Besides, it is considered to be the cheapest source of proteins available worldwide, which may help solve numerous feeding and agronomical problems. On top of that, soybeanbased products such as okara are a potential source of bioactive peptides that can be obtained using proteolytic enzymes. These peptides are of particular interest due to their antihypertensive, antioxidant, anticancer, antimicrobial or antidiabetic properties (Agyei, 2015;Puchalska et al., 2014a,2014b,2017). Several studies have analyzed the antioxidant properties (Yokomizo et al., 2002) and health-promoting effects of bioactive peptides (Jim enez-Escrig et al., 2010;Nishibori et al., 2017) and fiber (Jim enez-Escrig et al., 2008) derived from okara with promising results both in vitro and in animal models. The extraction of isoflavones using alcohol has been proposed by other authors and is maximal in theory (Jankowiak el al., 2014b) but expensive and not practical. Therefore, using water as extractant is preferred (Jankowiak et al., 2014a) despite the lower yield. In this article we hypothesized that the direct enzymatic hydrolysis of crude okara in aqueous solutions would enhance the extraction of isoflavones while also favoring the generation of bioactive peptides, thus contributing to the affordable formulation of a new product with higher antioxidant potency and increased value for the food and fertilizer industry. 2. Materials and methods 2.1. Okara and enzymes Okara from the same production batch was used for all experiments and it was supplied by Soria Natural S.L. (Garray, Soria, Spain). Briefly, soybeans were washed to eliminate impurities, soaked in cold water for 30 h at 33.3% (w/v) and heated up at 95 C for 5 min to inactivate any tripsin inhibitors and lipoxygenase present. Next, the grains were grinded in the presence of hot water at a 1:1 proportion (w/v) and soymilk was obtained by pressure and filtration, with the remaining by-product being the soy pulp (okara). Okara hydrolysates were made using a liquid formulation of the serine-endoprotease subtilisin (EC 3.4.21.62) from Biocon (Les Franqueses del Valles, Barcelona, Spain) with an enzyme activity of 100,000 AU/g. The enzyme was stored at 4 C. 2.2. Analysis of the chemical composition of okara The chemical composition of the okara used was analyzed by the Spanish Association for Standardisation and Certification (AENOR) laboratory (Madrid, Spain). The ash content was analysed according to standard AOAC methods (2016). The protein load was determined using the Kjeldahl procedure (AOAC, 2016) and multiplying the total nitrogen content in a protein by the conversion factor 5.80, which is applicable to most plant proteins. Crude fat was determined gravimetrically after extraction with hexane for 12 h in a Soxhlet extractor (Clemente et al., 1997). Total soluble carbohydrates were determined after extraction with a mixture of ethanol/water (2/3) for 2 h. After centrifugation at 4,000 g (MiniSpin, Eppendorf), the supernatant was filtered through a no. 1 Whatman paper and total soluble sugars were estimated colorimetrically at 490 nm (GeneQuant 1300, General Electric) by the phenol-sulphuric acid method, using a standard curve of glucose (Dubois et al., 1956). Organic matter was determined by the dry combustion method (MAPA, 1986). 2.3. Preparation of okara extracts Crude okara was diluted in water to a final concentration of 10% (dry w/v). Enzymatic hydrolysis using water as extractant was performed at pH 4.0, 6.0, 8.0 and 10.0 according to the pH-stat method (Brocklehurst, 2002). Conditions relative to the amount of enzymes, enzyme type, times of incubation and temperatures used were optimized in preliminary experiments not shown here. Briefly, incubation with 0.3% of liquid subtilisin (v/v) for 2 h at 55 C without stirring was chosen. This temperature is optimal for the subtilisin activity and kills most of the microorganisms present in the sample, thus deferring spoilage while contributing to the solubilization process. It has been described that after one hour at that temperature subtilisin retains 90% of its activity (Sigma Aldrich, 1996) and enzyme variants with increased stability have been developed too (Takagi et al., 1990). The samples were later centrifuged for 40 min at 4 C and 10,000 g (Avanti J-26XP, Beckman-Coulter). The soluble part (hereafter also referred as hydrolyzed okara) was heat dried and analyzed, while the pellet was weighted and discarded. As negative controls, we performed the same extractions without any enzyme, using water only (hereafter also referred as soluble okara). 2.4. Protein quantification Protein determinations were performed according to Bradford (1976), using bovine serum albumin as standard. 2.5. Analysis of the molecular weight of soluble proteins The protein content of the soluble fraction of okara was studied by size-exclusion chromatography using an € AKTA-purifier FPLC system (GE Healthcare), filtration chromatography and a Superdex Peptide 10/ 300GL column, with an exclusion range between 700 and 10,000 Da that discriminates free peptides and amino acids. Samples were centrifuged at 12,000 g for 30 min at room temperature (MiniSpin, Epperdorf) to remove insoluble molecules, and the supernatant was passed through a 0.2 μ mfilter and loaded into a 0.1 mL loop connected to an € AKTA-purifier system. The column was equilibrated, and eluted with 0.25 M Tris-HCl buffer (pH 7.0) in isocratic mode, at a flow-rate of 0.5 mL/min, and proteins/peptides were detected at 280 and 215 nm with a GE Healthcare UV900 module coupled to the column elution. 2.6. Analysis of the amino acid composition The amino acid composition was determined by reversed-phase HPLC analysis of 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC) derivatives using γ-aminobutyric acid as the internal standard. The samples were hydrolyzed using 6N HCl/1% (w/v) phenol vapor at 110 C for 24 h in a vacuum (Bidlingmeyer et al., 1984). The amino acids were treated with AQC to form AQC derivatives using the commercial method described by Waters AccQ-Tag, which were then analyzed using a Waters HPLC system (Millipore Ltd) fitted with a reversed-APAse C18 column. For cysteine estimation, aliquots were first oxidized with performic acid and analyzed as above. 