Stress Effect of Food Matrices on Viability of Probiotic Cells during Model Digestion

Abstract

The aim of this study was to evaluate the influence of model (alcohol, sugar, salt, protein and acid) and real foods and beverages on the viability of probiotics during incubation and artificial digestion. Viability of monocultures Lactobacillus acidophilus CCM4833 and Bifidobacterium breve CCM7825T, and a commercial mixture of 9 probiotic bacterial strains, was tested by cultivation assay and flow cytometry. In model foods, the best viability was determined in the presence of 0.2 g/L glucose, 10% albumin and 10% ethanol. As the most suitable real food for probiotic survival, complex protein and carbohydrate substrates were found, such as beef broth, potato salad with pork, chicken with rice, chocolate spread, porridge and yoghurt. The best liquid was milk and meat broth, followed by Coca-Cola, beer and coffee. Viability of probiotics was higher when consumed with meals than with beverages only. Addition of prebiotics increased the viability of probiotics, especially in presence of instant and fast foods. Generally, the highest viability of probiotics during artificial digestion was observed in mixed culture in the presence of protein, sugar and fat, or their combination. The increase of cell viability observed in such foods during model digestion may further contribute to the positive effect of probiotics on human health.

2. Ma e ials and Me hods
2.1. Ma e ial
2.1.1. P obio ic S ains
Bac e ial s ains Lac obacillus acidophilus CCM 4833 and Bi idobac e ium b e e CCM 7825T
used in his s udy we e pu chased om he Czech Collec ion o Mic oo ganisms in B no,
Czech Republic.
The comme cial p epa a ion Biop on 9 (Walma k, L d., T inec, Czech Republic) was
selec ed as an op imum sou ce o mixed p obio ic s ains. This ood supplemen con ained a
mix u e o 9 bac e ial cul u es o Bi idobac e ium bi idum,Bi idobac e ium b e e,Bi idobac e ium

2.1.2. Food Ma ices
All used samples o eal oods a e summa ized in Table 1. The se o eal samples
should ep esen ypical oods and be e ages consumed daily by mos o he popula ion.
Be e ages, ins an oods (ins an soups, pas a wi h c eam sauce, po idge) and snacks
(yoghu , pudding, chocola e sp ead, pas y, poppy seed cake, chips, po a o salad) we e
ob ained om local e ail. Ins an oods we e mixed wi h ho wa e o milk acco ding o he
p oduce s’ ecommenda ions. Homemade p oduc s (mixed ege able b o h, chicken b o h,
bee b o h, boiled chicken mea wi h ice, po a o salad wi h ied po k) we e p epa ed
immedia ely be o e expe imen s om na u al ma e ials (300 g) boiled in wa e (1 L) wi h
3 g o sal . Hambu ge was ecei ed om as ood e ail. F ui s and ege ables we e used
as mix u es o oma o, cucumbe and peppe , wi h he a io o 1:1:1.

2.2.2. Model Food Ma ices
As model ood ma ices, solu ions wi h a ious concen a ions o alcohol (e hanol
5–40%), suga (0.2–20 g/L o glucose), sal (0.4–10 g/L o NaCl) and p o ein (bo ine se um
albumin 5–20 g/L) we e p epa ed in s e ile dis illed wa e . As a model o acidic ood,
s e ile dis illed wa e adjus ed o pH = 3 by HCl was used. S anda d and pu e chemicals
we e pu chased om Sigma Ald ich (Sigma-Ald ich, Me ck KGaA, Da ms ad , Ge many).
All used model oods a e summa ized in Table 2.

2.3. Model Diges ion
A i icial s omach juice was p epa ed om 0.25 g o pepsin dissol ed in 100 mL o
dis illed wa e . To his solu ion, 0.84 mL o 35% hyd ochlo ic acid was added. Final pH
was adjus ed o 0.9. A i icial panc ea ic luid was p epa ed wi h 0.25 g o panc ea in and
1.5 g o sodium hyd ogen ca bona e in 100 mL o dis illed wa e (pH = 8.9). Bile luid was
composed o 0.4 g o bile acid sal s dissol ed in 100 mL o phospha e bu e . P obio ics we e
added o 300 g o be e age, liquid ood o solid ood homogenized in an app op ia e wa e
olume. A mix u e o p obio ic cells (1.10

