III RESUMEN El presente trabajo, tiene como objetivo principal ampliar el conocimiento existente sobre el manejo de reproductores, y la reproducción de Haliotis tuberculata coccinea (Reeve, 1846), especie considerada candidata potencial para diversificación de la acuicultura en Canarias. El Experimento I, se basa en el estudio de la reproducción y crecimiento de una población natural, para ello veinte ejemplares de Haliotis tuberculata coccinea fueron capturados mensualmente, entre junio del 2005 y mayo del 2007, del medio natural, en la isla de Gran Canaria. Se utilizaron técnicas histológicas con el propósito de investigar el ciclo reproductivo, y se estimaron los parámetros de crecimiento de von Bertalanffy. La morfología del ovario, reveló la existencia de seis estados de células germinales femeninas en el proceso oogénico, y un desarrollo ovárico por grupos sincrónicos. El ciclo reproductivo fue caracterizado como anual a lo largo de todo el año, aunque la máxima actividad reproductiva se observó entre agosto y febrero. La fecundidad varió entre 85.749 huevos (29 mm en longitud de concha) y 691.456 huevos (62 mm en longitud de concha). Se observó una fecundidad significativamente superior en las hembras superiores a 55 mm en longitud de concha, sugiriendo que esta es la talla mínima de reproductores que se debe utilizar en un criadero, para optimizar la producción de huevos. En relación a la estructura de la población, el radio de machos respecto a hembras fue de 1:1,02, y el análisis de clases de tamaños no reveló ninguna tendencia en los datos. Los parámetros de von Bertalanffy estimados para hembras y machos fueron de 72,5 y 78 mm en longitud de concha, y una tasa de crecimiento de 0,43 y 0,37 año -1 respectivamente. El Experimento II, se centró en el efecto de la calidad del alimento, sobre el perfil de ácidos grasos de las gónadas, sobre algunos aspectos reproductivos (la morfología de la gónada y la viabilidad de puestas inducidas), y sobre parámetros fisiológicos (tasas de crecimiento y alimentación) en adultos de Haliotis tuberculata coccinea. Los individuos fueron alimentados con dietas constituidas por macroalgas ( Gracilaria cornea Agardh, 1852) de diferente contenido proteico, designadas como Low-N, Medium-N e High-N G. cornea . Después de inducir la puesta, el mayor número de huevos expulsados por gramo de animal, y los mayores diámetros de huevos y de citoplasma, fueron encontrados en los individuos que se alimentaron con las dietas Medium-N y High-N G. cornea . Se observó, que la alimentación con la dieta High-N G. cornea , aumentó el contenido del ácido Araquinódico en las gónadas maduras de Haliotis tuberculata coccinea . Asimismo, el estudio histológico de la gónada reveló
IV que el estado maduro de individuos alimentados con Low-N G. cornea , tuvo una mayor cantidad de poblaciones de macrófagos y las áreas de oocitos degeneradas fueron más frecuentes. El presente experimento sugirió que las macroalgas cultivadas en agua enriquecida con amonio, puede ser una buena opción para alimentar las hembras reproductoras de Haliotis tuberculata coccinea en Canarias. El Experimento III, trata sobre la utilización de anestésicos en la manipulación de los reproductores. Se probó la eficacia del aceite de clavo como anestésico, en las tareas rutinarias en una granja, en ejemplares entre 42 – 80 mm en longitud de concha. El aceite de clavo fue comparado con el anestésico habitualmente utilizado en el cultivo de oreja de mar, 2fenoxietanol. El aceite de clavo fue mezclado previamente al 50 % con etanol absoluto y se testaron cuatro concentraciones: 0,1 ml/l, 0,3 ml/l, 0,5 ml/l y 0,7 ml/l. Para evaluar la eficacia de los anestésicos se consideraron los tiempos de relajación y de recuperación, la condición del músculo y la mortalidad. Los resultados mostraron que 0,5 ml/l de aceite de clavo fue la menor concentración que desprendió todos los individuos de sus refugios en 8,9 ± 3,5 minutos, con un tiempo de recuperación de 2 ± 1,5 minutos. En comparación con el 2-fenoxietanol, el aceite de clavo fue efectivo en una dosis 10 veces menor. En el Experimento IV, se identificó al poliqueto que afectó el cultivo de Haliotis tuberculata coccinea , en el Instituto Canario de Ciencias Marinas en 2008, y además evaluó la eficacia de la aplicación de dosis múltiples de mebendazol para controlar dicha infestación. El mebendazol fue aplicado en forma de baños con el producto denominado comercialmente lomper® (Laboratorio Esteve, España), en las siguientes concentraciones: 6 ml/l, 8 ml/l y 10ml/l, asimismo se utilizó 0 ml/l como control. Las orejas de mar infestadas fueron expuestas a tres baños de mebendazol, aplicados en tres días consecutivos (un baño de tres horas cada día), como se recomienda en las instrucciones del medicamento, para su uso en humanos. Después de cada baño, las orejas de mar se mantuvieron en los tanques de cultivo hasta el próximo baño. Cada mes, debido al efecto del mebendazol, los poliquetos moribundos y/o muertos se desprendieron de sus madrigueras en las conchas de la oreja de mar, y se encontraron en el fondo de los tanques, en los siguientes quince días a la aplicación de los baños de mebendazol. Estos poliquetos fueron contabilizados, recolectados y fijados en formaldehido al 10 % para su posterior identificación. Todo este proceso se repitió cada mes, hasta que no aparecieron más poliquetos en el fondo de los tanques, después de la aplicación de los baños de mebendazol. La especie de poliqueto fue identificada como Polydora hoplura (Claparède, 1870). La eficacia del lomper® fue aproximadamente de un 99 % en todas las concentraciones ensayadas, cuando fue aplicado durante siete meses consecutivos (cada mes, se aplicaba un baño durante tres días consecutivos). La mortalidad de las orejas de mar, ocurrió solamente en los animales muy
V infestados. El estudio de crecimiento durante el experimento, sugirió que el tratamiento con mebendazol no afectó significativamente el peso del animal. El Experimento V, del presente trabajo, se centró en un estudio sobre el acondicionamiento de un lote de reproductores de Haliotis tuberculata coccinea , proveniente del medio natural y estabulado en las instalaciones del Instituto Canario de Ciencias Marinas, sin modificar la temperatura del agua, y alimentados con una dieta mixta de algas. El objetivo fue la determinación del potencial reproductor de esta especie en cautividad. Se estudió histológicamente el índice gonadosomático y se comparó con el visual, con el propósito de determinar el momento adecuado para inducir la puesta y el potencial reproductor (i.e. fecundidad, número de hembras que alcanzan la madurez, respuesta a la inducción al desove, número de puestas etc.) a lo largo de un año, induciendo la puesta cada mes. Además se realizó un experimento para determinar la concentración idónea de espermatozoides para obtener el mayor porcentaje de fertilización. Si bien se obtuvieron puestas inducidas a lo largo de todo el año, no todos los reproductores acondicionados respondieron satisfactoriamente a las condiciones de cultivo, ni a la inducción a la puesta. Este hecho sugiere la importancia de realizar una selección de reproductores, para minimizar el trabajo y los costes que implican las tareas de acondicionamiento en un criadero. El momento más adecuado para inducir la puesta en Haliotis tuberculata coccinea fue cuando los reproductores estuvieron en un estado de desarrollo gonadal 3 (determinado visualmente), sin embargo, este estado no aseguró la puesta inducida. Los resultados mostraron que Haliotis tuberculata coccinea es una especie desovadora parcial y que requirió entre dos y cuatro meses de acondicionamiento para volver a responder satisfactoriamente a la inducción a la puesta. No se encontró ninguna relación estadísticamente significativa entre la fecundidad y la talla del animal (longitud de concha). La concentración óptima de espermatozoides a la cual se obtuvo un mayor porcentaje de fertilización, y un mayor porcentaje de desarrollo larvario normal, fue de 10 4 espermatozoides/ml.

VII ABSTRACT The main objective of the present work is to expand the existing knowledge about conditioning and reproduction of Haliotis tuberculata coccinea (Reeve, 1846), a species considered a potential candidate for the local aquaculture diversification in the Canaries. The Experiment I is based on a study of the reproduction and growth of Haliotis tuberculata coccinea in a natural population for this, twenty monthly samples were collected during June 2005 and May 2007 at Gran Canaria Island. Histological techniques were used to investigate the reproductive biology and, von Bertalanffy growth parameters were estimated. The ovary morphology revealed six stages of female sexual cells in oogenetic process and an ovaric development by group synchronous. The reproductive cycle was characterized as an annual spawning throughout the year, although the maximum reproductive activity of population was between August and February. Batch fecundity ranging from 85,749 eggs (39 mm in shell length) to 691,456 eggs (62 mm in shell lenght) and significantly highest fecundity was found in individuals over 55mm shell length, suggesting that this is the minimum broodstock size to be used in hatcheries to optimize egg production. Related to population structure, the overall sex ratio of males to females was 1:1.02 and the analysis by size class did not reveal a tendency in data. The von Bertalanffy parameters estimated for females and males were 72.5 and 78.8 mm SL of asymptotic length and, 0.43 and 0.37 years -1 of growth rate. The Experiment II focuses on the effect of the quality of food on the fatty acid profile of the gonads, on some reproductive aspects (the morphology of the gonad and the viability of spawning) and on physiological parameters (growth and feeding rates) in adult Haliotis tuberculata coccinea females. Abalones were fed on macroalgae diets ( Gracilaria cornea Agardh, 1852) with three different protein levels, namely Low-N, Medium-N and High-N G. cornea . After inducing the spawning, the higher number of eggs released per gram of abalone and the biggest egg and cytoplasm diameters were found in abalones fed on Medium-N and High-N G. cornea diets. Feeding High-N G. cornea increased the level of Arachidonic acid in the mature gonad tissue in Haliotis tuberculata coccinea . Likewise, a histological study on the gonadal morphology revealed that ripe-stage individuals fed on a Low-N G. cornea diet had a higher amount of macrophage populations and the areas of degenerated oocytes increased. The present study suggests that macroalgae cultivated in seawater enriched with ammonium may be a good choice for producing food for female Haliotis tuberculata coccinea broodstock in the Canaries.
VIII The Experiment III discusses the use of the anaesthetics in the manipulation of broodstock. We evaluated the effectiveness of clove oil as an anesthetic in routine tasks on a farm, and we evaluated in specimens from 42 - 80 mm in shell length. The efficacy of clove oil was compared with the anaesthetic commonly used in the cultivation of abalone, 2 - fenoxyetanol. The clove oil was previously mixed at 50% with absolute ethanol and four different concentrations were tested: 0.1 ml/l, 0.3 ml/l, 0.5 ml/l and 0.7 ml/l. Relax and recovery time, muscle condition and mortality after one week was consider to evaluating the efficiency of anaesthetics. The results showed that clove oil at 0.5 ml/l was the lowest concentration that removal all abalone from the rearing shelters in 8.9 ± 3.5 minutes, showing 2 ± 1.5 minutes of recovery time. In comparison with 2-phenoxyethanol, clove oil was effective in 10-fold lower doses. In the Experiment IV, we identified the shell-boring polychaetes that affected in 2008 the cultivation of Haliotis tuberculata coccinea in the Instituto Canario de Ciencias Marinas. We also evaluated the effectiveness of the implementation of multiple doses of mebendazole for controlling this pest. Mebendazole was applied as baths of lomper® (Esteve Laboratory, Spain) using the following concentrations: 6 ml/l, 8 ml/l and 10 ml/l and, 0 ml / l as a control measure. Abalones were exposed to 3 mebendazole baths over a three-day period (1 x three-hour bath applied for three consecutive days), as recommended in the drug directions for human use. After each bath, the abalones were returned to their original culture tanks until the following day. After these three days, the abalones were kept in their original culture tanks for a term of one month. Each month, due to the effect of mebendazole, moribund and/or dead polychaetes became detached from the burrows and were found at the bottom of the tanks during the first 15 days after the baths of mebendazole were applied. The polychaetes expelled from the shells were counted, collected and fixed in 10% buffered formaldehyde for the subsequent identification. This process was repeated month after month until no moribund and/or dead shell-boring polychaete was found at the botton of the tanks after the application of the baths of mebendazole. The shell-boring polychaete species was identified as Polydora hoplura (Claparède 1870).The efficacy rate of lomper® was around 99 % in all the concentrations tested when it was applied during a seven-month term (each month, 1 bath was applied for 3 consecutive days) and, abalone mortality was limited to highly-infested animals only. Monitoring of abalone weight gain suggested that the mebendazole treatments did not affect growth significantly during the study period. The Experiment V of this work focuses on the conditioning of a Haliotis tuberculata coccinea batch captured in the natural environment and stabled in the facilities of the Instituto Canario de Ciencias Marinas, without changing the water temperature and fed on a mixed algae
IX diet. The objective consisted of determining the reproductive potential of this species in captivity. A histological study of the gonadosomatic index was carried out and it was compared with the visual estimation of this index, in order to determine the most appropriate moment to induce spawning and the reproductive year-round potential (i.e. fecundity, number of females reaching maturity, response to spawning induction, number of spawning events) by inducing spawning on a monthly basis. Likewise, an experiment was conducted to determine the concentration of sperm necessary to get the best fertilization rate. Although induced spawning events occurred throughout the year, not all the conditioned broodstock responded satisfactorily to the culture conditions or to the induction of spawning either. This fact suggests the importance of the broodstock selection so as to minimize the work and costs involved in the hatchery and related to the conditioning tasks. According to the results obtained, the best moment to induce Haliotis tuberculata coccinea spawning was the period of time when individuals reached the stage 3 of the gonad index (visually determined). However, this stage did not ensure a successful spawning. The results showed that Haliotis tuberculata coccinea is a partial-spawning species and between two and four months were required so as to obtain again a satisfactory response to the spawning induction. No relationship, statistical significant, between the fecundity rate and the animal size (i.e. shell length) was found. The optimal sperm concentration with which a higher fertilization rate and a higher percentage of normal larval development were obtained were 10 4 spermatozoids/ml.