2.7. Analysis of isoflavones by UHPLC-MS/MS Isoflavones were analyzed using a Thermo Scientific liquid chromatography system consisting of a quaternary UHPLC Dionex Ultimate 3000 SD, connected to a quadrupole-Orbitrap Qexactive hybrid mass spectrometer with HESI ionization probe. Xcalibur software was used for instrument control and data acquisition. Separation was performed on a C18 column Xbridge BEH (2.1 100 mm, 2.5 μ m) (Waters). The injection volume was 5 μ L and the flow rate was 0.4 mL/min. Two different solvents were used as mobile phase: solvent A (water with 0.1% formic acid) and solvent B (acetonitrile with 0.1% formic acid). The following gradient was used: 0–1 min 5% of solvent B, 1–10 min from 5% to 100% of solvent B, 10–12 min 100% of solvent B, then 5% of solvent B up to 15 min. The Parallel Reaction Monitoring (PRM) was acquired in positive mode and a resolution of 17500 at m/z 200 FWHM, with an isolation window of 1 m/z. The HESI source parameters were as follows: spray voltage 3.5 kV, S lens level 50, capillary temperature of 320 C and probe heater temperature of 400 C, sheath flow, auxiliary flow and sweep gas flow (N 2 )of 50, 13 and 2 arbitrary units, respectively. Antioxidant capacity assays The ability of okara samples to reduce Fe þ3 to Fe þ2 was determined by the FRAP (ferric reducing antioxidant power) assay (Benzie and Strain, 1996) at pH 3.6. Absorbance was measured at 734 nm and Trolox (Merck Millipore) was used for the standard curve. 2.9. Statistical analysis Three independent experiments were performed and measured by triplicate using the same okara batch. Data was analyzed by a one-way analysis of variance (ANOVA) using Statgraphics Centurion XVIIII followed by Tukey's post-hoc test for multiple comparisons in order to confirm differences among the groups. A p-value <0.001 was considered significant. 3. Results and discussion 3.1. Chemical composition of okara The chemical composition of crude okara can vary depending on the soybean variety and method of obtention during soymilk production. Therefore, a prior characterization of the dried product was performed as shown in Table 1. In our case, crude okara contained 76.5% of humidity, which is in agreement with the literature and predisposes okara to fast spoilage. On the other hand, proteins represented around 40% of the total dry weight. However, around 90% of them were insoluble, reflecting their poor bioavailability. When soluble proteins were analyzed by exclusion chromatography (Fig. 1,Table 2), about 90% of them corresponded to proteins of more than 10 kDa, while almost 7% were small peptides and free amino acids. The whole amino acid composition of dried okara (which includes both soluble and insoluble protein content) was further analyzed (Table 3). Demonstrating the high potential value of its protein, we observed that nearly 30% of the amino acids found were essential for the human diet. Among these, branched-chain amino acids (BCAA) represented 15% of the total, and their presence in diets may have beneficial effects on the ageing process, muscle synthesis, body weight and glucose homeostasis (Bifari and Nisoli, 2017). Aromatic amino acids summed a 6% and most remarkably, hydrophobic amino acids counted for around one third of all amino acids detected. Most soluble proteins are removed during soymilk production. These proteins contain less than 25% of hydrophobic amino acids (Puchalska et al., 2017). Therefore, it is expected that proteins and peptides with a higher content of hydrophobic amino acids are retained in okara. Interestingly, the prevalence of hydrophobic amino acids in bioactive peptides has been related to their antioxidant properties (Kitts and Weiler, 2003; Puchalska et al., 2017;Samardi and Ismail, 2010;Zhang et al., 2012). Altogether, these values reflect the high potential for generating bioactive peptides from okara. Carbohydrates comprised nearly 30% of the total dry weight and for the most part were insoluble fiber. On the other hand, fats accounted nearly one fourth of the total weight (23.6%) and the most abundant oligoelements were K, P, S, Mg and Ca. Despite okara is rich in organic molecules, most of its proteins, carbohydrates and biomolecules embedded inside are insoluble, which hampers its use as a source of functional molecules. For this reason, we reasoned that the enzymatic hydrolysis would enable the extraction of compounds with functional activity, such as isoflavones, bioactive peptides and amino acids, hence contributing to the formulation of a new product with added value. 3.2. Chemical composition of the water extract and subtilisin extract of okara The chemical composition of the soluble extracts (treated with subtilisin or only with water) was analyzed after heat drying the samples (Table 1). Taking into account that the extraction efficiency at pH 10.0 was 30.4% for the subtilisin treatment and 12.6% for the water control Table 1 Chemical composition of okara (dry matter) used. As expected, no significant differences were observed in the extraction of carbohydrates (22.9% with subtilisin vs. 21.4% with water) and fats (2.9% with subtilisin vs 2.5% with water). 3.3. Effect of pH and subtilisin treatment on the solubilization of okara Unless stated otherwise, previous experiments were carried out at the starting pH value of 6.7, which corresponds to the initial pH of okara, without pH control. However, we wanted to check the effect of different pH values on the solubilization of okara. As shown in Fig. 2A, the solubility was minimal at pH 4.0 and increased at higher pH values for both the aqueous controls and the 0.3% subtilisin hydrolyzed extracts. This data suggests that alkaline pH favors solubilization. After the treatment, the enzymatic extracts reached a solubility of 27.3% at pH 8.0 and 30.4% at pH 10.0, respectively, despite that difference was not statistically significant. This represented an additional 17.1% and 17.8% solubilization at pH 8.0 and 10.0, compared to their respective controls, and is consistent with the optimal pH range activity for subtilisin (pH 7.5–10.0). At acidic pH values subtilisin samples yielded an extra 11.2–10.4% than the controls. Overall, the solubilization achieved with the subtilisin treatments at pH 8.0 and 10.0 was significantly much higher than any of the controls at any pH value tested, hence justifying the use of subtilisin in the alkaline pH range. 