Ma e ials and me hods: 1 g o chi osan hyd ochlo ide (molecula weigh 230.0 kDa, deg ee
o deace yla ion 85%) was dispe sed in 100 mL o e hanol and allowed o swell o 2 hou s. A e
swelling, a low-molecula -weigh eagen was added a a mola a io o chi osan o cyanoguanidine
= 1:1-4. The eac ion mix u e was s i ed o 5-10 hou s a 60-80°C. Upon comple ion, he p ecipi a e
o med was sepa a ed by decan a ion, dissol ed in 1% hyd ochlo ic acid, and ep ecipi a ed wi h
ace one. The esul ing solid was edissol ed in wa e and pu i ied by dialysis o 48 hou s, wi h i e
changes o dialysis wa e . Finally, he pu i ied p oduc was subjec ed o lyophiliza ion and analyzed.
Resul s: Upon eac ion o cyanoguanidine wi h he amino g oups o chi osan, a nucleophilic
addi ion occu s, leading o he o ma ion o guanidinium moie ies. The conduc ed s udies
demons a ed ha by a ying he eac ion condi ions speci ically he mola a io o eac an s, eac ion
ime, and empe a u e i is possible o ob ain chi osan de i a i es wi h a deg ee o subs i u ion anging
om 0.15 o 0.64 mol%. The p esence o guanidinium g oups in he chi osan backbone was
con i med by FTIR and NMR spec oscopy. In oduc ion o guanidinium agmen s in o he
mac omolecula chain signi ican ly imp o ed he solubili y o chi osan ac oss a wide pH ange. While
na i e chi osan is insoluble in neu al and alkaline media, he syn hesized de i a i es emained
soluble wi hin he pH ange o 3 o 9.
In addi ion, he included guanidine g oups inc eased he o e all cha ge o chi osan (38 mV)
and he ζ-po en ial alue was 45-58 mV.This inc ease in posi i e cha ge may ha e a di ec impac on
he physiological p ope ies o chi osan. Fo ins ance, s onge elec os a ic in e ac ions wi h cell
memb anes could enhance bo h i s an imic obial ac i i y and i s cell adhesion capaci y, he eby
b oadening he po en ial biomedical applica ions o guanidinium- unc ionalized chi osan.
Conclusions: Thus, he syn hesized supe cha ged chi osan de i a i es con aining guanidinium
g oups exhibi imp o ed physicochemical cha ac e is ics, including an expanded solubili y ange and