– Introducción General – 3 Unido (15.371 t) y España (10.092 t). Francia y Reino Unido, así mismo, lideraron las capturas de orejas de mar, bígaros y arcas (13.851 y 12.196 t respectivamente). La ostra fue la especie que menos se capturó y fueron Irlanda y Dinamarca, los principales países productores (2.256 y 1.490 t respectivamente). Figura 1 Producción mundial de moluscos marinos por grupos de especies en 2008 (FAO, 2011). Según FAO (2011), los mejillones ( Mytilus edulis y Mytilus galloprovincialis ) son los principales moluscos producidos por la acuicultura europea, con un total de 454.629 t, principalmente en España (180.265 t), Francia (72.760 t) e Italia (61.200 t). Cabe destacar, el incremento de la producción en países como Irlanda, Grecia y Reino Unido (con un total de 86.825 t, en conjunto), mientras que un país con gran tradición en el cultivo del mejillón, como es Holanda, ha visto su producción reducida en los últimos tiempos debido a las dificultades para conseguir semilla. La ostra fue el segundo molusco producido, la especie mayoritariamente cultivada fue la gigas ( Gassostrea gigas ), aunque también se produjeron otras dos especies más la ostra plana y el ostión ( Ostrea edulis y Crassostrea angulata ). El primer país productor fue Francia (112.960 t), aunque se están sumando otros países como Irlanda, que entre 1999 y 2008 pasó de producir 1.700 a 8.000 t. La almeja japonesa ( Ruditapes philippinarum ), ocupó el tercer lugar con una producción en 2008 de 72.087 t, Italia fue el principal productor aportando 61.800 t, Francia España y Reino Unido produjeron en conjunto un total de 8.100 t. 4 – Introducción General – cantidad se produjo la vieira ( Pecten maximus ) con un total de 224 t de los cuales Irlanda y Reino Unido aportaron un total de 102 t. No existen datos sobre la producción en los cultivos de orejas de mar, bígaros y estrombros. Figura 2 Producción europea de moluscos marinos por grupos de especies en 2008 (FAO, 2011). En España Según JACUMAR (2009), en 2008 la producción española de acuicultura alcanzó 265.298 t de las cuales, 65.262 y 199.923 t correspondieron a peces y a moluscos respectivamente y 113 t a crustáceos. La mayor producción de la acuicultura española corresponde a moluscos, especialmente al mejillón ( Mytilus edulis ) con un 72,69 % de la producción total. Cabe destacar las bruscas variaciones de su producción en los últimos años, en función de los episodios de mareas rojas que tienen lugar en Galicia, pero a pesar de ello, España continua estando entre los primeros países productores con 180.265 t producidas en el año 2008. En la cornisa Cantábrica y la región Noroeste, se ha desarrollado el cultivo del mejillón y ostras en bateas, y el de almejas y berberechos en parques de cultivo. Como cultivos secundarios se pueden destacar el de pectínidos y de forma emergente, el engorde de pulpo. En la zona Mediterránea y Suratlántica de aguas más templadas también tienen cierta importancia el cultivo de ostras, almejas y mejillón. En Canarias y Baleares, no se realiza el cultivo de moluscos a nivel comercial. LA OREJA DE MAR La oreja de mar es el nombre común, con el que se denomina a las especies pertenecientes a la clase Gastropoda (Cuvier 1797), orden Archeogastropoda (Thiele 1929), familia Haliotidae (Rafinesque 1815) y su a único género Haliotis . El primer haliótido conocido aparece en el Cretácico (hace aproximadamente 65 millones de años) en la actual California. Hoy en día, se distribuyen en todos los océanos (Lindgerg, 1992), batimétricamente se localizan en profundidades por encima de los 30 m donde las algas, su alimento natural, crecen abundantemente (FAO, 1990). Si bien existen 56 de especies de oreja de mar (Geiger, 2000), sólo alrededor de 15 tienen interés comercial (Hanh, 1989). Entre ellas, cabe destacar en Asia, la kuro ( Haliotis discus ) que es una oreja de mar de tamaño grande y considerada de gran calidad debido a su exquisito sabor, y la donkey's ear abalone ( Haliotis asinina ) y tokobushi ( Haliotis diversicolor ), que son orejas de mar pequeñas (hasta 10 cm en longitud de concha) y de crecimiento rápido (Cook y Gordon, 2010). En otros lugares del mundo, las especies comerciales incluyen al blacklip abalone ( Haliotis rubra ) y greenlip abalone ( Haliotis laevigata ) en Australia, paua ( Haliotis iris ) en Nueva Zelanda, perlemoen abalone ( Haliotis midae ) en Sudáfrica, red abalone ( Haliotis rufescens ) en México y Estados Unidos y green abalone ( Haliotis fulgens ) en Estados Unidos, todas ellas orejas de mar de gran tamaño (entre 20 y 30 cm en longitud de concha) (Hanh, 1989). Toda la carne de la oreja de mar es comestible, pero tradicionalmente el consumo primario es el del músculo del pie. Su carne es fileteada en las especies de mayor tamaño, y el resto es utilizado por muchos restaurantes para hacer sopas y otros platos preparados. En Japón se consideran “delicatesen” las gónadas, cuando se comen inmediatamente después de separar la concha de la carne en las orejas de mar vivas. En el mercado internacional se puede encontrar la oreja de mar: fresca, enlatada, congelada e incluso seca. Además, también se comercializa su concha, ya que posee un color irisado muy adecuado para la creación de abalorios y adornos. 1.4. LA PRODUCCIÓN DE LA OREJA DE MAR En el mundo Mientras la producción mundial de moluscos aumentó a un ritmo anual del 3,7 %, entre los años 2000 y 2008, la producción de orejas de mar aumentó a un ritmo anual del 39,9 %, desde una producción de 2.800 t en el año 2000 hasta 39.000 t en el año 2008 (FAO, 2010). La

La producción de la acuicultura europea se estima en aproximadamente 150 t anuales (González y Malo de Molina, 2002). Irlanda introdujo y comenzó la producción de las especies Haliotis tuberculata y Haliotis discus hannai en la década de los 70, y junto a las Islas del Canal de La Mancha, lideran hoy en día la producción europea (Huchette y Clavier, 2004). En Francia se cultiva Haliotis tuberculata , destinada principalmente a cubrir las demandas de los restaurantes parisinos (Huchette y Clavier, 2004). En Europa, la mayor parte de la producción se destina a Francia, donde la producción local no abastece la demanda (González y Malo de Molina, 2002). En España Según González y Malo de Molina (2002), la producción española de oreja de mar se ha centrado en Galicia mediante su marisqueo, y surge como consecuencia de su atractivo precio en los mercados asiáticos a principios de la década de los 90. Sin embargo, a partir de 1993, esta actividad permaneció paralizada nueve años debido a la presencia de la toxina PSP en las rías gallegas. Desde esa fecha sólo tres cofradías realizaron la extracción de Haliotis tuberculata , aunque varias más contaban con planes de explotación. La forma de comercialización de la oreja de mar consistía en subastar previamente, en lonja, una determinada cantidad y fijar un precio mínimo, a partir del acuerdo entre el mariscador y los compradores. Una vez fijados el precio y la cantidad, se realiza la extracción. Desde diciembre de 2005, en todos los análisis realizados se vuelve a superar los límites legales de contenido en PSP, lo que impide su comercialización. En el presente año, 2011, la empresa Galician Marine Aquaculture S.L. tiene previsto comenzar a cultivar en Galicia las especies Haliotis tuberculata y Haliotis discus hannai (especie introducida). 1.5. LA OREJA DE MAR CANARIA La oreja de mar presente en Canarias se conoce popularmente como almeja canaria, lapa burra u orejón. En realidad, es una sub-especie de Haliotis tuberculata (especie autóctona de Europa) y su nombre científico es Haliotis tuberculata coccinea (Reeve, 1846) sinonimia de Haliotis coccinea canariensis (Nordsieck, 1975) (Moro et al. , 2003) (Figura 5). Su distribución geográfica es Macaronésica, aunque se han encontrado algunos ejemplares en Melilla y Málaga (Geiger, 2000). En Canarias, la almeja canaria es una especie ubiquista y muestra una gran selección por el hábitat; aparece en pedregales formados por

Este hecho se debía principalmente a dos razones: por una parte, a la fuerte degradación de las costas canarias (con numerosos vertidos de todo tipo y edificaciones), sobre todo en el caso de Gran Canaria y Tenerife, y por otra a la gran presión marisquera en todas las islas excepto en El Hierro y en La Palma. En aquella época la almeja canaria se cotizaba a 48 € / kg. Desde el año 2001 esta especie se encuentra en el Catálogo Nacional de Especies Amenazadas (BOC-2001/097; BOE-2011/043). Es entonces cuando el Gobierno de Canarias se interesa en desarrollar su cultivo, y el Grupo de Investigación en Acuicultura (ICCM-ULPGC), con la financiación de varios proyectos nacionales y europeos, realiza experiencias de inducción a la puesta (Peña, 1986), cultivo larvario (Courtois de Vicose et al ., 2007, 2010; Courtois de Vicose, 2011) y engorde (Toledo et al ., 2000; Viera et al. , 2005, 2011). Estas experiencias han demostrado la viabilidad de su cultivo y la potencialidad comercial del mismo. 1.6. ASPECTOS BIOLÓGICOS DE LA OREJA DE MAR 1.6.1. Descripción anatómica La descripción anatómica se ha realizado de acuerdo a FAO (1990), y Pérez y Moreno (1991). La característica más llamativa del género Haliotis es la forma de la concha, similar a la de una oreja. El color de la concha depende en gran medida del tipo de roca en que habite y de las algas que componen su dieta (FAO, 1995; Liu et al ., 2009), y es el resultado de la adaptación y evolución de la especie como mecanismo de defensa contra la depredación en el medio natural (Liu et al., 2009). La concha contiene una espira corta formada por tres o cuatro vueltas, de las cuales la última es muy amplia y presenta una serie de orificios redondeados u ovales, alrededor de los cuales la superficie de la concha se eleva ligeramente (Figura 5). Por cada orificio aflora al exterior un tentáculo paleal, el cual funciona como poro respiratorio y canal de excreción de desechos, y de liberación de huevos y esperma. A medida que el animal crece, se forman nuevos orificios, mientras que los más antiguos situados en la parte posterior se sellan con carbonato cálcico. Haliotis tuberculata coccinea presenta 5 o 6 poros abiertos. La superficie de la concha presenta crestas con forma de espiral, donde las estrías de crecimiento son apreciables. La abertura oblicua, es muy amplia y el labro se encuentra ligeramente dentado. La superficie interna de la concha contiene un grueso estrato de nácar. La característica ventral más llamativa es el músculo del pie. La parte dorsal del pie está rodeada lateralmente por un collar llamado epipodium, el cual contiene pequeños órganos
– Introducción General – 11 sensoriales y tentáculos especializados, que permiten un contacto sensorial químico y táctil con el medio que rodea al animal (Figura 5). La parte ventral del pie tiene la capacidad de contraerse fuertemente y aparece con elevada mucosidad, de esta forma, la oreja de mar es capaz de adherirse fuertemente a las rocas u otras superficies duras. La parte anterior del pie es la cabeza e incluye la boca, dos ojos y dos tentáculos cefálicos largos con capacidad para retraerse (Figura 5). El aparato digestivo se encuentra en la parte izquierda del músculo aductor, localizado en la mitad del cuerpo (Figura 6). Los órganos digestivos consisten en la lengua, tráquea, estómago, intestino recto y el ano. Debido a la forma espiral del cuerpo, los órganos del digestivo aparecen curvados, y la lengua y el ano se encuentran en la misma orientación. La lengua tiene forma ovalada y en ella se encuentran el lóbulo de calyx, la rádula y las glándulas salivares. La tráquea del abalón es larga y estrecha. El estómago tiene forma de V y se encuentra posterior a la tráquea. El hígado (hepatopáncreas) está localizado en la parte derecha del músculo aductor, protuberante y con forma de cuerno de buey. El intestino que finaliza siendo el recto, comienza en la parte trasera hacia la parte marginal delantera rodeando la parte izquierda del músculo aductor. El intestino mide aproximadamente 3,27 veces la concha. Las heces y otros materiales de desecho se liberan a través de los poros respiratorios mediante contracciones del músculo del pie. El sistema nervioso está poco desarrollado (Figura 6). Contiene cinco pares de ganglios (cerebral, pedal, ventral, branquial e intestinal) y una conjunción de nervios que están conectados longitudinal y transversalmente. El aparato respiratorio presenta dos branquias localizadas inmediatamente debajo de los poros de la concha y funcionan como órganos respiratorios (Figura 6). Se encuentran en el centro de la cámara respiratoria, y la de la derecha es más grande que la de la izquierda. La parte izquierda y el fondo de la cámara respiratoria está cerrada, de esta forma, el paso del agua hacia las branquias se realiza desde la parte superior y derecha. El agua de mar pasa a través de las branquias (intercambio gaseoso) y se dirige hacia los poros respiratorios de la concha junto con otros productos de excreción procedentes del ano. En el aparato circulatorio (Figura 6), el corazón, localizado en la cavidad pericardial, consta de un ventrículo y dos aurículas. La sangre desde el corazón se transporta a través de las venas situadas entre los órganos por el sistema circulatorio, primero se dirige hacia las arterias de las branquias y una vez oxigenada, llega a la aurícula a través de las venas branquiales.

Estos poliquetos perforadores se han encontrado tanto en poblaciones naturales de orejas de mar, como en ejemplares cultivados (Sphepherd, 1973; Clavier, 1992a; Ruck y Cook, 1998; Lleonart et al ., 2003b), y afectan a la oreja de mar reduciendo significativamente la condición de la carne (Simon et al ., 2006; Sato-Okoshi et al ., 2008), causando incluso la muerte en infestaciones avanzadas (Lleonart et al ., 2003a). Si bien, en ejemplares cultivados, han sido numerosos los intentos para erradicar este tipo de infestación, actualmente no existe ningún tratamiento efectivo que no cause daños a la oreja de mar, y que demuestre una eficacia total contra los gusanos (Shields et al ., 1998; Kuris y Culver, 1999; Lleonart et al ., 2003a; Leighton, 1998; Moore et al ., 2007). Otro tipo de parasitismo, que pueden afectar a la oreja de mar, es la infestación de las gónadas por trematodos (Crofts, 1929; Bower, 2003), y también protozoos parásitos del género Haplosporidian , éstos últimos causantes de grandes mortalidades en la especie Haliotis iris presente en Nueva Zelanda (Diggles et al ., 2002). Enfermedades víricas La amiotrofia es una patología producida por un virus, que ocasiona importantes pérdidas económicas a los cultivadores de semillas de Haliotis discus en Japón (Nakatsugwa et al ., 1999). También se ha observado elevadas mortalidades por infecciones víricas en Haliotis diversicolor (Wang et al ., 2000, 2004) Enfermedades bacterianas A finales de la década de los 90, se produjo una mortalidad masiva en las poblaciones naturales de la oreja de mar francesas, que acabó con el 60 – 80 % de la población afectada (Nicolas et al ., 2002). Esta mortalidad fue causada por Vibrio carchariae , responsable también de importantes pérdidas en cultivos de Haliotis diversicolor supertexta (Nicolas et al ., 2002). Otras enfermedades bacterianas son las causadas por Vibrio fluvialis en Haliotis discus hannai , conocida como la enfermedad pustular (pustular disease) (Li et al ., 1998), Vibrio parahaemolyticus en Haliotis diversicolor supertexta en Taiwan (Liu et al ., 2000), Vibrio harvey y Vibrio splendidus en Haliotis rubra y Haliotis laevigata en Tasmania (Bower, 2003). Entre los tratamientos utilizados contra las infecciones bacterianas, Dixon et al. (1991) encuentran efectiva la exposición a agua ozonizada, y un tratamiento con un antibiótico de amplio espectro. El caso más conocido e investigado de mortalidades masivas, originadas por bacterias en especies de orejas de mar, es la enfermedad del síndrome de deterioro (withering
20 – Introducción General – syndrome). Se detectó por primera vez en California a mediados de los ochenta y a finales de esa década, las poblaciones de Haliotis cacherodii , Haliotis rufescens y Haliotis corrugata disminuyeron entre un 51 y 98 % en las Islas del Canal de California (Haaker et al ., 1992). La bacteria causante de esta enfermedad fue Candidatus xenohaliotis californiensis (Friedman et al ., 2000), y existe una relación entre las elevadas temperaturas y el incremento de mortalidad, de hecho, esta enfermedad se asocia al fenómeno El Niño (Steinbeck et al ., 1992; Friedman et al ., 1997). El contagio tiene lugar mediante el agua, sin necesidad de contacto físico entre individuos (Moore et al ., 2000), y parece que el tratamiento a base de oxitetraciclina permite disminuir las pérdidas en granjas de cultivo (Friedman et al ., 2003). 1.7. EL CULTIVO DE LA OREJA DE MAR El cultivo de la oreja de mar comenzó hace más de 80 años en Japón (Elbert y Houk, 1984), y se practica actualmente con éxito en varios lugares del mundo, siendo de gran importancia para la economía de muchos países (Cook y Gordon, 2010). Si bien, la tecnología de cultivo de algunas especies de oreja de mar está totalmente desarrollada (FAO, 1990), a la hora de cultivar nuevas especies, como es el caso de Haliotis tuberculata coccinea , hay que adaptar la tecnología existente a las condiciones específicas de la especie, y al lugar en donde se desarrolla el cultivo. El cultivo de oreja de mar se puede dividir en tres fases: el acondicionamiento de reproductores, la obtención de semilla y el engorde hasta la talla comercial. Las dos primeras fases se realizan en el criadero, mientras que la última en la granja de engorde. El criadero suele ser una instalación cubierta situada en tierra firme, y requiere del control de las condiciones de cultivo: temperatura, filtración y de la esterilización de grandes volúmenes de agua. El producto final de un criadero es la semilla de oreja de mar. Se denomina semilla a los ejemplares de una talla aproximada de 10 mm en longitud de concha, que son capaces de alimentarse de macroalgas frescas. Normalmente el acondicionamiento de reproductores se realiza en una sala destinada a este fin, donde se mantienen los reproductores y se realizan las puestas inducidas y la fertilización. La eclosión de los huevos y el cultivo larvario (fase pelágica), se realiza normalmente en otra sala en tanques diseñados para este fin. Cuando se acerca el momento de la fijación y de la metamorfosis, las larvas son transferidas a otros tanques provistos de placas de plástico para aumentar la superficie de fijación, cubiertas de films de diatomeas como sustratos de fijación y alimentación para el crecimiento de las postlarvas (Takami et al ., 1997). Esta es una fase crítica del cultivo, donde la mortalidad post-larvaria