3.3.1. Effect of pH and subtilisin treatment on the solubilization of proteins After testing the enzymatic treatment at different pH values we observed that the solubilization of proteins increased at higher pH values, being maximal at pH 10.0 (Fig. 2B). The addition of subtilisin doubled the amount of protein extracted compared to the respective controls at all pH values tested. In particular, the increase was 2.5 fold at pH 4.0, 2.9 fold at pH 6.0, 2.1 fold at pH 8.0, and 2.2 fold at pH 10.0, which represents almost 200 mg extra of solubilized protein per g of dry okara. When the soluble proteins of the enzymatic extract at pH 10.0 were analyzed by molecular exclusion chromatography and compared to the negative control extracted with water only (Fig. 1,Table 2) we observed that proteins with a molecular weight greater than 10 kDa decreased from 91.14% to only 7.85%, while the small protein/peptide (1–10 kDa) and small peptide/amino acid (<1 kDa) fractions increased from 1.09% to 15.87% and 6.98%–75.74%, respectively. It is worthy to mention that bioactive peptides, which usually consist of 2–20 amino acids, may be generated by proteolytic digestion and therefore be contained in these fractions. The biological and antioxidant properties of these peptides are related to their content in hydrophobic and aromatic residues (Kitts and Weiler, 2003;Puchalska et al., 2017; Samardi and Ismail, 2010;Zhang et al., 2012), whilst the abundance of hydrophobic amino acids also contributes to the bitter taste of soy (FitzGerald and O'Cuinn, 2006). Strikingly, subtilisin hydrolysates of soy protein have been found to exhibit the highest bitter taste compared to hydrolysates with other proteases (Seo et al., 2008), thus reflecting their idoneity for the potential generation of bioactive peptides. Taken together, these results underscore the adequacy of the subtilisin treatment to solubilize the protein content of okara and its potential to generate bioactive peptides. 3.3.2. Effect of pH and subtilisin treatment on the solubilization of isoflavones The solubilization of isoflavones was measured by mass spectrometry both in the presence and absence of subtilisin. Table 4 shows the total amount of isoflavones, aglycones, β-glucosides, and acetyl and malonyl derivatives extracted at different pH values. In general, we observed that alkaline pH values exerted the strongest effect on the solubilization of isoflavones. Thus, when the pH increased from 8.0 to 10.0 the total amount of solubilized isoflavones raised an extra 28% (763.40 vs 597.54 μ g/g of dry okara) in the presence of subtilisin and 38% (469.89 vs 338.97 μ g/g of dry okara) in the control samples. Compared to their water-soluble counterparts, the addition of subtilisin at pH 8.0 augmented the extraction of total isoflavones by 76% (597.54 vs 338.97 μ g/g of dry okara) and 62% at pH 10.0 (763.40 vs 469.89 μ g/g of dry okara). Remarkably, at pH 10.0 the subtilisin treatment increased 9.1 times the total amount of aglycones extracted (174.83 vs 19.18 μ g/g of dry okara). This can be due to an increased efficiency of the extraction at pH 10.0, but also to a higher interconversion of the thermally unstable conjugated forms (malonyl and acetyl glucosides) to their respective heat-stable non-conjugated β-glucosides and aglycones (Mathias et al., Table 3 Whole amino acid composition from dry matter of okara used in the experiments. Importantly, the higher recovery of aglycones is desired, since they are considered to be the most bioactive forms of isoflavone, due to their moderate lipophilicity that allows them to be better absorbed in the gut (Murota et al., 2002). For each of the aglycones measured (daidzein, genistein and glycitein), for the β-glucosides daidzin and genistin, and for total isoflavones, the highest solubilization values were achieved at pH 10.0. For the β-glucoside glycitin the maximum solubilization (96.04 μ g/g of dry okara) was achieved at pH 6.0 in the presence of subtilisin. Malonyl and acetyl derivatives increased gradually with pH and reached the highest values at pH 8.0. However, at pH 10.0 they were almost inexistent, except for the malonyl genistin extracted with subtilisin, which remained high (105.39 μ g/g of dry okara) despite some interconversion to its nonconjugated forms. No other major differences were observed for the malonyl and acetyl derivatives in the presence or absence of subtilisin, hence indicating a small effect of the enzyme on their extraction. In the okara hydrolyzed with subtilisin at pH 10.0, malonylderivatives accounted for 14.0% of the total isoflavones extracted, β-glucosides for the 63.1% and aglycones for the 22.9%. Also, genistein and its β-glucosides and malonyl-derivatives represented 63.9% of all the isoflavones extracted. The daidzein family represented one third of the total, while the glycitein-based isoflavones only summed 2.8%. Similar percentages were found at pH 8.0, being 60.8% genistein-based isoflavones, the remaining 38.3% daidzein-derivatives, and only 0.9% glycitein and its family. However, at pH 6.0 glycitein-based isoflavones, mainly in the glycitin form, accounted for half of the total isoflavones extracted in the presence of subtilisin. In agreement with other authors (Mathias et al., 2006), this behavior may be related to the extra H 3 CO- group present in the glycitein molecule and reflects how the chemical structure of isoflavones dictates their stability under variable pH conditions, which may be used to modulate the solubility of different isoflavone molecules. Malonyl genistin and malonyl daidzin are the most abundant forms in raw soybeans (Mathias et al., 2006) but their lower thermal stability favors their conversion into non-conjugated forms. Thus, the solubilization of isoflavones during our enzymatic treatment involved an incubation step at 55 C for 2 h. At this temperature isoflavones are relatively stable (Xu et al., 2002). Besides, it may increase