simila ly o GIT condi ions. Model diges ion was se as a sho ened con inuous p ocess.
2.4. De e mina ion o Cell Viabili y by Cul i a ion Me hod and Flow Cy ome y
The iabili y o p obio ic bac e ia was es ed by cul i a ion assay on Pe i dishes wi h
MRS medium by he o e low me hod. A e 48 h o cul i a ion, colony- o ming uni s,
e.g., bac e ial colonies (CFU/mL), we e coun ed. The cul i a ion me hod was used o
e alua ion o all ypes o oods and be e ages, including model oods.
The iabili y o p obio ic bac e ia exposed o model and eal liquid oods and be e -
ages was also ollowed by di ec analysis o cell iabili y using low cy ome y (Apogee
Flow Sys ems, Hemel Hemps ead, UK). As a luo escen p obe, p opidium iodide (Sigma-
Ald ich, Da ms ad , Ge many) was used. Fo analysis, 100

dilu ed o 1 mL and added o p opidium iodide. A e 5 min o incuba ion in da kness,

diges ion, hen a e 20 min incuba ion in s omach juice (pH 2.1), ollowed by cell coun ing
a e alkaliza ion and mixing wi h he mix u e o panc ea ic and bile juices and a he end
o he diges ion p ocess (pH 7.9, 2 h). Acidi y o he en i onmen (pH) was checked du ing
he whole diges ion p ocess.
2.6. Viabili y o Mix u e o P obio ic Cells in Di e en Types o Foods and Be e ages wi h/wi hou
Addi ion o P ebio ics
P obio ics om a comme cial capsule we e e-hyd a ed o 20 min and solubilized in
s e ile dis illed wa e . Then, cells we e mixed wi h eal ood ma ix and wi h o wi hou
he addi ion o p ebio ics (300 mg o inulin, Sigma-Ald ich, Me ck KGaA, Da ms ad ,
Ge many). The ecommended daily dose o p obio ics mix u e (1 capsule, 1.10

CFU) was
suspended in 300 g o homogenized ood o be e age. This p epa ed mix u e was hen
incuba ed a 37

C o 15 min. Then, samples we e aken o analysis o he numbe o

model ood ma ixes can be conside ed as s e ile (see Sec ion 2.2.2). Real oods we e
p epa ed immedia ely be o e incuba ion and, hus, he p esence o o he mic oo ganisms
was minimized. Each o he es ed p obio ics samples we e i s mixed wi h ood/be e age
and incuba ed o 15 min. Then, he samples we e exposed o model diges ion acco ding
o Sec ion 2.3. In egula in e als (a e o du ing incuba ion), he amoun o li ing cells
was measu ed. The iabili y o p obio ics du ing simula ed gas oin es inal condi ions
we e pe o med using cul i a ion me hods and low cy ome y (liquid samples). Cul i-
a ion me hods we e used o all he e ogenic samples, and o homogenous solu ions, i
was possible o de e mine hem di ec ly by FC. The e alua ion o iabili y and CFU in
indi idual model and eal oods a e complica ed by con inuous g ow h o p obio ic cells in
any en i onmen . Thus, he iabili y a e diges ion (o indi idual s ages) was compa ed
o he alue o CFU a he beginning o model diges ion, a e sho - ime incuba ion wi h
he ood.
3. Resul s
3.1. Viabili y o P obio ics in Model Food Ma ices
The aim o he p esen s udy was o e alua e he iabili y o es ed p obio ic cul u es
in he p esence o selec ed model and eal oods and be e ages in model condi ions o he
diges i e ac . As a model ood, solu ions wi h a ious concen a ions o alcohol, suga ,
sal , p o ein and ace ic acid we e p epa ed. Real oods we e selec ed acco ding o he mos
common die a y p e e ences in he popula ion.
In all model oods excep acidic ood, he pH alue was se o pH 7, and he model acid
ood is o pH 3. In Table 3, pH alues in es ed eal be e ages and oods a e p esen ed. Wi h
he excep ion o juice (pH 3) and bee (pH 5.1), all es ed be e ages and oods exhibi ed
pH in he ange o 6.0–7.3. Thus, we canno expec some d ama ic in luence o pH o es ed
ood ma ixes on iabili y o p obio ic cells. Howe e , a mo e acidic pH (app oxima ely
5.5–6.5) will p obably be mo e sui able o p obio ic bac e ia g ow h.

