Abs ac
En oloma is one o he op h ee gene a in species di e si y o Aga icomyco ina. In his
s udy, i e species om Qilian Moun ain in China we e documen ed. One new species E.
g iseopileum belonging o subg. Lep onia was in oduced based on mo phological and
phylogene ic analyses. This new species is cha ac e ized by i s g ay basidiomes wi h
ib illose pileus and wood-decaying li es yle. Fou known species a e E. glaucobasis, E.
incanum, and E. e ae in subg. Cyanula, and E. uligineocine eum in subg. Nolanea. All
i e species a e documen ed wi h desc ip ions, pho o pla es, and molecula da a includ-
ing ITS, LSU, and m SSU sequences.

En oloma (F .) P. Kumm., ypi ied by E. sinua um (Bull.) P. Kumm., is one o he
op h ee gene a in species di e si y o Aga icomyco ina. I is es ima ed ha
he e a e mo e han 1800 species o En oloma wo ldwide (He e al. 2019a). The
gene al mo phological cha ac e is ics o En olomaa e:basidiomes aga icoid,
seco ioid o gas e oid; aga icoid habi a iable as icholoma oid, mycenoid, col-
lybioid, cli ocyboid, omphalioid o pleu o oid; pink spo e p in o pinkish b own,
lamellae almos ee, adnexed o adna e, adna e-ema gina e o adna e-decu -
en , spo es angula in all iews which is en i ely co e ed wi h ace s delinea ed
by idges; clamp-connec ions o en absen ; cheilocys idia a e; pileipellis a i-
able (cu is, ichode m, hymenide m) wi h di e se pigmen a ion(Noo deloos
1981; La gen 1986). Mos species o his genus a e sap o ophic, bu he e
a e also some myco hizal o weakly pa asi ic axa. Fo example, membe s o
sec ions En oloma and Rhodopolia a e ec omyco hizal, and Rosaceae-asso-
cia ed membe s o sec ion Nolanidea a e conside ed weakly pa asi ic (Age e
and Walle 1993; Tede soo e al. 2010).

A obus molecula -based axonomic sys em o En oloma is no es ablished
ye . T adi ional axonomic s udies di ided En oloma in o 13 subgene a based
on mo phological cha ac e is ics, such as basidiome o ms, pileipellis, basidio-
spo es, cys idia, and clamp connec ions (Noo deloos and Ga es 2012a; Kaygu-
suz e al. 2024). Ano he opinion ea ed hese subgene a as gene a (Aime e al.
2010, Ka s ed e al. 2020; Ni ei o e al. 2021). En oloma cons i u es he domi-
nan species di e si y wi hin En oloma aceae. The i s comp ehensi e molecu-
la phylogene ic s udy o En oloma aceae indica ed he e a e wo main clades o
En oloma, basal and c own clades. The basal clade is equal o P unuloides clade
and c own clade is composed o Rhodopoloid clade, Inocephalus-Cyanula clade,
and Nolanea-Claudopus. Subgenus Lep onia is ound o be polyphyle ic and dis-
ibu ed in wo di e en subclades in he c own clade (Co-Da id e al. 2009).
In he pas decades, mos axonomic s udies o En oloma a e ocused on Ame -
ican and Eu opean a eas while species di e si y o En oloma in China is ela i ely
poo ly known. In ecen yea s, species di e si y o En oloma in China has been
explo ed.The e a e many new species o En oloma in oduced om China based
on mo phological and molecula da a (He e al. 2012, 2013, 2015, 2019b; Chen e
al. 2024, 2025). The Qilian Moun ains, loca ed be ween Qinghai and Gansu p o -
inces in no hwes China, is a c i ical biodi e si y co ido connec ing no he n
and sou he n China. Du ing ou mac o ungi di e si y su eys in Qilian Moun ains,
se e al no able samples o En oloma we e collec ed. In his s udy, based on mo -
phological and mul igene phylogene ic analyses, we iden i ied and documen ed
i e En oloma species including one new o science in subgenus Lep onia.
Ma e ials and me hods
Mo phological examina ion
Specimens we e collec ed du ing he ainy season (July o Augus ). Pho o-
g aphs we e aken immedia ely in he ield. Basidiomes we e w apped in alu-
minium oil o pu in plas ic boxes sepa a ely. Mac o mo phological cha ac e -
is ics and habi in o ma ion we e eco ded when specimens we e esh. E e y
specimen was d ied in an elec ical ood d ie a 40 °C, hen kep in a plas ic zip-
lock bag and deposi ed in He ba ium MycologicumAcademiaeSinicae (HMAS).
Ana omical and cy ological cha ac e is ics we e obse ed using d y specimens
unde he ligh mic oscope (SOPTOP EX33). Obse ed cha ac e is ics included
basidiospo es, basidia, cys idia and pileipellis. A leas 20 measu emen s we e
made o each cha ac e is ic. Measu emen s we e analyzed and eco ded as
X= he mean o leng h by wid h ± SD, Q = he quo ien o basidiospo e leng h o
wid h, and Qm = he mean o Q alues ± SD. All he p o ocols o mo phological
s udy ollowed La gen ’s me hodology (La gen 1986).
DNA ex ac ion and PCR
Genomic DNA o each specimen was ex ac ed by ab oad-spec um plan Rapid
Genomic DNA Ki ollowed he manu ac u e ’s p o ocol. Th ee genes we e ap-
plied in he phylogene ic analyses iz. in e nal ansc ibed space (ITS), la ge
ibosomal subuni (LSU), and mi ochond ial ibosomal small subuni (m SSU).
The p ime s o each gene a e ITS4/ITS5, LR5/LROR, and NS1/NS4, espec i ely.