the solubility of aglycones (Wu et al., 2010) and also favor the conversion of daidzin and genistin into their bioactive aglycone forms (Muliterno et al., 2017), particularly at pH 10.0. However, Jankowiak et al. (2014a) did not observe any significant difference in the total amount of isoflavones extracted within the 20–95 C range, despite a significant decrease of the malonyl-glucosides above 65 C. Similar to them (Jankowiak et al., 2014a), we also used water as extractant instead of ethanol. Whereas they tested water to dry okara ratios ranging from 20:1 till 100:1, we just used a 10:1 ratio. However, only when they assayed the 20:1 ratio at pH 10.0, they were able to increase the aglycone content up to 340 μ g/g of dry okara approximately and recover 911 μ g of total isoflavones per gram of dry okara. In our hands, water extraction alone did not recover high yields, and we only obtained 19.18 μ g of aglycones and 469.89 μ g of total isoflavones per gram of dry okara at pH 10.0. Since both methods are theoretically equivalent, the differences in the aglycone and isoflavone content at pH 10.0 without any enzymatic treatment may be essentially attributed to the type of soy used and the processing conditions during soymilk manufacture, as well as the distinct liquid-to-solid ratio used (10:1 in our case and 20:1 for Jankowiak et al.). It should be expected to increase our aglycone and total isoflavone content to some extent by raising the liquid-to-solid ratio from 10:1 to 20:1. However, the addition in our case of 0.3% subtilisin increased the total amount of isoflavones solubilized (763.4 μ g/g of dry okara) by 62.5% and raised the aglycone content 9.1 times up to 174.8 μ g/g of dry okara, whereas for Jankowiak et al. their highest extraction yield using 70% ethanol increased their isoflavone content only by 11.7% (1018 μ g/g of dry okara) and the aglycone levels barely added a 5% (357 μ g/g of dry okara) extra. 3.4. Protein/isoflavone relationship in okara extracts Remarkably, we found a direct correlation between the amount of proteins solubilized and total isoflavones extracted at all pH values tested, either with the subtilisin treatment or the water extraction (Fig. 3). However, after the enzymatic treatment, the yield of isoflavones extracted was lower than expected for the quantity of proteins solubilized. This reinforces the notion that the solubilization of isoflavones was an indirect effect provoked by the solubilization of proteins and that some isoflavones may remain trapped in the insoluble fiber matrix too, in agreement with previous works (Jankowiak et al., 2014a). 3.5. Reducing activity of okara extracts When the specific ability of the samples to reduce Fe þ3 to Fe þ2 was measured using the FRAP technique (Fig. 4), we found that the reducing activity of the samples was strongly affected by the pH, despite the fact that samples hydrolyzed with subtilisin were higher than their respective controls. At acidic values, the effect of the enzymatic treatment was considerable (3.2-fold at pH 4.0 and 2.4-fold at pH 6.0 over their respective controls), but slight at basic pH values, where it only represented a 1.2-fold at pH 8.0 and 1.1-fold at pH 10.0. Overall, such antioxidant capacity increased at higher pH values in all samples tested, except for the subtilisin extract at pH 8.0 that was lower than its equivalent at pH 6.0. That anomaly might be due to the solubilization of other Table 4 Effect of the subtilisin treatment at different pH values on the extraction of isoflavones from okara. It has been described that genistein and genistin have the highest antioxidant activities of all soy isoflavones (Lee et al., 2005). Accordingly, their elevated occurrence at pH 10.0 correlated with the highest antioxidant capacity of okara extracts at that pH value. Also, low-molecular weight peptides have been shown to have anti-oxidant activity (Jim enez-Escrig et al., 2010;Nishibori et al., 2017) that may also contribute to the total reducing activity observed in the okara extracts due to the solubilization of the protein content with the subtilisin treatment. 4. Conclusions We developed a method for the valorization of okara based on the application of 0.3% subtilisin at pH 10.0 to 10% (dry w/v) crude okara. This approach rendered high levels of soluble isoflavones and peptides, and a linear correlation between their extractions was observed reflecting potential interactions among them. Our process skips the drying of okara, which is energy intensive and may damage its components, decreasing its properties and economic value. However, it contains an initial step at 55 C that reduces the risk of spoilage by microorganisms and extends the half-life of crude okara. This technology avoids the use of ethanol, which is not profitable at industrial scale, but achieves comparable rates for the solubilization of proteins and isoflavones. In particular, it is noteworthy the rich content of aglycones. The subtilisin activity also facilitates the generation of bioactive peptides with potential uses in health and agriculture, which all in all results in the formulation a new functional product with high antioxidant potency. Therefore, this protocol may represent a functional alternative for the valorization of okara potentially suitable for industrial scale-up. To further explore the importance of toxicological estimation after enzymatic treatment, we studied the removal results of pentachlorophenol with laccase as polyphenol oxidase and the synthetic effluent. Additionally, we investigated the influence of enzyme load (4.55–134.28 U mL−1), pentachlorophenol concentration (2–75 mg L−1), reaction time (0–25min), and hydrogen peroxide concentration (5–820 mol L−1). The results indicated that the maximum removal percentage of pentachlorophenol was 87.4% after 14 min of reaction time when experimental parameters were as following: hydrogen peroxide 101.2 mol L−1, enzyme 74.3 U mL−1and pentachlorophenol 45.9 mg L−1.Lactuca sativa and Artemia salina were used to estimate the toxicity of the products which appeared after enzymatic treatment. Above results indicated that laccase was very efficient for removal of pentachlorophenol, simultaneously, the