Recientemente se han desarrollado otros métodos para la fijación, como la utilización de la macroalga verde Ulvella lens en las placas de fijación (Takahashi y Koganezawa, 1988; Seki, 1997; Daume et al ., 2000), y la utilización de sprays compuestos principalmente de agar-agar y microalgas secas (Stott et al ., 2004), con el fin de optimizar y minimizar las tareas en esta fase de cultivo. En un estudio realizado por Courtois de Vicose et al . (2010), se prueban diferentes especies de diatomeas frente a Ulvella lens y a un alga roja incrustante para inducir la fijación y el crecimiento de post-larvas de Haliotis tuberculata coccinea , y muestra que si bien, las mayores tasas de fijación se obtuvieron en placas de plástico envejecidas con algas coralinas incrustantes, a la hora de una producción masiva de semilla es más viable la utilización de la macroalga bentónica Ulvella lens . Así mismo, Courtois de Vicose (2011) estudia el potencial de las esporas de la macroalga Ulva rígida para inducir la fijación, y como alimento de post-larvas de Haliotis tuberculata coccinea y obtiene resultados satisfactorios. La fase de engorde comprende el cultivo de las semillas hasta alcanzar la talla comercial. Esta etapa del cultivo permite una mayor diversidad de las instalaciones, pudiendo realizarse en tierra firme (en tanques), o en diferentes tipos de estructuras en el medio marino (jaulas, barriles, nasas, etc.) (Gónzalez y Malo de Molina, 2002; Wu et al ., 2009). Recientemente, se ha prestado atención al cultivo en sistemas integrados por oreja de mar-macroalgas-oreja de mar (Robertson-Andersson et al ., 2008; Bolton et al ., 2009) y peces-macroalgas-oreja de mar (Neori et al ., 2000, 2007). El cultivo de macroalgas en sistemas integrados es interesante porque se produce alimento para el engorde de oreja de mar, y se eliminan parte de los desechos (nitratos y fosfatos) asociados al cultivo de oreja de mar y de peces y además es relevante, sobre todo en el caso de Canarias, porque no existe una producción de macroalgas a nivel comercial. Estudios recientes realizados por Viera et al . (2005, 2011) muestran que las macroalgas ( Ulva rígida , Gracilaria cornea , Hypnea spinella e Hypnea musciformis ), producidas en un sistema de biofiltración, son ricas en proteínas, y que su composición nutricional satisface los requerimientos de proteínas, lípidos e hidratos de carbono de la oreja de mar, resultando en un crecimiento satisfactorio y supervivencia de Haliotis tuberculata coccinea . 1.8. ACONDICIONAMIENTO DE REPRODUCTORES Se denomina acondicionamiento de reproductores al mantenimiento de individuos adultos bajo condiciones favorables, que garanticen una maduración correcta de las gónadas, y puestas exitosas en el momento y condiciones deseadas.
22 – Introducción General – Antes de que los ejemplares adultos sean capaces de reproducirse, es necesaria la maduración de sus órganos sexuales. Las especies de oreja de mar (ej. Haliotis tuberculata , Haliotis iris , Haliotis discus hannai , Haliotis midae y Haliotis laevigata ), pertenecientes a zonas de agua templada (Francia, Nueva Zelanda, Japón, Sudáfrica, Australia etc.), normalmente presentan una fase de reposo sexual durante el invierno, y el desarrollo de la gónada coincide con un cambio estacional de temperatura (un incremento o descenso), asociado a un incremento en la disponibilidad de alimento en la naturaleza (Shepherd y Laws, 1974; Hayashi, 1980a; Ault, 1985; Lleonart, 1992; Uki, 1989; Campbell et al ., 2003). En cambio, las especies de orejas de mar (ej. Haliotis varia y Haliotis asinina ) de zonas tropicales (Tailandia, Filipinas, etc.), muestran un ciclo reproductivo continuo a lo largo del año (Bussarawit et al ., 1990; Sobhon et al ., 1999; Jebreen et al ., 2000). El Experimento I del presente trabajo, estudia la biología reproductiva de una población natural de Haliotis tuberculata coccinea , lo que permitirá determinar el ciclo reproductivo y los factores que lo controlan, la talla adecuada para el acondicionamiento de reproductores y la mejor época para la inducción a la puesta. Estos conocimientos contribuirán a una mejor planificación en las tareas de un criadero (inducción al desove, cultivo larvario y preparación de los tanques del cultivo post-larvario). 1.8.1. Obtención de reproductores Para llevar a cabo una producción de oreja de mar, en primer lugar es necesario formar un stock de reproductores. Los reproductores pueden capturarse del medio natural, o pueden obtenerse en las granjas de cultivo escogiendo los individuos que han alcanzando la madurez sexual (Hooker y Morse, 1985; FAO, 1990). La selección de un stock de hembras reproductoras, que tengan una alta fecundidad y produzcan huevos de calidad en las puestas, es importante para el éxito del cultivo, puesto que reduce la necesidad de mantener un grupo numeroso de hembras, y como consecuencia, minimiza la cantidad de alimento necesario y las labores para mantener el stock de reproductores (Daume, 2007). En el cultivo de la oreja de mar, ha sido práctica habitual capturar los reproductores en la naturaleza, en el momento que presentan su máximo grado de maduración sexual (Fleming, 2001). Este es el caso de Nueva Zelanda, Estados Unidos y Sudáfrica con las especies Haliotis iris , Haliotis rufescens y Haliotis midae respectivamente, y permite por un lado tener un periodo corto de acondicionamiento, y por otro una mejor respuesta a la inducción al desove puesto que se utilizan individuos muy maduros (Tong et al ., 1992; Ault, 1985; Fleming, 2001; Najmudeen y Victor, 2004). Sin embargo, recientemente hay una mayor tendencia, por parte de
– Introducción General – 23 investigadores y productores comerciales, a realizar el acondicionamiento de reproductores, en condiciones de cultivo controladas durante todo el año (Daume y Ryan, 2004; Grubert y Ritar, 2005; Freeman et al ., 2006). Este sistema permite elegir la época de la maduración de las gónadas controlando los parámetros de cultivo (ej. temperatura), de esta forma, se puede elegir la época del año para la inducción a la puesta, y como resultado se rentabiliza mejor las instalaciones (Lleonart, 1992). Así mismo, permite la aplicación de programas de mejora genética (Daume, 2007). A día de hoy en Canarias Haliotis tuberculata coccinea está protegida legalmente (BOC2001/097), y su captura no se puede realizar de forma habitual en el medio natural. En este sentido, lo más viable es mantener un stock de reproductores permanente en las instalaciones de cultivo. 1.8.2. Condiciones de cultivo Durante el acondicionamiento es necesario que los reproductores se encuentren en óptimas condiciones, para que inviertan su energía en la producción y maduración de los gametos, y no en mitigar los efectos del estrés (Hooker y Morse, 1985; Fleming, 2001). En este sentido la temperatura, la alimentación, la iluminación, la calidad del agua y los cuidados de los animales son factores que hay que tener en consideración. Temperatura Cuando la alimentación es la adecuada, la temperatura es el principal factor exógeno que afecta la maduración de la gónada (Webber y Giese, 1969; Hahn, 1989). Kikuchi y Uki (1974b), fueron los primeros investigadores que demostraron la relación entre la temperatura y la maduración sexual en la oreja de mar, y desarrollaron el llamado EAT (Effective Accumulative Temperature), expresado como EAT (ºC/día). Este índice establece el momento adecuado para inducir la puesta, en base a la temperatura empleada en el acondicionamiento y la temperatura cero biológica (donde no se observa la maduración sexual) específica para cada especie de oreja de mar. De esta forma, dependiendo de la temperatura utilizada durante el acondicionamiento, se puede acortar el periodo para alcanzar la madurez de las gónadas. La manipulación de la temperatura permite la obtención de puestas en cualquier momento del año (Grubert y Ritar, 2005). Este método se ha aplicado en varias especies de oreja de mar, y parece que es totalmente efectivo en las especies pertenecientes a aguas templadas con un periodo de puesta en verano (Uki y Kikuchi, 1984; Hanh, 1989; Fleming, 2001; Grubert y Ritar, 2004). Sin embargo, en las especies de regiones tropicales, como Haliotis asinina , además de la temperatura, la ovulación y el desove aparecen fuertemente asociadas al régimen de mareas
24 – Introducción General – vivas (Jebreen et al ., 2000), incluso en condiciones de cultivo (Counihan et al ., 2001), lo que permite, si se cuenta con un número considerable de reproductores, la posibilidad de obtener puestas durante todo el año, sin la necesidad de controlar la temperatura durante el acondicionamiento. Alimentación Una alimentación adecuada es esencial para un desarrollo óptimo de la gónada. De hecho, se ha observado que la disponibilidad de alimento es una de las causas de la variación del ciclo reproductivo de poblaciones diferentes, incluso cercanas, de una misma especie de oreja de mar (Hayashi, 1980a; Hanh, 1989). La bibliografía existente sobre los requerimientos nutritivos durante el acondicionamiento es escasa. Estudios recientes muestran, en Haliotis laevigata , la importancia de la nutrición en la composición y tamaño de los huevos, y en la frecuencia de puestas (Daume y Ryan, 2004; Freeman et al ., 2006; Graham et al ., 2006). Es sabido que ciertos ácidos grasos específicos, incorporados en la dieta, juegan un papel importante en la maduración de la gónada de la oreja de mar (Uki y Watanabe, 1992; Bautista-Teruel et al ., 2001; Nelson et al ., 2002), y que 3 meses de alimentación son suficientes para modificar la composición bioquímica de los huevos (Daume y Ryan, 2004). Entre los ácidos grasos, cabe destacar el ácido Araquinódico (20:4n-6), principal precursor de las hormonas tipo Prostaglandinas que influyen en la reproducción de moluscos (Osada et al. , 1989). Así mismo, otros estudios muestran que la cantidad de proteína y de lípidos incorporados en la dieta, afectan al desarrollo de la gónada y a la fecundidad (Jackson et al ., 2001; Bautista-Teruel et al ., 2001). Si bien se han diseñado dietas artificiales específicas para el acondicionamiento de reproductores (Daume y Ryan, 2004; Daume, 2007), se recomienda la utilización de macroalgas frescas dada la poca estabilidad del pienso en el agua (Daume, 2007). De hecho, la práctica más común es ofrecer a los reproductores una dieta basada en una mezcla de algas frescas durante la fase de acondicionamiento (Fleming, 2001). Esta mezcla varía en función de las preferencias alimenticias de la especie y la disponibilidad de algas locales, de esta forma, especies como: Haliotis discus hannai son acondicionadas con Undaria spp. y Laminaria spp. , Haliotis rufescens con Nererocystis spp. (Fleming, 2001) y Haliotis tuberculata con algas del tipo Palmaria spp. (FAO, 1995). En Canarias, debido a que la cantidad disponible de algas, de arribazón, no es abundante, y no se cultivan de manera industrial, se hace necesario buscar un método alternativo a la obtención de algas del medio natural. Una alternativa es el cultivo de
– Introducción General – 25 macroalgas tanto en tanques como en estructuras marinas (Neori et al ., 2000; Viera et al ., 2005, 2011; Hwang et al ., 2009). Además, el cultivo de algas, bajo condiciones controladas, da la posibilidad de alterar la calidad nutritiva de las mismas (Neori et al ., 2000). En el Experimento II del presente trabajo, se realiza el cultivo en tanques del alga Gracilaria cornea , y tiene como objetivo principal determinar el efecto del porcentaje de proteína, en la dieta de los reproductores, sobre la reproducción de Haliotis tuberculata coccinea . Este experimento nos proporcionará información sobre la viabilidad de producir alimento adecuado para la fase de acondicionamiento, y ampliará el conocimiento sobre los requerimientos nutricionales de los reproductores. Iluminación Un estudio realizado por Setyono (2004), muestra en Haliotis asinina que el fotoperiodo, durante el periodo de acondicionamiento no influye en la maduración de la gónada, y se observa una correcta maduración tanto en un régimen de 24 horas de luz como, en uno 24 horas de oscuridad. En cambio, se ha observado que en puestas inducidas, el tiempo requerido para que los reproductores desoven, es menor si la inducción se produce una hora antes del inicio del periodo de la fase oscura utilizado durante el acondicionamiento (Uki y Kickuchi, 1982), es decir, al atardecer si se mantienen los reproductores bajo condiciones naturales de fotoperiodo, o antes de las horas de oscuridad, si se mantienen los reproductores bajo un régimen de fotoperiodo artificial. La práctica más extendida es el acondicionamiento en un régimen de 12 horas luz y 12 horas oscuridad (Hanh, 1989; Fleming, 2001; Grubert y Ritar, 2002, 2004, 2005; Daume, 2007). Así mismo, es posible alterar el fotoperiodo natural con el propósito de obtener puestas durante la mañana (Grubert y Ritar, 2005). Teniendo en cuenta que la oreja de mar es un animal foto-fóbico (Hanh, 1989), se aconseja durante el periodo de acondicionamiento mantener una intensidad de luz baja, entre 50 y 150 Lux, medida en el fondo del tanque (Fleming, 2001; Grubert y Ritar, 2004, 2005; Daume, 2007). Así mismo, se aconsejable incorporar refugios en los tanques de acondicionamiento, imitando las rocas y recovecos de la naturaleza donde la oreja de mar vive durante las horas de luz. Calidad del agua Un acondicionamiento adecuado requiere el mantenimiento de una buena calidad del agua, con el objetivo de mitigar el estrés de los reproductores. Los factores que determinan la calidad del agua son: un diseño adecuado de los tanques, una alta tasa de renovación del agua, una apropiada densidad de cultivo, una apropiada estrategia de alimentación y la eliminación regular de desechos (Hanh, 1989; Fleming, 2001; Grubter y Ritar, 2002).
26 – Introducción General – El diseño de los tanques de acondicionamiento debe permitir la eliminación de los desechos de cultivo (heces, mucus y restos de comida), perturbando los reproductores lo mínimo posible. Por ejemplo, en Japón es habitual el uso de tanques provistos de una cortina de aire a lo largo del fondo, que suspende continuamente los desechos del cultivo en la columna de agua, permitiendo su eliminación con la salida de agua del tanque (Fleming, 2001). Así mismo, se pueden incorporar cestas suspendidas en los tanques con el objetivo de separar los animales de las heces y restos de comida. Sin embargo, para una correcta higiene, es necesaria la limpieza manual periódica de los tanques sifonando el fondo del tanque y/o vaciándolo completamente (Fleming, 2001; Grubert y Ritar, 2002). Es importante entonces, que el diseño del sistema permita un llenado rápido del tanque después de la limpieza, para perturbar los reproductores lo mínimo posible. La regularidad de la limpieza manual depende principalmente de la estrategia de alimentación; la utilización de pienso produce más desechos que la utilización de macroalgas y como consecuencia requiere una limpieza más regular. En ambos casos, los niveles de nitritos y amonio no deben superar 1,12 mg NO 2 -N / l y 0,042 mg NH 3 -N / l, respectivamente (Harris et al ., 1997). La densidad de cultivo se mantiene más baja que en la fase de engorde. Por ejemplo, en Haliotis tuberculata se aconseja una densidad de cultivo de 10 g/l (González y Malo de Molina, 2002). Con Haliotis laevigata y Haliotis rubra se mantienen entre 15 y 20 reproductores de 100 a 120 mm en longitud de concha en tanques de 150 litros (Grubert y Ritar, 2002). La densidad de cultivo debe ser la apropiada para mantener una la saturación de oxigeno disuelto por encima de un 80 % (Uki y Kikuchi, 1981; Grubter y Ritar, 2002). Cuidado de los animales Es importante que los ejemplares que van a producir la nueva generación de orejas de mar se encuentren sanos. En este sentido, se debe evitar cualquier herida en los animales y la proliferación de enfermedades y/o infestaciones de parásitos. Las orejas de mar se fijan fuertemente a los refugios, y en las tareas que precisen su desprendimiento (ej. muestreos para elegir los reproductores, para controlar el crecimiento, el estado de salud, etc.), la manipulación mecánica (ej. uso de paletas o espátulas) puede causar heridas en el músculo del pie. Estas heridas pueden ser mortales, puesto que la oreja de mar posee una baja capacidad de cicatrización y ausencia de un mecanismo de coagulación de la sangre (Armstrong et al ., 1971; Genade et al ., 1988). En este sentido, el uso de anestésicos es una alternativa segura (White et al ., 1996). Entre ellos: el sulfato de magnesio, el 2phenoxyethanol, el pentobarbitúrico y la benzocaina (Sagara y Ninomiya, 1970; Prince y Ford,
– Introducción General – 27 1985; McShane y Smith, 1988; Hanh, 1989; Tong et al ., 1992; White et al ., 1996; Aquilina y Roberts, 2000; Sharma et al ., 2003), siendo todas ellas sustancias químicas no degradables. Las especies sometidas a cultivo son un medio apropiado para el desarrollo de enfermedades e infestaciones, ya que los animales se encuentran hacinados en un espacio limitado. En Canarias, abundan diferentes especies de poliquetos perforadores, asociados a sustratos que contienen carbonato cálcico (Pascual y Núñez, 1999), y al igual que sucede en otros lugares, donde se desarrolla el cultivo de oreja de mar (Lleonart et al ., 2003b; Simon et al ., 2006), puede darse el caso de infestaciones masivas. Actualmente no existe ningún método eficaz que elimine la infestación por poliquetos y no dañe la oreja de mar (Shields et al ., 1998; Kuris y Culver, 1999; Lleonart et al ., 2003a; Leighton, 1998; Moore et al ., 2007). El Experimento III del presente trabajo tiene la finalidad de probar el aceite de clavo como anestésico natural, en las tareas que requieran el desprendimiento de los individuos de los refugios, y el Experimento IV, tiene como objetivo la identificación de la especie de poliqueto que infestó, en 2008, a la oreja de mar en las instalaciones del Instituto Canario de Ciencias Marinas, y probar la eficacia del mebendazol como antihelmíntico en individuos adultos de Haliotis tuberculata coccinea . 1.8.3. La inducción a la puesta y fertilización Verificación del estado de maduración Durante el acondicionamiento, es útil emplear un “índice de maduración” que permita conocer la evolución gonadal de los ejemplares, y el momento en que están preparados para inducir la puesta artificialmente. Entre los índices más habituales se encuentra del descrito por Kikuchi y Uki (1974b), que establece la madurez gonadal en 4 estados que se designan visualmente: en el estado 0 no se distingue el sexo del animal y a medida que madura, la gónada aumenta en volumen y aparece más protuberante hasta alcanzar su estado de máxima madurez (estado 3). Sin embargo, no todos los individuos alcanzan el estado 3 de maduración durante el acondicionamiento (Fallu, 1991), por lo que es práctica habitual inducir la puesta a animales en estado 2 avanzado. La ventaja de la utilización de este índice reside en que no es un método invasivo, sin embargo, es subjetivo y no asegura la puesta inducida en todas las especies de oreja de mar (ej. Haliotis discus hannai ) (Fleming, 2001). El método más fiable para verificar el estado de maduración es el realizado mediante técnicas histológicas: medida del diámetro de los oocitos y morfología de la gónada (Hanh, 1989). En la clasificación propuesta para Haliotis asinina por Capinpin et al . (1998), se considera
28 – Introducción General – un ovario maduro cuando la mayoría de oocitos presentan un diámetro mayor de 125 µm y se observan oocitos libres de 150 µm, y se considera un testículo maduro, cuando se observan espermatozoides densamente empaquetados. Sin embargo, las técnicas histológicas implican el sacrificio de animales y puesto que la oreja de mar no posee un sistema de cicatrización eficiente (Armstrong et al ., 1971, Genade et al ., 1988), no es aconsejable tampoco la realización de una biopsia de la gónada puesto que el animal puede ser irrecuperable. En el Experimento V del presente trabajo se realiza un control histológico del índice visual propuesto por Kikuchi y Uki (1974b), en la gónada femenina de Haliotis tuberculata coccinea . Este estudio permitirá conocer el porcentaje de los diferentes estados de oocitos en los diferentes grados de maduración visual. Esta información permitirá determinar el momento más adecuado para inducir la puesta. Técnicas de inducción a la puesta Si bien, las puestas naturales son aprovechables en un criadero de oreja de mar, la inducción a la puesta presenta la ventaja de obtener huevos en un momento predecible que permite al granjero/a preparar los tanques larvarios y post-larvarios, así como obtener puestas durante las horas matinales de trabajo. En condiciones de cultivo, la liberación de los gametos se induce mediante un estímulo artificial. Hoy en día se conocen numerosas técnicas para inducir el desove en la oreja de mar (Hanh, 1989): Método físico : se basa en la exposición prolongada de los reproductores al aire y/o en someter a los animales a cambios de temperatura (elevando la temperatura del agua bruscamente entre 3-6 ºC y bajándola después gradualmente durante 3 horas, hasta alcanzar la temperatura ambiente). Método químico : se basa en un aumento de pH y/o en la dilución de hormonas (Kabir et al ., 2001) o gametos de la misma especie, en el agua que contiene los reproductores. Exposición a Rayos Ultravioleta (Kikuchi y Uki, 1974a): consiste en hacer pasar el agua del cultivo que abastece al tanque de puesta, a través de una cámara con una bombilla de UV (con una longitud de onda de 254 nm), de donde sale desionizada. Es uno de los primeros métodos desarrollados y se obtienen buenos resultados para todas las especies de oreja de mar (Fallu, 1991).

