DETERMINATION OF THE MONOSACCHARIDE COMPOSITION BY PAPER

The aim o his s udy was o de e mine he monosaccha ide composi ion o he ex ac s
ob ained om he ollowing plan s: Aco us calamus L., Helich ysi ma acandicum Pop ex Ki p., and
Ae a lana a L. using pape ch oma og aphy.
Ma e ials and me hods: Di e en polysaccha ide g oups we e sepa a ed using he abo e-
desc ibed me hod. Ch oma og aphic analysis e ealed ha he alcohol-soluble suga s consis ed o
uc ose, glucose, and suc ose. Wa e -soluble polysaccha ides we e ex ac ed using wa e ex ac ion.
Fi s , wa e -soluble polysaccha ides (WSP) we e isola ed, ollowed by he sequen ial isola ion o
pec in subs ances (PS) and hemicelluloses (HMC). Be o e analysis, he aw ma e ials we e cleaned
o o eign subs ances. One hund ed g ams o c ushed aw ma e ials (Aco us calamus L. ( oo ),
Helich ysi ma acandicum Pop ex Ki p. A. lana a L.( lowe s) and Ae a lana a L. (abo e-g ound
pa s), and h ee es mix u es we e ea ed wice wi h a ho mix u e o me hanol and chlo o o m (1:1)
o emo e dyes and non-ca bohyd a e componen s. The aw ma e ials we e hen sepa a ed by
il a ion and d ied a e wa d. The isola ed subs ances we e analyzed using pape ch oma og aphy
(PC) Fil ac-FN 13, 18 (Ge many). The ollowing sol en s we e used in he ollowing a io: n-
bu anol-py idine-wa e (6:4:3). The ollowing eagen s we e used o opening: 1) acid ph hala e
aniline (5 min, a 100°C), and 2) 5% u ea solu ion.
Resul s: The d ied aw ma e ial was ex ac ed wice wi h ho e hyl alcohol (1:6) a 82 °C o 1
h. The alcohol ex ac s we e combined, e apo a ed, and analyzed ch oma og aphically in sys em 1,
esul ing in he de ec ion o aces o glucose, suc ose, and uc ose. A e comple e acid hyd olysis
o WSP, PS, and HMC, 100 mg o he isola ed polysaccha ides we e hyd olyzed wi h 3 ml o 1n
H2SO4 solu ion a 100°C: WSP o 8 h and PS and HMC o 24 h. A e he speci ied ime, he
hyd olysa e was placed in a beake and neu alized wi h ba ium ca bona e. The esul ing p ecipi a e
was il e ed, and he il a e was deionized wi h KU-2 ca ioni e, e apo a ed o a small olume (0.5
ml), and ch oma og aphed on FN-18 pape in an n-bu anol-py idine-wa e (6:4:3) sys em wi h
speci ic monosaccha ides (con ol samples). A e d ying, he ch oma og ams we e ea ed wi h acid
aniline ph hala e and hea ed in an o en a 110°C o 1-2 minu es. Galac ose, a abinose, and glucose
we e ound in he monosaccha ide composi ion o he polysaccha ides.
Conclusion: The monosaccha ide composi ion o polysaccha ides in he ex ac s ob ained om
he collec ion o plan s Aco us calamus L., Helich ysi ma acandicum Pop ex Ki p., and Ae a lana a
L. was s udied using pape ch oma og aphy. Gi en ha hei composi ion is e y simila o ha o