products which were obtained after enzymatic action indicated relatively high toxicity. The lethal concentration LC50 which was gained for Artemia salina was 52.7% and the inhibition concentration IC50 which was obtained for Lactuca sativa was 26.9%. Moreover, the enzymatic treatment should be united with other kinds of treatment for further degradation and toxicity reduction of pentachlorophenol. Odorous and volatile organic compounds emissions represent a serious problem related to biowaste treatment, because they may be carried away several kilometres, depending on weather and topographical conditions. The most used systems are the biofilters, working in normal operating conditions in terms of temperature and pressure. Due to this, they are relatively cheap and highly efficient. Waste air bio-treatment with biofilters was developed as a reliable and cost-effective technology, the pollutants biodegradation by microorganisms leading to harmless end-products. Biofilters are also efficient systems for odorous off-gases from composting processes treatment. Results revealed that the main part of the volatile organic compounds load was degraded within the biofilters. As a conclusion, the benefits of the biotechnology use, such as environmental monitoring and inspecting, pollutant removal, soil and groundwater treatment, molecular nanotechnology, green energy and so on, really make the great changes in our environment and living life. http://dx.doi.org/10.1016/j.jbiotec.2017.06.1010 Okara valorization process by fermentation with Bacillus licheniformis: Obtention of hydrolytic enzymes, bioactive compounds, and protein hydrolysates Angel Orts Gomez1,∗, Juan Parrado Rubio1, Manuel Tejada Moral2, Bruno Rodriguez Morgado1, Pablo Caballero Jimenez1 1Faculty of Pharmacy, University of Seville, Seville, Spain 2ETSIA, University of Seville, Seville, Spain E-mail address: [email protected] (A.O. Okara is a by-product obtained during the manufacturing process of soy milk that has a great potential for food industry due to its richness in proteins and high content in bioactive compounds (isoflavones). However, it is underused due to its low bioavailability caused by its high fiber content and insolubility. B. Licheniformis is a Gram-positive bacterium used in the biotechnology industry to produce industrial enzymes (proteases, ␣-amylase, lipases, chitinases, etc.) that also can be used to modify unsoluble by-products and hence obtain highly valuable biological products. The aim of this work was to develop a biorefinery process of Okara using fermentation with B. Licheniformis as a biological tool. Once the optimal fermentation conditions (pH, temperature, substrate concentration) were determined, we obtained a new product that was rich in enzymes (proteases and lipases) and where the organic matter from Okara was modified by the hydrolytic enzymes excreted by B. Licheniformis. In second place, a biorefinery process to separate the valuable fractions – (1) hydrolytic enzymes, (2) soluble protein hydrolysates, and (3) soluble extracted bioactive compounds – was evaluated. This separation was designed using size

Kati). The present study is focused on isolating actinomycetes from compost sample, Ulva lactuca and investigating the enzyme and antibacterial substance producing abilities of these isolates. The Ulva lactuca was prepared as compost material. Actinomycetes were isolated from compost sample by serial dilution method on agar medium with or without potassium dichromate. The presence of extracellular amylase, lipase/esterase, chitinase, pectinase, protease and cellulase enzymes in these isolates were qualitatively investigated. The isolates were tested for antibacterial activity. Fourty actinomycete isolates have been obtained from Ulva lactuca compost sample. Many isolates produced one or more enzymatic activities. From these isolates, 34 (85%) showed amylase, 21 (53%) lipase/esterase, 21 (53%) chitinase, 29 (73%) pectinase, 40 (100%) protease and 30 (75%) cellulose activities. Extracellular enzyme activity results were evaluated and found that totally eight isolates have quite high amylase and ten isolates have quite high protease activities. Two isolates exhibited quite high antibacterial activity. This study clearly indicated that algae compost can be considered as an important source for actinomycetes. Some actinomycete isolates showed high enzymatic and antibacterial activities. These isolates may be used for commercial, industrial and pharmaceutical applications in the future. The optimum pH and temperature of the enzyme were found as 11.5 and 30 ◦C, respectively. However, the activity of the enzyme was high at temperatures ranging from 40 to 90 ◦C. The alkaline protease was 91% stable for 30 min at 60 ◦C. The enzyme activity was slightly inhibited by 1 mM MnSO4and 5 mM NaCl with, 3% and 1%, respectively. The proteolytic activity was moderately stimulated by the chelating agent EDTA (113%, 115%) at 1 and 5 mM concentration. 1% Triton X-100, Tween-20 and Tween-80 increased the enzyme activity up to 124%, 113% and 115%, respectively. In contrast, the higher concentration (5%) of Triton X-100, Tween-20 and Tween-80 was significantly inhibitory to protease stability and this activity decreased to 28%, 22% and 14%, respectively. Alkaline protease activity of Bacillus megaterium was determined as 42.97 U/mL. The molecular weight of alkaline protease was estimated to be 83 kDa by SDS-PAGE. Considering these properties Bacillus megaterium alkaline protease has significant application potential in laundry detergents. Additionally, as a food processing aid, lasparaginase can significantly reduce the level of acrylamide in starchy food. In this study, l-asparaginase was procured from soil isolates. The highest l-asparaginase producing strain was identified as Bacillus subtilis. The optimum activity of the purified enzyme was observed at 40 ◦C, at pH 8.0. In treating the enzyme with different buffers of pH, 100% of the stability was obtained at pH 7.0. By pre-incubating the enzyme at 20 ◦Cto100◦C, it was 100% stable at 50 ◦C, whereas the enzyme became unstable at temperatures over 70 ◦C. While urea and EDTA greatly decreased the enzyme