e al ee h, he longes mandibula
pec en wi hin he genus, he highes numbe o body s iae, and an excep ionally an e io ly
posi ioned ing o gan. Mic o endipes obus us is dis inguished by a uni o mly da k b own
men um, ema kably wide en omen al pla es, he mos a iable s iae coun , and a medium-
sized mandibula pec en. Mic o endipes ube osus exhibi s a uni o mly da k b own men um
wi h median ee h conspicuously sho e han he second la e al ee h, he smalles body
size in he genus, he sho es mandibula pec en, he ewes s iae, and he mos pos e io ly
loca ed ing o gan. A e ised la al key o Chinese Mic o endipes is p esen ed, imp o ing
eshwa e biomoni o ing and add essing challenges associa ed wi h c yp ic di e si y.

The genus Mic o endipes Kie e was es ablished by Kie e in 1915 and cu en ly
comp ises o e 60 desc ibed species (F eeman and C ans on 1980; C ans on
and Ma in 1989; Ashe and C ans on 1990; Oli e e al. 1990; Zo ina 2001; Qi
and Wang 2006; Qi e al. 2014; Yamamo o and Yamamo o 2014; Haz a e al.
2016; Tang and Hi omi 2017; Song e al. 2023). In China, 18 eco ded species
ha e been desc ibed (Qi and Wang 2006; Qi e al. 2012, 2014; Song e al. 2023).
Taxonomic esea ch on Mic o endipes has p ima ily ocused on adul mo -
phology, and la al desc ip ions a e sca ce. This gap is la gely due o he high
mo phological homogenei y o la ae and he lack o eliable diagnos ic ai s,

es pe sis in specimen collec ion and analysis, equi ing specialized appa a us
and mic oscopic echniques, and da abase limi a ions hinde eliable la a–adul
ma ching. Despi e hese challenges, DNA ba coding has eme ged as an e ec i e
iden i ica ion ool (Ca ew e al. 2005, 2007; Gadawski e al. 2022).
DNA ba codes a e included in mos new species desc ip ions as a s an-
da d me hod o delimi ing non-bi ing midges (Song e al. 2016, 2022; Yan e
al. 2017; Lin e al. 2019). DNA ba coding has also p o en e ec i e o associ-
a ing specimens ac oss di e en li e s ages (Song e al. 2018). Howe e , he
la al s ages o hese species ha e emained undisco e ed. To add ess his
gap in he li e a u e, we conduc ed ex ensi e la al collec ions and pe o med
DNA ba code analyses. Consequen ly, we success ully collec ed and iden i ied
he la ae o h ee Mic o endipes species: M. baishanzuensis Song & Qi, 2023,
M. obus us Song & Qi, 2023, and M. ube osus Qi & Wang, 2006, p o iding he
i s desc ip ions o hei la al s ages.
Ma e ial and me hods
La al specimens we e collec ed om mul iple i e s and s eams using D-shaped
ne s. All specimens we e p ese ed in 75% e hanol and s o ed a −20 °C in he labo-
a o y un il subsequen mo phological and molecula analyses. Fo mo phological
examina ion, specimens we e slide-moun ed in Eupa al and examined unde a mi-
c oscope. Measu emen s ollowed s anda dized p o ocols, and mo phological e -
minology and abb e ia ions we e adop ed om Sæ he (1980) and Maschwi z and
Cook (2000). Values a e epo ed as anges accompanied by mean alues, wi h he
numbe o obse ed specimens (N) indica ed in pa en heses. All specimens a e
deposi ed in he College o Li e Sciences, Taizhou Uni e si y.
Genomic DNA was ex ac ed using he p o ocol ou lined by Song e al.
(2018). The s anda d ba code egion o he 5′ po ion o he mi ochond ial
gene cy och ome c oxidase I (COI-5P) was ampli ied using he uni e sal p im-
e s LCO1490 and HCO2198 (Folme e al. 1994). PCR ampli ica ions we e pe -
o med as desc ibed by Song e al. (2018). PCR p oduc s we e elec opho esed
on a 1.0% aga ose gel, pu i ied, and sequenced using an ABI 3730XL capilla y
sequence (Beijing Genomics Ins i u e Co., L d., Hangzhou, China). Raw se-
quences we e edi ed using BioEdi . 7.2.5 (Hall 1999).
Publicly a ailable Mic o endipes sequences (≥500 bp in leng h) we e e-
ie ed om he Ba code o Li e Da a Sys em (BOLD; h p://www.boldsys ems.
o g/) on June 20, 2024 (see Suppl. ma e ial 1 o de ails). Sequence alignmen
was conduc ed in MEGA . 12 (Kuma e al. 2024) using he Clus alW algo i hm.
Pai wise gene ic dis ances we e calcula ed using he K2P model in MEGA . 12.
A neighbo -joining (NJ) ee was cons uc ed based on he Kimu a 2-Pa ame e
(K2P) subs i u ion model wi h 1000 boo s ap eplica es, using he “pai wise
dele ion” op ion o handle missing da a. A maximum-likelihood (ML) ee was
gene a ed using IQ-TREE . 2.1.3 (Nguyen e al. 2015), wi h node suppo as-
sessed h ough ul a as boo s apping wi h 1000 eplica es. Bayesian in e -
ence (BI) analysis was conduc ed using M Bayes . 3.2.7 (Ronquis e al. 2012),
employing Ma ko chain Mon e Ca lo (MCMC) andomiza ion o 10 million