36 – Material y Métodos Generales – 2.1. TANQUES EXPERIMENTALES 2.1.1. Tanques de 60 l Estos tanques eran de plástico utilizaron para el acondicionamiento de reproductores estantería en dos alturas. Se colocó tanque y separada del fondo lo que permitió separar las heces y restos de comida de los reproductores, y facilitó las labores de l tejas de PVC, que proporcionaron a los individuos refugio. Así mismo, se colocó encima de cada tanque una malla negra de invernadero protección a la luz. Los tanques estaban dotados de un sistema de circuito abierto, el agua, sin filtrar, era conducida por una tubería de PVC de 40 mm de diámetro. La entrada en cada tanque era superior a través de un tubo de polipropileno de 10 mm de diámetro y se reg una llave. La renovación diaria fue de aproximadamente de 48 veces el volumen del tanque. El nivel de agua se controló con un tubo de nivel exterior al tanque. Cada uno de los tanques fue provisto de un tubo aireador de polietileno de 16 mm diámetro perforado y conectado a una tubería general de polietileno de 32 mm de diámetro. El sistema de aireación disponía de llaves de plástico que permitieron regular la entrada de aire. Figura 10 Tanques de 60 l en el Instituto Canario de tanques dispuestos en la estantería. El agua que se bombeó del tanque que recoge los efluentes de la nave de cultivos del ICCM, se suministró sin filtrar a través de una tubería de PVC de 40 mm de diámetro, entró en cada tanque por la parte superior, y se reguló por una llave de 20 mm de diámetro. El nivel de agua se mantuvo con una salida de 32 mm de diámetro, situada en la parte de superior de los tanques y a través de una manguera que desembocaba en una canaleta general. La renovación se mantuvo aproximadamente en 4 volúmenes al día. El suministro de aire se realizó desde el fondo, mediante una manguera perforada de 16 mm de diámetro, y se regulaba con una llave de polietileno de 16 mm de diámetro. 2.1.4. Tanques de 50 l Estos tanques se utilizaron para el mantenimiento de reproductores (en algunos casos), y para la inducción a la puesta por el método de agua irradiada con radiación ultravioleta (Figura 13). Los tanques eran rectangulares de plástico azul y 50 l de capacidad, y se dispusieron en una plataforma de madera. Se introdujeron dos tejas de PVC a modo de refugios en cada tanque que fueron cubiertos con una malla negra para proporcionar a los reproductores una adecuada protección a la luz. El agua fue filtrada hasta 1 µm, e irradiada con radiación ultravioleta a través de un esterilizador (Wedeco GmbH D-4900, Herford, Alemania). El agua se condujo a través de una tubería de PVC de 40 mm de diámetro y fue distribuida en cada tanque, por la parte superior y regulada con una llave de 20 mm de diámetro de PVC. La renovación fue de aproximadamente 5 volúmenes al día. La salida del agua se realizó por debajo de los tanques, conectado esta salida a un tubo de nivel, exterior al tanque, de 32 mm de diámetro de PVC. El desagüe de todos los tanques se realizó sobre una canaleta. Cada uno de los tanques fue provisto de un tubo aireador de polietileno de 16 mm de diámetro perforado y conectado a una tubería general de polietileno de 32 mm de diámetro. El sistema de aireación disponía de llaves de plástico que permitieron regular el flujo. 2.1.5. Cubetas de 2 l Las cubetas se utilizaron en el experimento del control de parásitos externos, y como recipientes en la inducción a la puesta por el método de peróxido de hidrógeno. Estas cubetas eran de plástico transparente y de 2 l de capacidad. En cada cubeta se dispusieron dos tejas de PVC a modo de refugio.

40 – Material y Métodos Generales – 2.1.6. Agua de mar La toma de agua de mar se realizaba a través de un pozo de captación excavado en el espigón del Muelle de Taliarte. El agua era elevada mediante un grupo de bombas que suministraba un caudal de 180 m 3 por hora, y era conducida hasta un depósito regulador principal situado en el exterior en una zona anexa a las instalaciones propias de la planta de cultivo. Todo el sistema de conducción de agua de mar estaba montado en PVC. Desde este depósito el agua de mar era distribuida directamente, por gravedad, a los tanques experimentales de reproductores. La salinidad y el pH del agua de los tanques de reproductores se mantuvieron aproximadamente en 36,70 ‰ y 8 respectivamente. 2.1.7. Aireación El suministro de aire a toda la instalación, para conseguir una oxigenación adecuada del agua, se realizaba mediante tres turbinas ventiladoras (Siemens-Elmo, Alemania), de funcionamiento continuo que suministran aire a baja presión. 2.2. CONDICIONES DE CULTIVO 2.2.1. Producción de alimento En los tanques de 1500 l se cultivaron Gracilaria cornea y Ulva rigida con el fin de producir el alimento para los reproductores. La densidad de inoculación fue de 6 g/l para Gracilaria cornea y 3 g/l para Ulva rígida . Cada dos semanas se cosechó toda la biomasa algal, se eliminó el exceso de agua del alga, se limpió el tanque y se volvió a inocular en la misma densidad inicial. Regularmente fueron eliminados manualmente los epífitos de las algas. En el experimento de nutrición (Experimento II), la macroalga Gracilaria cornea fue cultivada en los tanques de 90 l. Los tanques se inocularon a un densidad de 1000 g de Gracilaria cornea en 90 l de agua. Con el objetivo de conseguir diferentes niveles de proteína en el alga, en cada tanque se realizaron pulsos de fertilización con amonio (sulfato amónico, Canarias Explosivos S.A.) y con fosfato (fosfato monopotásico, Kemira) 3 días a la semana. Para ello, se cerró el flujo de agua durante 6–8 horas y se añadieron las siguientes concentraciones en cada tanque: 33.3 mg/ l -1 día -1 NH 4 SO 3 y 3.3 mg l -1 día -1 NaH 2 PO 4 , 11.1 mg l -1 día -1 NH 4 SO 3 y 1.1 mg l -1 día -1 NaH 2 PO 4 y 1.1 mg l -1 día -1 NH 4 SO 3 y 0.1 mg l -1 día -1 NaH 2 PO 4 . Cada dos semanas se cosechó toda la biomasa algal, se eliminó el exceso de agua del alga, se limpió el tanque y se volvió a inocular con la misma densidad inicial. Regularmente los epífitos de Gracilaria cornea fueron eliminados manualmente.