activity, however, MgCl2increased the activity. The enzyme was seen to be a single band with a molecular weight of 128 kDa by using the zymogram method. http://dx.doi.org/10.1016/j.jbiotec.2017.06.1014 Freeze-drying microencapsulation of anthocyanins from sour cherries in the -lactoglobulin matrices Nicoleta St˘ anciuc∗, Ana Maria Oancea, Ioana Otilia Ghinea, Vasilica Barbu, Gabriela Bahrim, Gabriela Râpeanu, Iuliana Aprodu Faculty of Food Science and Engineering, “Dunarea de Jos” University of Galati, Romania E-mail address: [email protected] (N. [email protected] Resumen* En el presente trabajo se ha desarrollado un proceso enzimático destinado a la valorización de la okara, mediante su conversión en un producto bioestimulante agronómico/ambiental altamente biodisponible y con alto contenido en C, N, P, K.. Abstract ** In the present work, an enzymatic process has been developed for the valorization of okara, through its conversion into an agronomic/environmental biostimulant product with high bioavailability and high content in C, N, P and K. Okara is an insoluble by-product resulting from the process of obtaining soy milk and tofu. Although it has a high nutritional value (proteins, fiber and lipids), where its composition of bioactive compounds such as isoflavones, vitamins and trace elements stands out, it presents problems of utilization due to its high water content, which makes it a very perishable and easily fermented product. All this makes it an ideal substrate for obtaining agronomic/environmental compounds. The process is based on the use of different hydrolytic enzymes, which lead to the solubilization and hydrolysis of organic (proteins and sugars) and inorganic (P and K) molecules. The enzymatic process designed consists of a sequential multi-enzymatic hydrolysis involving proteases, phytases and carbohydrolases. The enzymatic extract has been chemically characterized, highlighting its high content in peptides and free amino acids (soluble organic content composed of 64% of molecules <1 KDa, solubilization of 61% of total proteins), as well as in organic phosphate (solubilization of 79.1% of total P), K (solubilization of 86.6% of total K) and free glucose. Datos referidos a materia seca material.

Enzymatic hydrolysates showed a different chemical composition compared with fermented hydrolysates and OK. It had a higher protein concentration as well as C, P and K. The proteins of OK were converted into peptides with a lower molecular weight, the fermented hydrolysates being those with the lowest molecular weight profile. The influences of hydrolysates and OK were tested in soil, finding that β-glucosidase, phosphatase and dehydrogenase activities were stimulated by every treatment. However, it was observed that EH produced a greater stimulation of dehydrogenase and phosphatase than both OK and fermented BS. The bacterial and fungal phospholipid fatty acids were also higher in soils amended with BS than those of the control and soils with OK. It has also been found that β-glucosidase, phosphatase and microbial biomass were dose-dependent in every treatment, but dehydrogenase only was dose-dependent in EH and OK treatments. Introduction The addition of organic matter to soils is a common agricultural and environmental practice used to supply nutrients and improve the soil properties with the aim of enhancing either the productivity of the agroecosystem or reclaiming degraded soils [1–3]. In order to achieve these objectives, different sources rich in organic matter such as crop residues, animal manures, sewage sludge, by-products of food processing industry, etc. have been used [4–7]. However, it is necessary that these residues break down to provide the nutrients needed to stimulate the soil microbiology but, as found by several authors, this breakdown takes some time which depends on both the soil properties and the nature of the added organic wastes [1,8,9]. Therefore, recently, new products, called biostimulants (BS), have been used. These products are obtained by hydrolytic processes, enzymatic hydrolysis being the most used one. In order to produce these BS, different industrial by-products have been used, such as carob germ [10], sewage sludge [11], wheat-condensed distiller [12], defatted sunflower seed meal [13], chicken feathers [3], rice bran [3] and by-product of the dry-mill bioethanol process [14]. Enzymatic hydrolysis of these organic wastes yields a product rich in peptides, amino acids, polysaccharides, humic acids, etc., which provide a higher concentration of organic matter, macro and micro nutrients than those of the raw materials. Therefore, they provoke an enhancement of the microbial soil activity, improving its capability to reclaim contaminated soils; thus several researchers have studied the effect of BS on the degradation of different xenobiotics in soils, such as policyclic aromatic hydrocarbon (PAH) [15] and pesticides [16]. Likewise, the use of BS as biofertilisers exerts a positive influence on plant growth and fruit quality and production [10,13,17,18] Taking into account what has been found so far, any organic waste rich in organic matter can be used to obtain BS. In this sense, okara (OK) is an interesting raw © 2018 Informa UK Limited, trading as Taylor & Francis Group CONTACT Isidoro Gómez-Parrales [email protected] Reasearching Group Edafología Ambiental, Dpto. OK is a by-product of soy milk and tofu production rich in dietary fibre and proteins which contains essential amino acids [19]. Besides, it has a little quantity of fats, other carbohydrates and isoflavones. This composition makes OK an exceptional source of functional compounds in the food industry [20–23]. Nevertheless, due to the fact that OK ferments rapidly and has a high content of water, its conservation, although possible, is unprofitable. Consequently, OK has to be processed quickly and so, currently in the Western World, this by-product is mainly used as feed or fertiliser [19]. To overcome this aspect, OK could be submitted to a hydrolytic process to obtain a product with a high concentration of peptides and other functional compounds which can be used not only as a source of organic nitrogen and phosphorus but also as