The in luence o basic model oods on concen a ion and iabili y o p obio ic cells
du ing a i icial diges ion was s udied (Figu e 1A–F). As he p obio ic cul u e, a mixed
comme cial p epa a i e was used. The measu emen was pe o med using low cy ome y,
and as model oods wa e (wa e -based ood), ace ic acid solu ion o pH 3 (acid ood),
glucose (swee ood), p o ein (p o ein-based ood), sodium chlo ide (sal ed ood) and
e hanol (alcohol-based ood) a di e en concen a ions (see Table 2) we e used. The
e alua ion o iabili y and CFU in indi idual model and eal oods is complica ed by
he con inuous g ow h o p obio ic cells in any en i onmen , hus, in some samples, an
inc ease o CFU a e model diges ion was obse ed. I is necessa y o no e ha he numbe

signi ican inc ease o cell concen a ion in sample wi h he addi ion o 10 g/L o bo ine
albumin was obse ed. In his sample, abou 3 imes mo e han he o iginal numbe o

Fu he , he e ec o model ood wi h glucose on cell g ow h du ing diges ion was
moni o ed (Figu e 1A). The highes inc ease o cell numbe was eco ded in he sample
wi h 2 g/L o glucose. A e wo hou s o model diges ion, abou 5 imes highe CFU in
his sample was de e mined. Finally, he numbe o cells in acidic model ood was es ed
(Figu e 1E). A he beginning o he in es inal s age, a dec ease o he numbe o cells in
acidic ood was obse ed. Howe e , a e wo hou s o exposi ion o he in es inal juice,
he CFU numbe o acidic ood was al eady 2 imes highe .
Gene ally, acco ding o o al CFU numbe s in model oods, he mos sui able en i-
onmen s o p obio ic p epa a i e in ake we e ecognized as: (i) wa e , (ii) p o ein a he
concen a ion o 10 g/L, (iii) glucose a 2 g/L and low e hanol concen a ion (5–10%). The
mos nega i e e ec was exhibi ed p edominan ly by sal , which was p obably caused by
osmo ic s ess, simila o he e ec o highe glucose concen a ions. A nega i e e ec o
e hanol as a bac e ios a ic agen was obse ed a e long- e m exposi ion o highe concen-
a ions (20% and mo e), while lowe concen a ions o e hanol exhibi ed posi i e e ec s
on p obio ic iabili y. The eason could be a c oss- esponse o he ex e nal s ess e ec o
e hanol and p obably also an abili y o some p obio ics o use e hanol as an addi ional
ca bon sou ce.
3.1.2. Flow Cy ome y De e mina ion o Viabili y o P obio ics Exposed o Model
Diges ion in Liquid En i onmen s
In he ollowing sec ion, he in luence o model oods on he CFU numbe and ia-
bili y o p obio ic monocul u es o Lac obacillus acidophilus and Bi idobac e ium b e e a e
model diges ion was s udied (Figu e 2). Fo compa ison, a mixed p obio ic cul u e was
used. The scheme o he expe imen is desc ibed in Sec ion 2.5. A e a i icial diges ion
in s omach juice ollowed by incuba ion in in es inal juices, he CFU numbe was di ec ly
de e mined by low cy ome y. A he end o incuba ion, he cul u e o Lac obacillus aci-
dophilus exhibi ed a high inc ease o CFU p edominan ly in model ood con aining p o ein,
bu also sligh ly in o he model oods (Figu e 2A). Con e sely, he s ain Bi idobac e ium
b e e g ew in ensi ely, p edominan ly in model oods con aining saccha ide (Figu e 2B). In
he comme cial p obio ic mix u e, he highes inc ease o CFU numbe was ound a e he
model diges ion in he p esence o wa e a pH = 7 and in he p esence o acidic solu ion
(pH = 3). Fu he mo e, he CFU inc ease was de ec ed in oods con aining p o ein and 10%
alcohol as well (Figu e 2C).

Resul s a e illus a ed in Figu e 3. When compa ed wi h model oods, monocul u es
as well as mixed p obio ics cul u e exhibi ed simila ends, co esponding o some o he
ypical model oods, such as p o ein (mea b o h) o suga ( ege able b o h). The bes
en i onmen o all es ed samples was chicken b o h, con aining high concen a ions o
p o eins. Excep o in his en i onmen , monocul u es g ew qui e slowly (app oxima ely
4–5x lowe biomass o ma ion) when compa ed wi h he mixed cul u e. G ow h o mixed
cul u e in mea ex ac was lowe han in he p esence o 10 g/L o p o ein.