Mantenimiento de los reproductores En el presente trabajo, el mantenimiento de reproductores se ha realizado con una densidad de 10 animales por tanque (en los tanques de 50 l y 60 l). La alimentación consistió en macroalgas frescas ( Gracilaria cornea y Ulva rígida ) cultivadas en los tanques de 1500 l, excepto en el Experimento II que se utilizó únicamente Gracilaria cornea . La limpieza y alimentación se realizaron una vez por semana. Este día se vaciaron los tanques, se eliminó el alimento no ingerido, se limpió el tanque con agua dulce y se introdujeron las macroalgas frescas. La temperatura del agua y el fotoperiodo no se modificaron. 2.3. TRATAMIENTO ESTADÍSTICO DE LOS DATOS Los resultados obtenidos se han expresado siempre como media ± desviación estándar de la media. Los datos de un mismo experimento se compararon estadísticamente utilizando el análisis de la varianza (ANOVA). Una vez habían sido detectadas diferencias estadísticamente significativas con el ANOVA, las diferencias entre medias fueron puestas de manifiesto mediante el test de comparación múltiple de las medias de Tukey (como criterio general se tomó el 5 % de nivel de significación) (Sokal y Rohlf, 1996). Los datos se analizaron utilizando el programa estadístico STATGRAPHICS (versión 3.1 Plus for Windows; Graphic Software Systems, Inc. USA). Para correlacionar dos variables, se utilizaron regresiones lineales y polinomiales (Sokal y Rohlf, 1996). En el caso del estudio de la población natural, Experimento I, las diferencias entre medias fueron puestas de manifiesto mediante el test de comparación Kolmogorov-Smirnov y el test t-Student. El estudio de crecimiento se realizó mediante el método descrito por Pauly (1983) mediante el programa informático FAO-ICLARM Stocks Assessment Tools (FISAT), versión 1.2) 2.4. MÉTODOS ANALÍTICOS DE COMPOSIÓN Los análisis de composición se realizaron en el laboratorio del Instituto Universitario de Sanidad Animal y Seguridad Alimentaria (IUSA) de la Universidad de Las Palmas de Gran Canaria (ULPGC). En el Experimento II, se diseccionaron los animales al finalizar el experimento de nutrición. Se tomaron muestras de gónada, vísceras y músculo del pie. Estas muestras se lavaron cuidadosamente con agua destilada. Posteriormente fueron secadas con papel de filtro y guardadas a – 80 ºC en atmósfera de nitrógeno. También se guardaron muestras de las algas
42 – Material y Métodos Generales – utilizadas como dieta, y antes de los análisis, fueron liofilizadas 24 horas en el liofilizador Cryodos-50 (Telstar, España). En este apartado se describen los métodos de análisis utilizados con las diferentes muestras. Todos los análisis se hicieron por triplicado para asegurar un mínimo error analítico en los datos obtenidos. Las muestras se homogenizaron manualmente o con el homogenizador Pulverisette 6 (Telstar, España). 2.4.1. Humedad La humedad de las muestras se determinó por desecación en estufa a 105 ºC, hasta obtener peso constante, según el método de la Asociación Oficial de Analistas Químicos (AOAC, 2005). 2.4.2. Ceniza El contenido en ceniza fue determinado mediante la incineración de la muestra en horno de mufla a 450 ºC, hasta peso constante según el método de la AOAC (2005). 2.4.3. Proteínas El contenido en proteínas (AOAC, 2005) se calculó a partir de la composición en nitrógeno total de las muestras determinada mediante la técnica Kjeldhal. El método consiste en la digestión de las muestras con ácido sulfúrico concentrado a 400 ºC, en presencia de un catalizador de mercurio con el aparato Tecator Digestor 20 (Foss, Suecia), seguido de una destilación con Na(OH) al 40%, con ácido bórico como sustancia receptora, en una unidad destiladora Kjeltec 2100 (Foss, Suecia). Por último se realiza una valoración con HCl 0,1 N. La conversión a porcentaje de proteína bruta se realizó mediante la siguiente fórmula: % Proteínas = (V – P) x N x P m x F / M Donde: V es el volumen de HCl usado en la valoración en ml; P es la media de la valoración de los patrones en ml; N es la normalidad de HCl; P m es el peso molecular del nitrógeno que es 14,007; F es el factor de conversión empírico que tiene un valor de 6,25; M es el peso de la muestra en mg. 2.4.4. Lípidos El contenido en lípidos totales de las muestras fue determinado mediante el método descrito por Folch et al . (1957), haciendo uso de una mezcla de cloroformo-metanol (2:1
– Material y Métodos Generales – 43 volumen/volumen) conteniendo 0,01 % de BHT. Por decantación se separan las fases acuosa y orgánica, el contenido de lípidos totales de la muestra, se determina después de la evaporación completa del cloroformo con una corriente de nitrógeno. Los lípidos se guardaron a -80 ºC, en viales etiquetados, disueltos en cloroformo y en atmosfera de nitrógeno para evitar la oxidación. Para la determinación de ácidos grasos, los lípidos obtenidos por el método de Folch et al . (1957) se transesterificaron con ácido sulfúrico al 1% en metanol (Christie, 1982), y se añadió tolueno para favorecer la disolución de los lípidos neutros, diluyendo los ésteres metílicos de los ácidos grasos (FAMEs) en hexano a una concentración de 20 mg por ml de hexano. Los ésteres metílicos fueron identificados y cuantificados mediante cromatografía de gases, con el aparato Shimadzu GC-14A (Kyoto, Japón). 2.5. ANÁLISIS HISTOLÓGICOS Los análisis histológicos se realizaron en el laboratorio del IUSA. En los experimentos I y II del presente trabajo, las muestras de gónadas fueron cuidadosamente diseccionadas y fijadas en formol al 10 % por al menos 24 horas. Posteriormente las muestras fueron introducidas en casetes, que permitieron el paso de los reactivos sin la pérdida de muestra y fueron lavadas durante 30 minutos en agua. A continuación tuvo lugar un proceso con una duración de 24 horas, donde las muestras son deshidratas en alcohol ascendente, transferidas a xilol e infiltradas en parafina. Todo ello en un procesador de tejidos Histokinette 2000 (Leica, Nussloch, Alemania). Los bloques fueron conservados en el congelador, para ser tallados primeramente a 15 – 20 µm, enfriados nuevamente y posteriormente se realizaron cortes a 4 – 5 µm en un micrótomo de rotación Jung Autocut 2055 (Leica, Nussloch, Alemania). Los cortes fueron recogidos en un baño de flotación con agua destilada a 45 ºC. Los portaobjetos con los cortes de la muestra fueron secados en estufa (1hora a 60 ºC). Las preparaciones fueron posteriormente inmersas en xilol e hidratadas en alcohol descendiente hasta llegar a agua, según el siguiente protocolo: Xilol 15 minutos Alcohol absoluto 5 minutos Alcohol absoluto 5 minutos Alcohol 96 % 5 minutos Alcohol 90 % 5 minutos Alcohol 70 % 5 minutos Agua 5 minutos Una vez terminado el proceso anterior se realizó la tinción Hematoxilina – Eosina (Socorro, 2006). Se introdujeron los portas con las muestras en cubetas que contenían: Hematoxilina de Harris 15 – 20 minutos Alcohol ácido 3 baños cortos Lavado en agua Agua amoniacal 20 segundos Agua continua 15 minutos Eosina de Puttis 3 – 4 minutos Lavado en agua Tras la tinción se procedió a la deshidratación y clarificado de las preparaciones, de acuerdo con el siguiente protocolo: Alcohol 96 % 5 minutos Alcohol 100 % 10 minutos

52 – Experimento I – 4.1.1. INTRODUCTION The reproductive seasonality of several abalone species from various regions in the world has been studied (Boolootian et al ., 1962; Tomita, 1967; Newman, 1967; Webber and Giese, 1969; Poore, 1970; Young and DeMartini, 1970; Hayashi, 1980a; Tutschulte and Connell, 1981; Capinpin et al ., 1998; Fukuzawa et al ., 2007). Most of the studies mentioned here have contributed to the development and improvement of the artificial propagation techniques for a number of abalone species, as well as to the growth of abalone aquaculture around the world (e.g. Haliotis discus hannai and Haliotis rufescens ). Ormer Haliotis tuberculata tuberculata Linnaeus (1758) is currently the only commercial abalone species in Europe. It was considered as a potential candidate for aquaculture development in the 1970s in France, and its commercial fishing and culture interest significantly increased all through Europe during the early 1990s as a result of much easier international trade (rapid transport and market globalization) and an increase in its value in Asian markets (Huchette and Clavier, 2004). New practices for the production of juveniles have recently emerged and enabled a rapid expansion of this industry (Daume et al ., 2004; Roberts et al ., 2004). Ireland and the Channel Islands (Huchette and Clavier, 2004), Scotland (Kelly and Owen 2002), and France are currently the only established producing countries. Spain has also carried out some experimental culture trials. A subspecies of ormer, Haliotis tuberculata coccinea Reeve (1846), lies in the Macaronesian region (Geiger, 2000). In the Canary Islands, it has a patchy distribution on exposed and semiexposed areas from low intertidal to subtidal depths, shallower than 15 m, with an affinity for specific habitats (Nuñez et al ., 1994; Espino and Herrera, 2002). Coralline red algae are generally considered to provide an optimum substratum for larval settlement and metamorphosis (Morse and Morse, 1984). In fact, Espino and Herrera (2002) found high densities of Haliotis tuberculata coccinea juveniles and adults on rock surfaces encrusted with red coralline algae. Furthermore, this habitat is closely linked to a regular supply of drifting algae carried by the currents. The amount of such drifting algae determines the abalone population density where its habitat is suitable (Clavier and Chardy, 1989). This subspecies have been overexploited because of its high social and economic value. After Nuñez et al . (1994) and Espino and Herrera (2002), it has been classified as an endangered species in the Catalogue of Threatened Species of the Canary Islands (BOC-2001/097). These studies have assessed some populations around the islands and concluded that this subspecies cannot withstand the fishing pressure in the Canaries. Recently, the government of the Canary
– Experimento I – 53 Islands has shown interest in restocking this natural ecosystem and considered Haliotis tuberculata coccinea as a new species for the diversification of local commercial aquaculture. Its artificial spawning induction was studied by Peña (1986) and more recently, to improve knowledge about its culture, embryology, complete larval development, and juvenile nutrition requirements have also been described (Viera et al ., 2005; Courtois de Vicose et al ., 2007). This study aims to provide more accurate biological information on sexuality, reproduction, fecundity, and growth of this species. This knowledge, when applied to hatcheryrelated activities, may improve the culture of this species and is essential to studying restocking in natural ecosystems. 4.1.2. MATERIALS AND METHODS Sampling Monthly samples of 20 individuals were collected during the day by diving between June 2005 and May 2007 at a depth shallower than 10 m at El Castillo del Romeral Bay, Gran Canaria Island (Figure 15). Total shell length to the nearest 0.5 mm and total body weight to the nearest 0.1 g were taken for each specimen. Conical appendages, consisting of the digestive gland and the surrounding gonad, were carefully removed and weighted to the nearest 0.001 g. 54 – Experimento I – Ovary morphology The basal, middle and apical regions (Young and DeMartini, 1970) of the conical appendage were fixed in 10% buffered formaldehyde for histological study. After 24-48 hours, they were dehydrated, embedded in paraffin wax, sectioned into cross-sections (4 - 5 µm thick), and stained with Harris´hematoxylin-eosin stain. Three regions of conical appendages were observed microscopically to determinate the oogenetic processes in female gonads. Oocyte stages were assigned according to Apisawetakan et al . (2001). Sexuality and spawning season Sex and maturity stages (MS) of gonad were assigned according to the classification described by Capinpin et al . (1998) (Table 1). Monthly changes in the three following variables were analyzed to determine the spawning season (Hanh, 1989): 1) Gonad bulk index (GBI) is the percentage of gonad tissue at the base of the conical appendage. 2) GBI= (Area of the gonad in the section / Total area of the section) × 100. 3) Percentage frequency of the maturity stages (EMS). 4) Oocyte diameter (OD): Diameters of 100 oocytes were measured using Image Proplus software (ver. 5.0.1.). Measurements were made only of oocytes sectioned through the nucleus. Fecundity The pattern of annual fecundity was assessed by oocyte size frequency distribution. Ripe females ranging between 37 and 47 mm in shell length in the ripe stage were selected for analysis of oocyte size frequency distribution in November and March, representing gonad development early and late during the spawning season. Because the mean size ( t -test, P>0.05) was not significantly different between both samples, one frequency distribution was obtained for each month. The gravimetric method was used to estimate batch fecundity (modified from Hayashi, 1980a). Conical appendages of 12 ripe stages females, collected in October, were isolated from gonads and weighed to the nearest 0.001 mg. Subsamples from 3 different locations (basal, middle and apical regions) of each conical appendage were weighed, and oocytes loosened from trabeculae were counted. Fecundity was defined as the number of oocytes in stages 4 and 5. To calculate the percentage of oocytes in each region, gonad was fixed in 10% buffered formaldehyde for histological study. Oogonia cells are growing from the trabeculae and form clusters on trabecular walls. The gonad shows vacant spaces. 3: Maturing A layer of spermatids or spermatozoa is present in the testis. Vitellogenesis is initiated at this stage. Early maturing ovary is characterized by newly stalked oocytes measuring about 25µm. A late-maturing ovary has oocytes larger than 50 µm that extend from the trabeculae toward the digestive gland, with each attached by a stalk and is shapep like a teardrop. 4: Ripe The testis is densely packed with spermatozoa. A few early gametogenic stages may be present, but are restricted around the tubules. The ovary has oocytes larger than 150 µm. Mature oocytes free from the trabeculae have a mean diameter of 150 µm. A slight proliferation of oocytes less than 25 µm is common, especially near the peripheral wall of the gonad. 5: Partly spawned The testis has obvious spaces around the tubules where spermatozoa have vacated. Other areas appear as ripe. The ovary contains reduced densities of mature oocytes relative to ripe gonads. The gonad lumen is partially collapsed with folded trabeculae. A moderate number of ripe oocytes still present in some areas is densely packed. 6: Spent A spent gonad is characterized by lack of ripe gametes and slight gametogenic activity. Macroscopically, the gonad is greatly reduced. The gonad lumen is collapsed, with folded trabeculae or tubules. A spent ovary may have few residual oocytes. The females were divided into 3 classes according to their shell length (35-44, 45-54 and 55-64 mm) and the batch fecundity results were submitted to a statistical analysis applying a 1-
56 – Experimento I – way ANOVA followed by a Tukey test for multiple comparisons of means, at a 5% significance level (P<0.05). Population structure Normality of length data was previously determined by means of the KolmogorovSmirnov test for comparing the mean sizes of the different study groups of abalone. The same statistical test was used for comparing different distributions of length data. The Student´s t -test was applied as a non-parametric test for comparing 2 means (Zar, 1996). Carapace length-perwet weight relationships were calculated on each sex or group with respect to the whole period studied. The overall ratio (males to females) and the sex ratios by size intervals (2 mm) were determined considering the number of males and females. Sex ratios were tested statistically for significant deviations from the expected 1:1 ratio with a chi-square test (P>0.05). Growth Length frequency data were analyzed for estimating growth in each sex by means of Pauly´s method (Pauly, 1983). Length frequency was represented quarterly during 2 years to simulate the time sequence. The FAO-ICLARM Stock Assessment Tools (FISAT) program (version 1.2) was used for data processing and calculating the von Bertalanffy growth parameters. Bhattacharya’s method was used for decomposition of mixtures of normal distribution. The estimation of L ∞ was obtained by means of Gulland and Holts´ method, whereas the k value was determined using the routine ELEFAN I (Gayanilo and Pauly, 1997). For the sake of comparison, growth performance of both sexes Munro´s phi prime (φ´= log k + 2 log L ∞ ) was assessed according to Sparre et al . (1989). 4.1.3. RESULTS Ovary morphology The histological study revealed that the conical appendage included the conical digestive gland surrounded by the ovary, which was classified into 3 regions according to the relative importance of both tissues. In the basal region the digestive gland occupied most of the cross-section, in the medium region the ovary filled most of the cross-section, and in the apical region only ovarian tissue was found. The ovary was enclosed within 2 walls that separated the ovary from the mantle and the digestive gland. The connective tissue extended perpendicularly into the gonads to generate the trabeculae, dividing the gonad into compartments around the lumens, where oocytes were developing. The first stages of the sexual cells appeared attached
– Experimento I – 57 to the trabeculae, whereas mature oocytes were formed near the lumen. The trabeculae decreased in thickness as the female gonad ripened. Two phases and 6 stages of female sexual cells were recognized (Figure 16 A-E): a) Previtellogenic phase: Oogonia (Og): round cells with 10.66 ± 2.7 µm diameter, closely attached to the trabeculae, showing a highly basophilic nucleous surrounded by homogeneous basophilic cytoplasm (Figure 16A). Stage 1 Oocyte (Oc 1 ): oval shaped with a maximum diameter of 26.47 ± 6.44 µm and a minimum diameter of 15.48 ± 4.05 µm, strongly attached to the trabeculae, round nucleus containing a diffuse nuclear envelope, follicular cells surrounding (Figure 16B). Stage 2 Oocyte (Oc 2 ): elongated shaped with a maximum diameter of 60.51 ± 17.32 µm and a minimum diameter of 33.15 ± 13.04 µm, attached to the trabeculae only in the basal region, oval nucleous, first lipid droplets appear (Figure 16B, C). b) Vitellogenic phase: Stage 3 Oocyte (Oc 3 ): pear shaped with a maximum diameter of 110.30 ± 27.63 µm and a minimum diameter of 59.64 ± 21.02 µm, increasing number of lipid droplets, appearance of the first yolk eosinophilic vacuoles (Figure 16C). Stage 4 Oocyte (Oc 4 )): polygonal shaped with a maximum diameter of 162.25 ± 28.76 µm and a minimum diameter of 103.2 ± 23.49 µm, attached to the trabeculae by a small contact surface, cytoplasm contained yolk vacuoles and numerous lipid droplets, secretory granules with basophilic propriety migrated toward the plasma membrane, follicular cells located around the narrowest part of the cell, appearance of a thin eosinophilic layer of jelly coat on the outer surface of the cell membrane (Figure 16D). Stage 5 Oocyte (Oc 5 ): polygonal shaped with a maximum diameter of 171.77 ± 28.75 µm and a minimum diameter of 114.91 ± 26.51 µm, oocyte is released into the lumen of the ovary, follicular cells absent, yolk vacuoles aggregated (Figure 16E). Sexuality and spawning season Of the total number of abalone examined, 207 were females and 204 males. Assignation of sex macroscopically was easy in the most cases, except in the smaller or spent gonads.