a sustainable tool to enhance the soil quality, contributing to making OK profitable and to reduce chemical fertilisers [24,25]. In addition to enzymatic hydrolysis to valorise a wide variety of agricultural and agro-industrial by-products, several authors have carried out a fermentative process using different microorganisms to recycle these residues, most of which focus on the production of bioethanol [26–30] and enzymes [31–37]. On the other hand, there are some research works that study the application to soils of the bio-based products obtained by fermentation. Among them, it is worth highlighting those which focus on the production of bioherbicides [38], bio-pesti- cides [39,40] and biosurfactants [41,42]. Many research works have brought to light the importance of biosurfactants in the restoration of contaminated soils. In this sense Magthalin et al. [43] found that cationic biosurfactants obtained from chicken tallow were very efficient in the bioremediation of chromium-contaminated soils. On the other hand, other authors have highlighted the importance of these substances in reclamation of organic xenobiotics-contaminated soils. In this sense, Bezza et al. [44] and Mnif et al. [45] concluded that biosurfactants enhanced the bioavailability and hence the biodegradation of PAHs and diesel oil. Finally, Wang et al. [46] observed that the phytoremediation of DDT- contaminated soil was improved when biosurfactants were used. However, there are few articles in which the production of BS by fermentative processes is studied, such as Chintagunta et al. [30] who, in fact, obtained a biomanure as a co-product in the bioethanol manufacturing process, Oliveira Mendes et al. [47] and Aceves- Díez et al. [48] who used the broth obtained after the fermentation of a culture medium with Aspergillus niger and Bacillus thuringiensis as biofertiliser, respectively. Hence, the present paper aimed to produce BS from OK through two different processes, namely an enzymatic hydrolytic process, using an endoprotease, and a fermentative process, using Bacillus licheniformis. After obtaining the BS, its effect on soil biological properties and soil microbial community was assessed. 2. Materials and methods 2.1. Substrate, enzymatic hydrolysis and fermentative processes The substrate used in the fermentative as well as in the enzymatic hydrolysis process was OK, a by-product of the soy milk production provided by the company Soria Natural located in Garray (Soria, Spain). The enzymatic hydrolysis process was carried out according to the pH-stat method [49] using as hydrolytic agent the enzyme bioprotease LA-450 (Biocon Española S.A.) which is subtilisin. The process took place in a bioreactor operating under controlled temperature without either stirring or pH control and with the following conditions: (a) temperature: 55°C; (b) OK concentration: 10% (w/v); (c) subtilisin concentration: 0.05% and (d) operating time: 3 h. Finally, the hydrolysed product was centrifuged and the hydrolysed liquid (EH) was concentrated in a rotary evaporator until obtaining 58% of dry matter (Table 1). Regarding fermentation, this was carried out in 500 mL Erlenmeyer flasks under controlled stirring and temperature by B. licheniformis ATCC 21415, which was stored under freezing at −80°C and refreshed 24 h before inoculation in LB medium (10 g tryptone, 10 g NaCl, 5 g yeast extract and 1 L of water). Then the insoluble fibre was separated by centrifugation, obtaining a supernatant with hydrolysed proteins, bioactive products, enzymes and bacteria (FHEB). Secondly, a fraction of this supernatant was centrifuged to eliminate the bacteria, and thus a soluble fraction with hydrolysed proteins, bioactive compounds and enzymes was obtained (FHE). Thirdly, a portion of this last fraction was heated at 80°C for 20 min to denature the enzymes. Therefore, the resultant product was composed of hydrolysed proteins, bioactive compounds and denatured enzymes (FH). Finally, all these fractions were concentrated in a rotary evaporator obtaining the following dry matter content for each BS: FHEB 77.4%, FHE 31.5% and FH 38.9% (Table 1). 2.2. Chemical characterisation of OK, enzymatic and fermented hydrolysates Macro elements in OK (previously burned) and in enzymatic and fermented hydrolysates were analysed by inductively coupled plasma atomic emission spectrometry. The equipment used standard operational conditions, which are detailed in Parrado et al. [37]. Ash was analysed according to standard AOAC methods [50]. Total soluble carbohydrates were determined after extraction with a mixture of ethanol/water (2/3) for 2 h. After centrifugation at 4000×g, the supernatant was filtered through a No. 1 Whatman paper, and total soluble sugars were estimated colorimetrically using the phenol–sulphuric acid method with a standard curve of glucose [51]. Crude fat content was determined gravimetrically after OK extraction with hexane for 12 h in a Soxhlet extractor [52]. Organic matter was analysed by dry combustion at 550°C for 6 h [53]. Total nitrogen was determined by the Kjeldahl method [50]. The protein content was determined by multiplying the total nitrogen content in a protein by the conversion factor 5.71, which is applicable to soy protein. Fibre content was determined gravimetrically according to standard AOAC methods [50]. OK solubility was determined in 200 mL of water with an OK concentration of 10% at 55°C for 24 h. 2.3. Protein molecular weight and solubility determination The molecular mass distribution of protein in the samples was determined by size-exclusion chromatography using an ÄKTA-purifier (GE Healthcare). The operational conditions are described in Parrado et al. [10]. Protein solubility was determined using the method described by Adler-Nissen [54]: briefly, 5 mL of 2.4 M trichloroacetic acid was added to 10 mL of the sample (1% w/v), the precipitate was removed by centrifugation (8000g, 10 min) and the nitrogen concentration of the supernatant was determined. 