Resul s and discussion
DNA ba codes analysis
This s udy analyzed a o al o 161 sequences, including eigh newly ob ained la al
sequences. A clea ba code gap o 4–6% was obse ed (Fig. 1), con i ming he
e ec i eness o DNA ba coding o species delimi a ion wi hin his genus. Based
on h ee known male specimens, he maximum in aspeci ic gene ic dis ance was
0.7% (mean: 0.5%) o M. baishanzuensis, 5.8% (mean: 2.9%) o M. obus us, and
3.2% (mean: 1.4%) o M. ube osus. A e including la al DNA sequences, hese
alues changed sligh ly: M. baishanzuensis exhibi ed a maximum o 0.9% (mean:
0.5%), M. obus us a maximum o 5.8% (mean: 2.5%), and M. ube osus a maxi-
mum o 6.1% (mean: 2.6%) (Suppl. ma e ial 2). No ably, he maximum in aspeci ic
dis ance obse ed in M. ube osus (6.1%) emained below he minimum in e spe-
ci ic dis ance (8.0%) be ween M. baishanzuensis and M. obus us.

Phylogene ic analyses (Neighbo -joining, maximum likelihood, and Bayesian in-
e ence) e ealed s ong conco dance be ween la al and adul sequences (Fig. 2).
Fo M. ube osus, al hough mino opological a ia ions occu ed ac oss ees, all
h ee me hods p o ided obus suppo o he same clade s uc u e. In M. obus-
us, consis en esul s ac oss all h ee me hods (NJ/ML/BI) con i med la al–adul
conspeci ici y ia DNA ba coding. Fo M. baishanzuensis, all h ee phylogene ic
econs uc ions yielded well- esol ed, cong uen clades. This high consis ency in
b anching pa e ns, combined wi h he low maximum pai wise gene ic dis ance
(0.9%), conclusi ely iden i ies he la ae as M. baishanzuensis using DNA ba coding.

Rema ks. A compa a i e analysis was conduc ed be ween ou da a and he
la al mo phological measu emen s epo ed by Tang (2006). The la ae o
M. baishanzuensis esemble Mic o endipes pedellus (De Gee , 1929) and Mic o-
endipes chlo is (Meigen, 1933) in he shape o he men um. Howe e , M. bais-
hanzuensis has mo e s iae han M. pedellus (mean 37.5 s. 30.0). The o al
leng h o he pos men um in M. baishanzuensis is sho e han in M. pedellus
(mean 201.3 μm s. 223.0 μm). The ing o gan in M. baishanzuensis is loca -
ed a 0.2–0.3 (mean 0.267) o he basal segmen , compa ed o app oxima ely
0.33. Compa ed o M. chlo is, M. baishanzuensis has a dis inc ly sho e blade
(mean 115.5 μm s. 145.0 μm), a longe pos men um (mean 201.3 μm s.
215.0 μm), and mo e s iae (mean 37.5 s. 28.0).

o M. ube osus esemble hose o Mic o endipes b i eni (Edwa ds, 1983) in
men um mo phology. Howe e , M. ube osus is no iceably smalle in body
size (mean 2.8 mm s. 6.9 mm in M. b i eni) and has a sho e mandibu-
la pec en (mean 153.3 μm s. 195.0 μm in M. b i eni). Among congene s,
M. ube osus is no ably smalle o e all. Bo h i s median and la e al ee h a e
uni o mly da k b own.