Partly spawned and spent-stage individuals appeared yearlong excluding January, probably because of the few number of samples (N=11 over two years). 68 – Experimento II – 4.2.1. INTRODUCTION The abalone present in the Canary Islands is Haliotis tuberculata coccinea Reeve (1846). It is a small subtropical species with a maximum shell length of 8 cm and its spawning season occurs all over the year (Bilbao et al ., 2010a) as well as other tropical species (e.g. Haliotis asinina and Haliotis diversicolor ) (Takashima, 1978; Capinpin et al ., 1998). The Government of the Canary Islands has recently declared Haliotis tuberculata coccinea as a new species for the diversification of local commercial aquaculture. However, its culture should be developed from broodstock kept in captivity and separated from natural populations, since Haliotis tuberculata coccinea is legally protected and its capture is prohibited in the natural environment (cf. Official Gazette of the Canary Islands BOC-2001/097). The broodstock conditioning is mainly base on a proper diet in order to reach an optimal development of the gonad. Thus, it is essential to know the nutritional requirements of the species to be conditioned. Although the nutritional studies carried out on broodstock abalone are scarce, it is known that the amount of proteins in the diet plays an important role in gonadal morphology and the fecundity. Jackson et al . (2001) reported that the amount of macrophages in Haliotis asinina mature gonads increases as a result of stress or malnutrition of animals fed on artificial diets with both low protein content and relative high lipid content. Likewise, Bautista-Teruel et al . (2001) observed that, to some extent, Haliotis asinina fecundity increased due to the higher amount of proteins supplied in the diet. Specific lipids such as polyunsaturated fatty acids (PUFA) (e.g. Arachidonic acid, AA, 20:4n-6 and Ecosapentaenoic, EPA, 20:5n-3) play an important role in the gonadal development of abalone (Nelson et al ., 2002). It is also known that feeding Haliotis laevigata on an altered diet over a 3-month period is sufficient to alter the PUFA profile of the eggs (Daume and Ryan, 2004) and Graham et al . (2006) suggested that depending on spawning frequency, broodstock diet can influence the cytoplasm diameter and jelly coat thickness. Feeding abalone with seaweeds of the genera Gracilaria is appropriate in terms of growth and survival rates (Reyes and Fermin, 2003; Naidoo et al ., 2006; Qi et al ., 2010). Furthermore, species such as Gracilaria flagelliformis have a high content of arachidonic acid, playing an essential role in the mollusk reproduction (Daume and Ryan, 2004). The species Gracilaria cornea is found in the Canary Islands and it is a good feed for Haliotis tuberculata coccinea when it fertilized in an effluent from fish cultures (Viera et al ., 2005). Since the genera of Gracilaria captures the nutrients present in the water, with one of the highest nutrient intake capacity known, the seaweed protein content can be manipulated under specific culture
– Experimento II – 69 conditions (Neori et al ., 2004). This is interesting for two reasons; firstly, because Gracilaria cornea becomes an ideal species for an experimental study on the effects of the nutritional value of macroalgae on abalone reproduction. And secondly, because it provides a commercial approach, since wild algae are not commercial valuable in the Canary Islands and, therefore, they are not harvested. Thus, the availability of this natural resource is scarce and insufficient to sustain the commercial production of abalone. Hence, the culture of seaweed for feeding broodstock is an alternative to wild food. This study aims to understand the effect of protein content variations of the macroalga Gracilaria cornea fertilized by pulses on feeding Haliotis tuberculata coccinea and to improve our understanding about the nutritional requirements in broodstock conditioning. The experiment specifically compares the influence of the protein content variation of Gracilaria cornea on the growth, the consumption, the viability of spawning and both the fatty acid composition and the morphology of the gonad in Haliotis tuberculata coccinea . 4.2.2. MATERIALS AND METHODS Experimental diets The macroalga used in this study was Gracilaria cornea (Agardh, 1852). Six circular white plastic tanks with a capacity of 90 l were used for the cultivation of macroalgae. An initial stock of 1000 g of seaweed was added in each tank. The tanks were located in a greenhouse, where maximum irradiance was close to 1600 µmol photons / m 2 per second. The water exchange rate was 4 volumes per day and aeration was supplied by a bottom-central linear pipeline. To produce Gracilaria cornea with different protein contents, 33.3 mg / l day (High-N), 11.1 mg / l day (Medium-N) and 1.1 mg / l day (Low-N) NH 4 NO 3 was supplied to two culture tanks per protein treatment for a minimum of 15 days, to ensure stable protein content. 3.3, 1.1 and 0.1 mg / l day NaH 2 PO 4 was then added to each culture, respectively. These nutrients were added by pulse-fertilizing three times a week for 6 to 8 hours. Every other week, the macroalgae were removed from the tanks and the excess water was removed by blotting them, afterwards, the macroalgae wet weighed to assess algal biomass and restocked at the original density. The excess macroalgae were harvested for use in the experiment. The epiphytes were regularly removed from the plants. Experimental animals and culturing condition Abalones ( Haliotis tuberculata coccinea ) were produced within the experimental hatchery production unit of ICCM. A total of 90 females aged 3 years (10 abalones / replicate)
70 – Experimento II – with an average shell length a nd weight of were selected for the trial. Females were acclimatized to the diets and new cultured conditions for 3 weeks and were induced to spawn. Three diet types were tested: high protein content medium protein content G racilaria Gracilaria cornea (Low-N G. cornea ) body weight and 0.1 mm in shell length and assigned to an experiment 60 l ). Abalones were distributed among replicates so that there were no significant differences in shell length and body weight. – Experimento II – 71 The experiment was carried out between November 2008 and August 2009. The experimental tanks were cleaned and fecal material removed once per week. Same day, the uneaten macroalgae were removed and replaced with fresh algae. Growth and algal consumption To determine diet intake (D), the collected algae to be the fed to the abalone and the remaining algae in the tanks were weighted (± 0.01 g) at the end of the week. Prior to weighing of algae the excess water was removed by blotting them with tissue paper. The weight of unconsumed food was deducted from the total weekly ration and is expressed in grams. Reproductive performance The spawning acts in Haliotis tuberculata coccinea are related to spring tides around new or full moons throughout the year in natural populations (Bilbao et al ., 2010a). Under culture conditions, sometimes some of conditioned-animals spontaneously spawn around the new or full moons, however these gametes are not viable in the activities of an abalone hatchery because they are unpredictable. Thus, we take into consideration the viability of spawning defined as the number of eggs released per gram of abalone after the spawning induction. Females were induced to spawn on March and September 2009. Females from each replicate were placed in 9 plastic tanks and spawning was conducted using a flow-through system of UV irradiated seawater over 3 days around the full moon. The spawned eggs were immediately separated from the animals with a siphon and were concentrated into a known volume of clean seawater. The water containing the eggs was gently stirred to ensure a homogenous distribution of eggs through water column, and three subsamples were taken for estimating the
72 – Experimento II – amount of eggs released. The viability of spawning was considered as the total number of eggs released and it was divided by the weight of females that responded to spawning induction. We also measured the maximum diameter and the maximum diameter of the cytoplasm in 50 eggs using a profile projector (Mitutoyo PJ – 3000A, Kanagawa, Japan). At the end of the experiment conical appendages of at least 2 mature females of each replicate were preserved in 10 % formaldehyde for histological study. The choice was based according to the visual gonad index proposed by Uki and Kikuchi (1984) and we only take into account the most mature females (2-avanced and 3 stages). The half part of conical appendage was carefully removed, and they were dehydrated, embedded in paraffin wax, sectioned in cross-sections (4 – 5 µm thick) and stained with Harris´ hematoxylin eosin. The slices were microscopically observed in order to assess the stage of maturity gonad according to the classification proposed by Capinpin et al . (1998) and we only take into account the ripe-stage gonads. Them, we evaluated the morphology of ripe-stage gonads. Biochemical Analysis At the end of the experiment, we made dissection of gonad, digestive gland and muscle of abalones since these body components are likely sites of energy storage for subsequent use in gonad maturation. The samples collected were cleaned and frozen in -80 ºC. The biochemical analysis of each diet type was run during the conditioning period. The samples collected were cleaned, washed with freshwater to remove salts and epizoos, frozen in - 80 C freezer and lyophilized at least 24 hours. The biochemical analysis was carried out as following: The dry matter was determined by drying at 110 ºC for 24 hours. Ash content was determined by incinerating samples at 600 ºC for 24 hours. Total protein was calculated from total Kjeldahl nitrogen according to AOAC (2005) standard methods. Total lipids were extracted with chloroform-methanol (2:1) mixture as described by Folch et al . (1957). Fatty acids in the lipid extracts were transesterified to methyl esters (FAMEs) with 1% sulphuric acid: methanol complex (Christie, 1982). FAMEs samples were extracted into hexane and stored at -80 ºC. Fatty acids were analyzed in a Thermo Finnigan-GC Focus gas chromatography equipped with a flame ionization detector (260 ºC). FAMEs were separated with capillary column using helium as the carrier gas under the conditions described by Izquierdo et al . (1989).
– Experimento II – 73 Statistical analysis The results were submitted to a statistical analysis applying a 1-way ANOVA with diet as the fixed factor. Data was tested for normality prior to analysis using the Kolmogorov–Smirnov test. Bartlett's test was used to test for homogeneity of variances (Zar, 1996) and Tukey's multiple comparison test was used in pair-wise comparisons among diets when significant differences (P<0.05) were determined. 4.2.3. RESULTS Algal nutritional composition The nutritional compositions of the three Gracilaria cornea diets are showed in Table 2. The protein content exhibited statistical differences among three diet types: the highest value was observed in High-N G. cornea (29.88 ± 2.38 %), followed by Medium-N G. cornea (25.01 ± 1.07 %) and Low-N G. cornea (10.37 ± 2.04 %). There were also significant differences in the carbon content of the three diet types: the carbon content of Low-N G. cornea exhibited a 50 % higher value than observed in High-N G. cornea . The total lipid content did not vary among the diets. The ash content varied inversely to carbohydrate content: the lowest value was observed in Low-N G. cornea and it was significantly lower than in the other diets. 74 – Experimento II – Survival and growth performance The survival rate was 90.0 ± 10.0, 83.3 ± 15.3 and 83.3 ± 15.3 % in the Low-N, Medium-N and High-N treatments respectively. Hence, final shell length was significantly highest in animals fed on Medium-N G. cornea and High-N G. cornea and lowest in those fed on Low-N G. cornea . Accordingly, DSIL followed the same pattern, this parameter being highest in animals fed on Medium-N G. cornea and High-N G. cornea and lowest in those fed on Low-N G. cornea (Table 3). However, the final body weight of abalones fed on High-N G. cornea was similar than those fed on Medium-N G. cornea and higher than those fed on Low-N G. cornea . In this way, SGR and WG followed this same pattern (Table 4). – Experimento II – 75 The utilization of feeding The feed utilization of Haliotis tuberculata coccinea is showed in Table 5. MTFI showed statistical differences among treatments. The highest value was observed in the High-N treatment and it was similar than those in the Medium-N treatment and higher than the value observed in the Low-N treatment. Although, FR of abalones fed on High-N G. cornea exhibited the highest value, we did not observe any statistical differences among treatments. On the contrary, PER was significantly higher in abalones fed on Low-N G. cornea than those fed on Medium-N and High-N G. cornea . The proximate composition of abalone The proximate composition of abalones fed on Low-N, Medium-N and High-N G. cornea is showed in Table 6. The protein content of abalone viscera fed on High-N G. cornea was significantly higher than observed in the others treatments. The carbohydrate content of viscera varied inversely to protein content of the diet and the ash content did not show statistical differences among the three treatments. The lipid content of viscera did not show a clear relation to protein content of diets. The abalone muscle was the most affected organ to relation the protein content of the diets. The protein content of the muscle exhibited a positive relation to protein content of the diet. The highest value was observed in the animal muscle fed on High-N G. cornea followed by those fed on Medium-N and Low-N G. cornea . On the contrary, the carbohydrate content of the

