10). Recen me agenomic s udies ca ied ou in he sal e ns in

he highly saline ponds (c ys allize s), analysis o he me age-
nomic da a se o an in e media e-salini y pond wi h 19% sal s
indica ed ha a much wide p oka yo ic di e si y han expec ed
was p esen (11). In his epo , we desc ibe he sequencing o a

salini y o a ma ine sal e n loca ed in Isla C is ina, Huel a, sou h-
wes Spain, on he A lan ic Ocean, a away om he San a Pola
sal e n, which is on he Medi e anean coas .
A 50-li e wa e sample was ob ained om he su ace o a
pond wi h 21% o al sal s (Global Posi ioning Sys em [GPS] co-
o dina es 37°13=08⬙N, 7°20=17⬙W) on July 2010. The sample was
sequen ially il e ed h ough 5.0- and 0.22-
m-po e size polyca -
bona e il e s (Millipo e) as p e iously desc ibed (11). To al en i-
onmen al DNA was ex ac ed and pu i ied ollowing he p o o-
cols desc ibed elsewhe e (11,12). The pu i ied p oka yo ic DNA
was sequenced by py osequencing (Roche 454 GS-FLX sys em,
Ti anium chemis y) by GATC, Cons ance, Ge many. A o al o
1,223,923 eads wi h an a e age ead leng h o 397 bp we e ob-
ained. The size o he me agenomic da a se was 486,317,437 bp.
Sequences ela ed o 16S ibosomal RNA genes we e iden i ied
using he Ribosomal Da abase P ojec (RDP) (13). In addi ion,
sequences we e compa ed agains he MG-RAST se e (14),
Communi y Cybe in as uc u e o Ad anced Mic obial Ecology
Resea ch and Analysis (CAMERA) (15), and In eg a ed Mic obial
Genomes wi h Mic obiome Samples-Expe Re iew (IMG/MER)
(16) da abases.
Eu ya chaeo a (~84%) was he mos abundan phylum p esen in
his sample, ollowed by Bac e oide es (~8%) and Gammap o eobac-
e ia (~7%), sugges ing a dec ease o he mic obial di e si y in sal
pondso sal e nswhen hesalini y eachessal concen a ions⬎20%,
being mo e simila o hose ound in c ys allize ponds han hose
ound in in e media e-salini y ponds wi h ⬍20% sal s.
