Pu pose o he s udy: Nowadays, much a en ion is paid o amino acids as biologically ac i e
subs ances ha can be used in medical p ac ice. The aim o ou esea ch was o s udy he composi ion
and amoun o ee amino acids in he ui s o he bu naki plan - Lu a echina a ohb., which is
widely used in olk medicine.
Me hods and echniques: he composi ion and amoun o ee amino acids in he ui s o he
bu naki plan was s udied by p ecipi a ing p o eins and pep ides in he aqueous ex ac o he samples
in cen i uge ubes. Fo his, 20% ichlo oace ic acid was added o 1 ml (p ecise olume) o he
sample unde s udy. A e 10 minu es, he p ecipi a e was sepa a ed by cen i uga ion a 8000 pm
o 15 minu es. A e sepa a ing 0.1 ml o he supe na an , i was lyophilized. The hyd olyza e was
e apo a ed, he d y esidue was dissol ed in a mix u e o ie hylamine-ace oni ile-wa e (1:7:1) and
d ied. This p ocess was epea ed wice o neu alize he acids. Acco ding o he me hod o S ephen
A., Cohen Da iel, phenyl hioca bamyl de i a i es (FTK) o amino acids we e ob ained by eac ion
wi h phenyl hioisocyana e. The de e mina ion o amino acid de i a i es was ca ied ou by high-
pe o mance liquid ch oma og aphy (HPLC). HPLC condi ions: Agilen Technologies 1200
ch oma og aphy wi h DAD de ec o , column 75x4.6 mm Disco e y HS C18. Solu ion A: 0.14 M
CH3COONa + 0.05% TEA pH 6.4, B: CH3CN. Flow a e 1.2 ml/min, abso p ion 269 nm. G adien
%B/min: 1-6%/0-2.5 min; 6-30%/2.51-40 min; 30-60%/40.1-45 min; 60-60%/45.1-50 min; 60-
0%/50.1-55 min.
Resul s: The composi ion o amino acids was de e mined by hei ligh abso p ion ime, and
he amoun (mg/g) was de e mined by he a ea o he peaks. Acco ding o he esul s o he s udy, he
composi ion and amoun o ee amino acids (mg/g) in he composi ion o bu naki ui s a e in he
ollowing dec easing o de : Glu amic acid (1.421) > P oline (1.397) > Alanine (1.153) > Th eonine
(1.048) > Phenylalanine (0.849) > His idine (0.648) > Aspa ic acid (0.54) > Se ine (0.427) >
Glu amine (0.378) > Ty osine (0.346) > Glycine (0.314) > Aspa agine (0.304) > Leucine (0.26) >
Me hionine (0.233) > Valine (0.232) > Lysine (0.212) > Isoleucine (0.191) > T yp ophan (0.133) >
A ginine (0.126) > Cys eine (0.121).
Conclusions: The esul s o he s udy show ha bu naki ui s con ain 20 ypes o amino acids,
he o al amoun o which is 10,334 mg/g. 9 o he amino acids a e essen ial amino acids, which
accoun o 36.83% o he o al amoun o amino acids. These a e: Leucine - pa icipa es in he
egula ion o blood suga le els, issue epai and p o ein syn hesis. Isoleucine - suppo s ene gy
me abolism and muscle issue eco e y. Valine - is impo an o muscle ene gy and he ne ous