The contents of lipid, protein, ash and carbohydrate of abalone gonads did not show significant differences among the three treatments. The protein was the principal component of gonad, followed by lipid, carbohydrate and ash. Fatty acid composition of diets and gonads High-N G. cornea , Medium-N G. cornea and Low-N G. cornea had a similar composition of fatty acid (FA) (Table 7). Palmitic acid (16:0) was the major FA, followed by Arachidonic acid [AA 20:4(n-6)]. The FA composition of gonads is showed in the Table 8. Palmitic acid (16:0) was the major FA. The second most abundant FAs were Vaccenic acid [18:1 (n-7)], Arachidonic acid [AA, 20:4(n-6)], Oleic acid [18:1 (n-9)] and Myristic acid (14:0).
– Experimento II – 83 treatments were similar to the results previously reported on both wild and cultivated abalone fed on formulated diets and macroalgae (Floreto et al ., 1996; Chiou et al ., 2001; Nelson et al ., 2002; Litaay and DeSilva, 2003; Grubert et al ., 2004). With the exception of EPA, the amount of different fatty acid levels found in gonads was within the range obtained by Nelson et al . (2002) throughout one year in Haliotis fulgens adults fed on different macroalgae: palmitic acid (16:00) was the dominant saturated fatty acid and vaccenic acid (18:1n-7) was reported as the dominant monounsaturated fatty acid (35-37 and 18-20 % respectively). Likewise, the percentage of EPA, total n-3 and n-6 fatty acids in the gonads of Haliotis tuberculata coccinea were similar (1.4, 6 and 20-25 % respectively) to those obtained by Grubert et al . (2004) in mature gonad of Haliotis rubra and Haliotis laevigata species fed on a formulated diet (1.9, 4.4-6.2 and 23 % respectively). However, in the present study, the percentage of AA (6-8 %) was higher than the value obtained in Haliotis rubra and Haliotis laevigata (0.7 and 0.9, respectively). Daume and Ryan (2004) compared the eggs of abalones fed on formulated diets and macroalgae and proved that the PUFA composition of the broodstock diet influenced the PUFA profile of the eggs. It must be considered that, in the three studies (the present study; Nelson et al ., 2002; Grubert et al ., 2004), the diets fed to broodstock (formulated diet and diet based on different macroalgae species) had different fatty acid profiles and, thus, the differences of the PUFA composition of the Haliotis rubra , Haliotis laevigata and Haliotis tuberculata coccinea gonads may be due to the different PUFA profiles of the diets. Regarding the gamete development and the broodstock nutrition of abalone species, significant attention has been paid long-chain PUFAs, such as AA, EPA and DHA, since abalones are unable to synthesis these PUFA de novo (Uki et al ., 1986; Mai et al ., 1996). Abalone require EPA in the diet rather than DHA (Uki et al ., 1986), and when the dietary EPA is low (e.g. < 1% total FA in this study) they are able to retroconvert DHA into the EPA form (Dunstan et al ., 1996).This may explain the amount of EPA observed in the gonads in comparison to those found in the three diets. Although high levels of AA may not required for muscle growth, it may be required for ovogenesis and embryogenesis (Daume and Ryan, 2004). The AA is related to the cell transmission functions and is the precursor of the eicosanoids, particularly prostaglandins (Bell et al ., 1994; Nelson et al ., 2002; Bell and Sargent, 2003,), which are involved in several reproductive processes (Moore, 1985; Nelson et al ., 2002; Laas and Vosloo, 2010), including the production of steroid hormones and the gonadal development such as ovulation. The lipid of the eggs is major parts of the energetic during early ontogeny and larval metamorphosis (Litaay et al ., 2001; Daume and Ryan, 2004). Moreover, Jensen et al . (1990) showed that the metamorphosis of Haliotis rufescens larvae was influenced by free fatty acids, especially AA. As
84 – Experimento II – we discussed above, the gonad and the egg PUFA profile depends on the broodstock diet, especially the AA, considering that abalones are not able to synthesize AA from n-6 series with lower FAs (Floreto et al ., 1996). Considering that Gracilaria cornea had a high amount of AA, it is an appropriate food for abalone broodstock. Moreover, when the highest protein diets were used, the n - 6 PUFA and specifically the AA slightly increased (2 %) in the gonads. Cook et al . (2007) showed that a high protein content in the diet promotes the accumulation of AA in the tissue of the sea urchin gonad ( Psammechinus miliaris ) during maturation. However, this behavior has not been clearly observed in the present study because increasing the amount of protein in Gracilaria cornea also increased the amount of AA (from 22 to 27 %). Thus, the increase of the AA amount observed in the gonads of the three treatments may be due to a different amount of AA in the diet rather than to a protein supplement to the diet. The relationship between the dietary protein content and the fecundity has been little considered in mollusk aquaculture. Farías and Uriarte (2001) showed that a protein supplement to a high-protein diet fed to scallop ( Arcopecten purpuratus ) broodstock was mainly used for the production of a larger amount of eggs and not for the production of larger eggs. Likewise, Bautista-Teruel et al . (2001) suggested that a higher amount of macronutrients in the artificial diet used in their study, such as proteins and lipids, improved the reproductive performance of Haliotis asinina (i.e. fecundity and egg hatching rates). Although the real fecundity of Haliotis tuberculata coccinea has not been considered in the present study, as we did not count the spontaneous spawning, the results showed the highest values of the viability of spawning in females fed on Medium-N and High-N G. cornea . The broodstock can respond to nutritional stress conditions by reabsorbing the mature gametes. Martinez et al . (1992) concluded that a reduction in the gonad index of a hatcheryreared scallop ( Argopecten purpuratus ) was caused by the reabsorption of mature gametes because the broodstock was under nutritional stress. Likewise, Jackson et al . (2001) showed that juvenile Haliotis asinina fed on two artificial diets (with low protein content and a relatively high amount of lipids) grew less and mature gonads contained a large amount of macrophage populations reabsorbing mature eggs as a result of malnutrition or stress in the animals. In the present study, the morphology of the ripe-stage gonads of abalones fed on Medium-N and High-N treatments were similar to those found in the ripe-stage gonads of wild Haliotis tuberculata coccinea (Bilbao et al ., 2010a). However, the ripe gonads of females fed on Low-N G. cornea showed a higher amount of macrophage populations. In fact, with the Low-N treatment, the eggs expelled after the spawning induction were significantly smaller than those found in the other two treatments. Graham et al . (2006) showed that the eggs spawned by Haliotis laevigata broodstock fed on different diets (red seaweed and formulated diets) had different
– Experimento II – 85 sizes, suggesting that the lack of some macronutrients or micronutrients in the diets affected the egg size. In the present study, the protein content was the major difference among the diets. Hence, we used a single macroalga species as food for the broodstock. It must be taken into account that the larger eggs are not necessarily obtained from the highest AA-content diets (Graham et al ., 2006). Thus, the large amount of macrophages in the ripe gonads receiving the Low-N treatment may result in a smaller size of the eggs expelled after the spawning induction, since the macrophages reabsorbed the most mature oocytes. This may due to the malnutrition of the animals, which may arise from an insufficient protein content in the diet. Further research in this field with more appropriate experimental designs (i.e., increasing the sample size and increasing the frequency of gonad sampling) may provide a better understanding of the impact of the dietary protein content on the gonad morphology. In conclusion, the use of Gracilaria cornea enriched with 33.3 mg/ l day NH 4 NO 3 resulted in the highest growth rate, in an increased of the viability of spawning and in a higher AA content mature gonad. Thus, the methodology described in this study would be a simple and inexpensive method to obtain a better conditioning of broodstock abalone, especially in the Canary Islands, where there is not a stable market for harvesting seaweed and their geographical isolation makes the purchase of artificial feed more expensive.

88 – Experimento III – 4.3.1. INTRODUCTION In abalone aquaculture, there are many instances requiring some sedation to facilitate handling animals without injuring or stressing them excessively. Abalone is well adapted to hold tightly onto the substratum, and mechanical assistance is necessary to remove them. Dislodgement by mechanical means often results in injury or death because their slow healing rate, absence of a blood coagulation mechanism and stress (Armstrong et al., 1971; Genade et al., 1988). Chemical removal, by anaesthetizing the animals provides a safer alternative to mechanical removal (White et al., 1996). On other hand, in pearl culture of abalone, animals require some degree of relaxation to facilitate nucleus implantation. When choosing an anaesthetic, a number of considerations are important, such as efficacy, cost, availability and ease of use, as well as toxicity to animal, humans, and the environment (Soto and Burhanuddin, 1995). Aquilina and Roberts (2000) consider that the anaesthetic used for relaxation to allow for the insertion of pearl nuclei in abalone has to have the following characteristics: rapid onset of relaxation, full extension and softening of the muscles for at least 10 minutes, and rapid recovery. Several anaesthetics have been suggested for abalone aquaculture (Sagara and Ninomiya, 1970; Prince and Ford, 1985; McShane and Smith, 1988; Hanh, 1989; Tong et. al, 1992; White et al., 1996; Aquilina and Roberts, 2000; Sharma et al., 2003); however, most of them are chemical products such as magnesium sulphate, 2-phenoxyethanol, pentobarbitone, and benzocaine. Cloves are the dried flower buds of Eugenia caryophylata , and its active ingredient is eugenol. Aroma chemicals present in natural leaves and flowers have been widely used in aroma therapy since ancient times, suggesting that they have some beneficial health effects. Clove oil has been traditionally used by humans as an oral analgesic, and it has been investigated as sources of anti-microbial agents against oral bacteria (Cai and Wu, 1996), it has demonstrated its antifungal (Martini et al., 1996) and antioxidant activity in human food comparable with α-tocopherol (vitamin E) (Kwang-Geun and Shibamoto, 2001), and it is effectively used for asthma and various allergic disorders by oral administration (Kim et al., 1998). In aquaculture, clove oil is considered superior to a number of anaesthetics, such as benzocaine and MS-222 (Munday and Wilson, 1997; Keene et al., 1998). Because clove oil is a natural product and is less expensive than other anaesthetics used in fish, it has received considerable attention (Endo et al., 1972; Soto and Burhanuddin, 1995; Cho and Heath, 2000; Wagner et al. , 2002; Hoskonen and Pirhonen, 2004; Mylonas et al. , 2005). However, in mollusc
– Experimento III – 89 aquaculture, its anaesthetic effect has been demonstrated only in the common octopus, Octopus minor (Seol et al., 2007) and in pearl oyster, Pinctada albina (Norton et al. , 1996). The objective of the present study was to evaluate the use of clove oil as natural anaesthetic in Haliotis tuberculata coccinea , comparing its efficacy to the commonly used 2phenoxyethanol. 4.3.2. MATERIALS AND METHODS The experiment was undertaken in July 2008, using adults of Haliotis tuberculata coccinea produced at the Instituto Canario de Ciencias Marinas, Gran Canaria Island, Spain. The anaesthetics chosen for evaluation were clove oil at 4 different concentrations and 2phenoxyethanol at 2 ml / l (Table 10). The trial was conducted in transparent plastic aquaria holding 2 l seawater equipped with a shelter. To aid dissolution, the clove oil dose was shaken at 50% in absolute ethanol, and 2-phenoxyethanol was dispensed directly. Thirty animals per treatment (3 per aquaria), between 42 and 80 mm in shell length, previously tagged, were firmly attached to the shelter. The temperature was maintained as natural (22.3 ºC) and, PH was measured before and after adding the anesthetics. The animals were kept in the solution for 30 minutes. The relaxation time was defined as the length of time between the introduction of the animals into the aquaria and releasing from the shelter (Figure 29). At the conclusion of

Omitting these animals for statistical analyses, the relaxation time of the rest animals treated with clove oil decreased with an increase in concentration (855 ± 287 seconds at 0.1 ml/l, 786 ± 248 seconds at 0.3 ml/l, 536 ± 208 seconds at 0.5 ml/l and 472 ± 139 seconds at 0.7 ml/l). Statistical analysis showed significant differences in the mean of the high concentrations of clove oil (0.5 ml/l and 0.7 ml/l) compared with low concentrations (0.1 ml/l and 0.3 ml/l; Figure 30). The recovery time did not show statistical differences among different concentrations (181 ± 89 seconds at 0.1 ml/l, 143 ± 133 seconds at 0.3 ml/l, 121 ± 90 seconds at 0.5 ml/l, and 186 ± 137 seconds at 0.7 ml/l; Figure 31). The relaxation time observed with 2-phenoxyethanol was less (29 9± 56 seconds), and less dispersal was observed for clove oil at the 0.5 ml/l concentration (536 ± 208 seconds; Figure 30 and Table 11). However, the recovery time was more (433 ± 215 seconds at 2 ml/l of 2phenoxyethanol and 121 ± 90 seconds at 0.5 ml/l of clove oil), with more dispersal (Figure 31 and Table 11). Qualitatively, the effect of each chemical as a muscle relaxant was similar, producing a contracted muscle with a little exudation of mucus.

RESULTS Shell-boring polychaetes The shell-boring polychaete species found in the shells of abalone were identified as Polydora hoplura Claparède (1870). These large worms were 6.50 mm in length and 0.5-0.6 mm in width, with 56 setigers. The prostomium was fairly deeply notched, and often blackened by four small eyes. Notosetae were absent on the first setiger, and there were only ventral capillary setae. Branchiae start from setiger 7 and were absent in posterior region. Enlarged hooks (3-4) on the fifth setiger each had a lateral twisted spur, and on setiger 7 and there were 2-3 bidentate sigmoid hooded hooks in the ventral position. On the last setigers, the notosetae included one enlarged, yellow, curved hook. The pygidium was open and shaped like a suction cup, with a dorsal notch and, occasionally was dark in color. The chimneys created by Polydora hoplura on the outside surface of the abalone shell were initially located along the growing edge, although the worms were also capable of burrowing throughout the shell. When the infestation progressed, the shell-boring polychaetes tended to be evenly distributed within the shell. The chimneys were gray because of the accumulation of sediment (Figure 33). As the infestation became increasingly heavy, the abalone formed blisters by shell material secretion (i.e., conchiolin covered with nacre) that projected into the mantle cavity to isolate the shell-boring polychaetes. The shells were occasionally broken in the area of the blisters. In these cases, the abalone were extremely vulnerable and, in most cases, died (Figure 34). However, some of them were able to reconstruct the shell and, consequently, the shell became deformed. Treatment against shell-boring polychaetes The initial number of shell-boring polychaetes in the abalone shells was equal (P>0.05) in all replicates (Table 12). At the beginning of the trial, the results showed a positive association between the number of shell-boring polychaetes expelled and the concentration of mebendazole used (Figure 35). Afterwards, this relation disappeared and it was necessary to apply the drug 21 times over a term of 7 months (one-time 3-hours bath given for 3 consecutive days per month) to expel most shell-boring polychaetes from the abalone shells, regardless of the mebendazole concentration used.

104 – Experimento IV – Growth The size class of 40-59 mm in shell length had a higher specific growth rate than the size class of 60-75 mm in shell length, because these individuals were both younger and less infested (Tables 13 and 14). The SGR and WG rates did not differ statistically among all the mebendazole treatments and the control group (Tables 13 and 14). Thus, this result suggested that the mebendazole did not affect significantly the growth of the abalone during the period of study. However, it is worth noting that WG of the bigger abalones (60-79 mm in shell length) was positive in the control and negative in the various treatments. 106 – Experimento IV – In the current study, the efficacy rate of all the mebendazole doses tested (120 mg/l, 160 mg/l and 200 mg/l) in multiple applications were higher (EF 98.8-99.1%) than that found by Lleonart et al . (2003a) in a single application (200 mg/l and EF 32.5%) against the shell-boring polychaete Boccardia knoxi in the abalone species Haliotis rubra . Therefore it can be argued that when using mebendazole, which is a broad-spectrum anthelmitic drug (Krízová-Forstová et al ., 2011), multiple applications are more effective than single applications for eradicating shellboring polychaete pests. Although the growth rate results did not show statistical differences among the various mebendazole treatments and the control group, the bigger abalones (60-79 mm in shell length) lost weight. Taking into account that these animals were highly infested, this result confirms the observations of previous studies, which associated a high level of infestation with a loss in abalone condition (Lleonart et al ., 2003a; Simon et al ., 2006). This may be associated with the abalone tendency to respond to the infestation by depositing a layer of nacre over the worms to isolate them. The cost of continuously forming this layer is believed to result in a significant detriment to abalone condition (Handley, 1998). Our results suggested that the repeated application of mebendazole was more important than the concentration used. At this point, it would be interesting to test lower concentrations of the drug in multiple applications; however, it should be taken into consideration that the extended exposure to subtherapeutic dosages may result in mebendazole-resistant parasite populations (Buchmann et al ., 1992). It is also noteworthy that, among the causes of parasite resistance to drugs, one of them is the presence of active anthelmintics in the natural environment of the parasite (FAO, 2003; Krízová-Forstová et al ., 2011). This may be due to the disposal of drug residues directly into the environment along with the wastewater from the aquaculture facilities. In conclusion, the current study proved that using 120 mg/l mebendazole applied 21 times for 7 months (one-time 3-hours bath given for 3 consecutive days per month) eradicated the Polydora hoplura pest in approximately 99 % of highly infested abalones. However, it should be noted that the use of this type of chemical drug is a nonrenewable resource (Vial et al ., 1999; Geary et al ., 1999), in other words, if the parasites develop a resistance to the drug, the product will become unusable. Furthermore, total dependence on a single parasite control method has proved to be poorly sustainable and not profitable in the long term (Waller, 1997; Barger, 1999). Following the recommendations of FAO (2003), it is preferable to use the anthelmintic as a timely support for a rational and sustainable control strategy and not as the exclusive tool for such purposes.