we e sequenced in Sangon Bio ech (Shanghai) Co., L d. Bo h di ec ions we e
sequenced o ensu e accu acy o each gene.
Phylogene ic analyses
In o ma ion o e e ence and new gene a ed sequences we e lis ed in Suppl. ma-
e ial 1. New gene a ed sequences we e checked in Geneious P ime .2025.0.3.
Alignmen s we e made using Muscle o each gene sepa a ely (Edga 2004), hen
adjus ed by hand o emo e he ambiguous egion. The da ase was made o ge-
nus En oloma. The En olomada ase includes 72ITS, 53LSU, and 43m SSU. Max-
imum likelihood (ML) analysis and boo s ap alues calcula ion we e pe o med
in axmlGUI 1.5b1 wi h GTRGAMMA model wi h 1000 eplica es (Sil es o and
Michalak 2012). ModelFinde 2.2.0 (Kalyaanamoo hy e al. 2017) was used o
selec he bes - i pa i ion model (Edge-linked) using AIC c i e ion. In he En olo-
ma da ase , Bes - i model acco ding o AIC a e GTR+F+I+G4 o ITS, GTR+F+I+G4
o LSU, and GTR+F+I+G4 o m SSU. Bayesian In e ence (BI) analysis was pe -
o med by M Bayes 3.1.2. Ten million gene a ions we e un o six Ma ko chains
and sampled e e y 1000 h gene a ion. Bu n-ins was de e mined in T ace 1.6 wi h
e ec i e sample sizes (ESS) highe han 200 (h p:// ee.bio.ed.ac.uk/so wa e/
ace ). Remaining ees we e used o calcula e Bayesian pos e io p obabili ies
(PP). Maximum likelihood (ML) analysis and boo s ap alues calcula ion we e
pe o med in axmlGUI 1.5b1 wi h GTRGAMMA model wi h 1000 eplica es (Sil-
es o and Michalak 2012). Phylogene ic ees we e p esen ed in Fig. 1.
Phylogene ic esul s
The ML ee o En oloma is p esen ed in Fig. 1. The e a e 75 samples in ol ed ep-
esen ing 48 species o En oloma and one ou g oup species Cli opilus hi neolus.
These 48 species ep esen wel e subgene a o En oloma. Samples collec ed
om Qilian Moun ains dis ibu ed in h ee subgene a, iz. subg. Cyanula, subg.
Nolanea, subg. Lep onia. In he Cyanula clade, h ee samples om Qilian Moun-
ains clus e ed in E. incanum, E. e ae,and E. glaucobasis clades wi h ull boo s ap
alues. Fou samples clus e ed in E. uligineocine eum clade in subg. Nolanea
wi h ull boo s ap and PP alues. Th ee samples clus e ed in En oloma and sis-
e o he E. allingio um wi h he s a is ical alues PP/BS = 1.0/99.Th ee Qinghai

o E. glaucobasisin he phylogene ic ee (Fig. 1). This species is o iginally
desc ibed om Ge many. Ou sample gene ally coincides wi h he o iginal de-
sc ip ion o E. glaucobasis. Di e ence is ha he cheilocys idia is basidia-like
in ou sample while is cla a e o esicula acco ding o he o iginal desc ip ion
(Noo deloos 1985).