2.4. Biostimulation experimental design The soil used in this experiment is a Haplic Regosol (Calcaric) [55]. Soil samples were collected from the 0–25 cm surface layer. The main soil characteristics and methodology used to determine the physical and chemical parameters in soil are presented elsewhere [11,17]. The biostimulation design was performed in microcosms consisting of a glass pot with 300 g of dried and 2 mm sieved soil. Prior to the treatments, soil was preincubated at 25°C for 7 days at 40% of its waterholding capacity according to Tejada et al. [56]. Soil was amended with OK and the four BS obtained at two concentrations so that the organic matter was 1% and 1.5%. An unamended soil was used as control. Distilled water was added to each soil to achieve 60% of its water-holding capacity. The BS were liquid, so they were dissolved in distilled water before the application. On the other hand, OK, having been dried, was a powdery solid and so was dry mixed with soil. Therefore, there were five treatments per each concentration plus the control soil, with the following incubation conditions: (a) C: unamended soil; (b) OK1.5: amended soil with OK so that organic matter in soil was 1.5%; (c) OK1: amended soil with OK so that organic matter in soil was 1%; (d) EH1.5: amended soil with EH so that organic matter in soil was 1.5%; (e) EH1: amended soil with EH so that organic matter in soil was 1%; (f) FHEB1.5: amended soil with FHEB so that organic matter in soil was 1.5%; (g) FHEB1: amended soil with FHEB so that organic matter in soil was 1%; (h) FHE1.5: amended soil with FHE so that organic matter in soil was 1.5%; (i) FHE1: amended soil with FHE so that organic matter in soil was 1%; (j) FH1.5: amended soil with FH so that organic matter in soil was 1.5%; (k) FH1: amended soil with FH so that organic matter in soil was 1%. Triplicates of every treatment were performed and were incubated for 55 days at room temperature. For each treatment and each incubation time, a sample of 20 g of soil was taken and stored at −20°C in sealed polyethylene jars prior to enzymatic activities and phospholipid fatty acid (PLFA) analyses. 2.5. Statistical analysis In order to test the significance of differences among means of the dependent variables measured in the soil for each treatment during the incubation (repeated measurements model) a mixed ANOVA model with a between-subjects factor (incubation time) and a withinsubject factor (treatment) was performed. The analysis was made with the function aov of R statistical software [62]. When the differences were statistically significant the post hoc Tukey HSD test was applied using the R package ‘agricolae’[63]. 3. Results 3.1. BS obtaining process As mentioned above, OK is a by-product of the soy milk manufacturing process with only 23.3% of dry matter which is composed of fat (23.6%), insoluble fibre (26%) and protein (41.28%) (Table 1). OK’s chemical composition makes it an interesting product to be used as biostimulant/biofertiliser but due to its low water solubility (17%) it has very little bioavailability. In order to make the most of OK’s chemical composition, it is necessary to degrade the insoluble fraction and make it more bioavailable. To achieve this goal, two biorefinery processes have been proposed, namely enzymatic hydrolysis and a fermentative process. The enzymatic hydrolysate showed an increase of 25% in proteins content with respect to OK. Most of these proteins in OK (91.14%) have a molecular weight higher than 10 kD while in EH 75.74% of them have a molecular weight lower than 1 kD (Table 1). On the other hand, the fermented hydrolysates FHEB, FHE and FH had practically the same chemical composition since the differences among them are due to physical processes; so only the FHE chemical analysis is shown. The protein content is 42.25% (24% lower than EH), almost the same as that of original OK; nonetheless, 28.42% of these have a molecular weight between 1 and 5 kD and 71.47% are below 1 kD, that is, 99.89% have a molecular weight under 5 kD. Table 1 also gathers the elemental analysis of every product. The concentration of phosphorus is higher in EH (11.16 mg kg −1 ) than both in OK (6.53 mg kg −1 ) and in FHE (6.89 mg kg −1 ). Regarding the potassium values, EH has the highest one (17.14 mg kg −1 ), followed by FHE (11.25 mg kg −1 ) and OK had the lowest quantity (9.11 mg kg −1 ). Nitrogen behaves in a similar way as phosphorus, i.e. EH presents the greatest value (10.64%) among the three products. On the contrary, the carbon content of OK (53.7%) is larger than those of EH (44.81%) and FHE (33.6%). 3.2. Soil microbial activity 3.2.1. β-glucosidase activity In Table 2 it can be observed that every treatment produced a significant stimulation (p< .05) of this activity regarding control soil from the 7th day on, showing a sharp increase until reaching the maximum value between 10th and 15th days in soils amended with either enzymatic or fermentative hydrolysates. Afterwards, an abrupt decrease was produced until the end of the incubation period, reaching values similar to those of the first day. The evolution of βglucosidase activity in soils amended with OK followed a quite different behaviour, namely, there was an increase during the entire experimental period, reaching the maximum value at the end of the incubation. The highest β-glucosidase activities were achieved in soils amended with FHE1.5 and FHE1 on the 15th day, being 2.16 and 1.96 µmol PNP g −1 h −1 , around fourfold higher than that of the control and two-fold significantly higher than the maximum values of the other products at both concentrations. On the other hand, the maximum β-glucosidase activity in soils amended with EH, FHEB, FH and OK was twice as much that of the control but did not show significant differences among them. According to the evolution of ß-glucosidase activity over the incubation period, the maximum values, achieved on different days depending on the treatment, were significantly higher than those of the first day. The geatest differences were observed again in soils amended with FHE1.5 and FHE1. This enzymatic activity was dose-dependent; so the higher the dose the greater the β-glucosidase activity. The differences were significant in the period between the 7th and 30th days, except when FHEB was used, in which case, practically, there were no significant differences during the entire period. 4Á.