108 – Experimento V – 4.5.1. INTRODUCTION The abalone found in the Canary Islands is Haliotis tuberculata coccinea Reeve and it is synonymous with Haliotis coccinea canariensis Nordsieck. This is a small subtropical native species with a maximum shell length of 8 cm (Bilbao et al ., 2010a). Recently, the local Government of the Canary Islands identified Haliotis tuberculata coccinea as a new species intended for the diversification of local commercial aquaculture. Currently, various techniques for the experimental culture of this species are being developed at the Instituto Canario de Ciencias Marinas (ICCM) (Peña, 1986; Viera et al ., 2005, 2011; Courtois de Vicose et al ., 2007, 2010; Bilbao et al ., 2010 a,b, 2011). An important aspect to be considered in the culture of any new species is the control of its reproduction. The natural spawning period of the temperate abalones species (i.e. Haliotis laevigata and Haliotis rubra ) is limited to a few months when the water temperature reaches its highest levels (Hanh, 1989). Under culture conditions, in order to extend the period of spawning throughout the entire year, some authors suggest a broodstock conditioning at a constant temperature (16 ºC and 17 ºC for Haliotis rubra and Haliotis laevigata , respectively) with an adequate diet during 10 and 26 weeks in order to induce spawning after this period (Grubert and Ritar, 2005; Freeman et al ., 2006). However, the reproductive cycle of tropical species (i.e. Haliotis asinina ) extends almost throughout the year, and a broodstock conditioning during 1535 days with an adequate diet and with the same water temperature of its natural environment may be enough to obtain natural spawning (Capinpin et al ., 1998; Bautista-Teruel et al ., 2001). In natural populations of Haliotis tuberculata coccinea , a subtropical species, the maximum reproductive activity occurs between August and February (with an average temperature of 1823 ºC in the Canary Island waters), although some mature individuals and some spawning events are also observed throughout the entire year (Bilbao et al ., 2010a). Under culture conditions, spontaneous spawning events have been sporadically observed (personal observation) around the lunar phase changes. However, these events are not regular. Thus, the most suitable procedure to control the spawning of Haliotis tuberculata coccinea and to schedule the tasks of a hatchery (i.e. the preparation of culture tanks for the culture of larvae and post-larvae) is the spawning induction of mature animals. The most common methods used to induce spawning in abalone species are the ultraviolet method (Kikuchi and Uki, 1974a) and the hydrogen peroxide method (Morse et al ., 1977). Peña (1986) adapted the hydrogen peroxide method for Haliotis tuberculata coccinea and achieved satisfactory results (100% spawning rates for males and females). However, the
– Experimento V – 115 once. The monthly spawning response in individuals with a VGI 3 was 19.3 ± 10.3 % in females and 30.3 ± 14.6 % in males during the period of study (Table 15). The monthly distribution of fecundity is given in the Table 15; we obtained a total of 10,438,775 eggs. The results showed that all individuals lost weight after 48 hours of the spawning induction. The weight loss of individuals that spawn was significantly greater (3.29 ± 1.98 g) than those failed to spawn (1.98 ± 1.81 g). Optimal sperm concentration The concentration of sperm had an obvious effect on fertilization success of Haliotis tuberculata coccinea with a polynomial correlation (P<0.05) between both parameters (Figure 41). The maximum fertilization rate (90.7 ± 0.56 %) was obtained with 10 5 spermatozoids / ml and this value was significant higher than those observed with others sperm concentrations. The fertilization rate obtained with 10 3 , 10 4 and 10 6 spermatozoids / ml was similar and was ranging from 64.9 to 72.33 %. The fertilization rates significantly decreased in the following sperm
116 – Experimento V – concentrations 10 2 , 10 1 and 10 7 spermatozoids / ml (50.93 ± 4.14, 34.7 ± 7.75 and 15.73 ± 1.35 % respectively). – Experimento V – 117 The effects of seven sperm concentrations (spermatozoids / ml) on the normal trochophore larvae are showed in the Figure 42. The sperm concentrations ranging from 10 1 to 10 4 spermatozoids / ml gave consistent normal trochophores (ranging from 90.73 to 73.83 %) with no significant differences between these densities. The percentage of normal trochophore decreased significantly at higher concentrations of 10 5 to 10 7 spermatozoids / ml (ranging from 55.03 to 1 %). We found a negative correlation (P<0.01) between both parameters. 4.5.4. DISCUSSION No individuals in the stage VGI 0 were found during the period of conditioning, whereas individuals in stages VGI 1, 2, and 3 were present throughout all the year. In addition, the histological study showed that stages VGI 1, 2 and 3 had simultaneously early, pre-mature and fully mature oocytes, although in different proportions. Therefore, it seems that under culture conditions, the reproduction of Haliotis tuberculata coccinea was continuous throughout the entire year (i.e. there was not a reproductive rest) and the ovary developed by synchronous groups. These results are consistent with those found by Bilbao et al . (2010a) for the same species in a natural population. Likewise, under culture conditions, Haliotis tuberculata coccinea responded to temperature variations and a higher percentage of individuals in stages VGI 2 and 3 was found between May and January, the period when the maximum water temperatures were recorded. Under culture conditions, the highest reproductive activity occurred three months earlier than in natural populations (Bilbao et al ., 2010a) and it may be due to a higher food availability for the broodstock observed in the present study. In fact, temperature and food availability seem to be the major factors affecting reproduction in different abalone species (Uki and Kikuchi, 1984; Hahn, 1989; Grubert and Ritar, 2005). Forty percent of induced females and 70.6% of induced males spawned more than once a year, suggesting that Haliotis tuberculata coccinea is a partial-spawning species as others tropical and warm water species such as Haliotis asinina and Haliotis discus hannai (Jebreen et al ., 2000; Fukazawa et al ., 2007). Fukuzawa et al . (2007) found in Haliotis discuss hannai that the mature oocyte cohort could be released in several natural spawning events within a period around a month (with a constant conditioning temperature of 20 ºC) before they are absorbed by the ovary and, the second oocyte cohort starts to develop after the first oocyte cohort disappears from the ovary. The time required to complete the oogenesis varies among the different abalone species and the culture conditions (i.e., temperature and conditioning period). Jebreen et al . (2000) show that the period of time necessary to complete the oogenesis in the tropical species Haliotis asinina is around one month and this species is capable to develop at least 3 cohorts of oocytes per year, without
118 – Experimento V – changing the water temperature. Fukuzawa et al . (2007) show that the oogenesis in Haliotis discus hannai (warm water species) is completed within a term of one month and this species is capable to develop at least 2 cohorts of oocytes per year with a broodstock conditioning at 20 ºC. Likewise, the time required to complete oogenesis in Haliotis rubra and Haliotis laevigata (a warm water species) is around three months with a broodstock conditioning at 16 ºC (Grubert and Ritar, 2005). Although the present study was not designed to determine the time required to complete the oogenesis in Haliotis tuberculata coccinea (i.e., no histological study of the gonad during conditioning period was performed), the specimens which responded to the spawning induction required between 2 and 4 months to respond again to the stimulus without changing the conditioning temperature. The broodstock used in this study came from a natural population and not all the individuals responded satisfactorily to the culture conditions. This fact is reflected in the number of specimens which did not reach stage VGI 3 during the study period (24 females and 15 males). In the present study, the broodstock conditioning was not performed individually. Thus, it is unknown whether all the animals ate the same amount of diet and the manner this fact affects the gonad maturation. However, all the animals had the same opportunity to be fed to satiation, since leftover food was found every week in the conditioning tanks. Therefore, it seems that the genetic profile of some individuals could result in a better adaptation to the culture conditions. This fact underlines the importance of the broodstock selection in order to ensure the conditioning success in an abalone hatchery. According to our findings, the right time to induce spawning in Haliotis tuberculata coccinea is when females have reached stage VGI 3, since the ovary has a significantly higher (P<0.05) amount of mature oocytes (52.93 ± 15.6 %) during this stage. Furthermore, it was also found that the ovary size was bigger during this stage (i.e., the area in the base of the conical appendage). However, the stage VGI 3 did not ensure the spawning induction success in Haliotis tuberculata coccinea , according to the results founded by Freeman et al . (2006) in Haliotis laevigata. The response rate of the spawning induction by the hydrogen peroxide method was lower than 100% founded by Peña (1986) in Haliotis tuberculata coccinea . In the experiment carried out by Peña (1986), the number of individuals was lower than those used in the present study (5 females and 5 males). Out of the total broodstock induced to spawn, 19.3 ± 10.3 % of females and 30 ± 14.6 % of males responded successfully and spawning events were obtained. In the reviewed bibliography, the spawning rate varies between 4 and 100 % according to the different abalone species, the period and/or the temperature of the conditioning and the spawning induction method used (Lleonart, 1992; Moss et al ., 1995; Moss, 1998; Mccormick and Brogan, 2003; Grubert and Ritar, 2005). Moss (1998) suggested that the stress caused during the
– Experimento V – 119 collection of the broodstock from a natural population may cause a gamete resorption and this fact may explain why it is difficult the induce spawning in Haliotis australis soon after the individuals are collected. However, he did not found good spawning rates after a 14-week broodstock conditioning at a constant temperature of 18 ºC (an average of 17 % for males and 10 % in females). In a previous study, Moss et al . (1995) also found low spawning rates in a broodstock batch of Haliotis iris , captured in a natural population and induced to spawn by the hydrogen peroxide method (15 % of females and 43 % of males). Despite the large number of females induced to spawn during this trial, no relationship was found between the number of eggs released and the animal size (i.e., shell length). Similar observations have been reported in other abalone species induced to spawn (Babcock and Keesing, 1999; Grubert and Ritar, 2005). On the contrary, Peña (1986) observed an exponential relationship between the number of eggs released and the shell length in Haliotis tuberculata coccinea . The results obtained in this experiment (i.e., low spawning rates and absence of a relationship between the fecundity rate and the animal size) may be due in part to the fact that the broodstock came from the natural environment and did not adapt well to the conditioning or to the induction method either and, on the other hand, to the generated stress in the animals related to the spawning induction tasks, which were performed on monthly basis (i.e., the induced animals which did not respond satisfactorily to the spawning induction lost weight), or to a combination of both reasons. The animals which spawned required between 2 and 4 months to be able to successfully respond again to the spawning induction. By making longer intervals between spawning induction operations (spawning inductions not performed on a monthly basis), on the one hand, it would entail a lower manipulation of the broodstock and would avoid stress, and on the other hand, the broodstock would have more time to produce a higher amount of mature oocytes. In this case, it may possible for the broodstock in stage VGI 3 to better respond to the spawning induction. In the fertilization trial, by increasing the sperm concentrations, the fertilization success increased rapidly until 90 % with 10 5 spermatozoids / ml. With higher sperm concentrations, the fertilization success rapidly declined. In the reviewed bibliography, the sperm concentrations necessary for the maximum fertilization rates vary among different abalone species and not all the studies reached 100 % fertilization rates (Babcock and Keesing, 1999; Baker and Tyler, 2001). In Haliotis discus hannai , 100% of fertilization rate was obtained with 10 5 spermatozoids / ml (Kikuchi and Uki, 1974c). In Haliotis tuberculata fertilization rates between 90 and 100 % are obtained within a range of 5x10 4 and 10 6 spermatozoids / ml (Clavier, 1992b; Baker and Tyler, 2001). The sperm concentrations between 5x10 3 and 10 5 spermatozoids / ml have been described as optimal in Haliotis rubra (Huchette et al ., 2004). Leighton and Lewis (1982) studied
120 – Experimento V – the optimal concentration and the subsequent normal development of embryo and larvae and concluded that, for Haliotis rufescens , Haliotis corrugata , Haliotis fulgens and Haliotis sorenseni , the optimal concentration is 10 6 spermatozoids / ml. In Haliotis asinina the maximum fertilization rate and a normal embryonic development is within a lower order of concentration (10 5 spermatozoids / ml), according to the results obtained by Encena et al . (1998). More recently, Luo et al . (2010) showed that the hybridization of the species Haliotis gigantea and Haliotis discus hannai requires a concentration of 10 7 spermatozoids / ml, with which they obtained the maximum fertilization rate and the highest percentage of normal larval development. In the present study, considering the results of the percentage of normal larvae development, it seems that the optimal sperm concentration for the fertilization of Haliotis tuberculata coccinea is 10 4 spermatozoids / ml. As found in other studies (Encena et al ., 1998; Baker and Tyler, 2001), the results obtained in this experiment suggest that the abnormal larval development is a consequence of polyspermy as, by increasing the sperm concentration, the percentage of normal larvae decreases. In fact, in the present trial, the dissolution of the egg membrane was observed with a concentration over 10 6 spermatozoids / ml. In other species (i.e. Haliotis rubra and Haliotis tuberculata ), the dissolution of the egg membrane was observed with a concentration over 10 7 spermatozoids / ml (Clavier, 1992b; Baker and Tyler, 2001; Huchette et al ., 2004). Polyspermy may occur during the finite time delay between initial egg fertilization and any polyspermy block activation and this event is increasingly likely as sperm suspensions are more concentrated (Styan, 1988). Blocks to polyspermy are common to many taxa (Togo et al ., 1995), but it remains unresolved which anti-polyspermy mechanisms occur in abalone species (Baker and Tyler, 2001). In the present study, with low sperm concentrations, low fertilization rates were obtained, according with the results founded by Baker and Tyler (2001) in Haliotis tuberculata ; these authors suggest that this fact is a result of the decreased probability of successful eggsperm encounters. Apart from the sperm concentration used in the fertilization tasks, some studies showed that the age of gametes is also important and it is recommended that the semen used is not older than 30 minutes of age after it was released (Baker and Tyler, 2001) and that the eggs should not be older than 2 hours of age after they were released in Haliotis asinina, Haliotis laevigata and Haliotis tuberculata (Encena et al ., 1998; Babcock and Keesing, 1999; Baker and Tyler, 2001). However, it seems that the contact time between gametes does not significantly affect the percentage of fertilization of Haliotis rufescens and Haliotis tuberculata and a contact time between 15 and 30 minutes is recommended for these species (Elbert and Houk, 1984; Babcock and Keesing, 1999).
– Experimento V – 121 In conclusion, this study showed that it is possible to obtain induced spawnings by the hydrogen peroxide method all the year round by conditioning Haliotis tuberculata coccinea broodstock using a mixed diet and without changing the water temperature. However, the results showed that not all the individuals used in this study (captured in the natural environment) had the same response to culture conditions or to the spawning induction either. Thus, it is important to keep a log of specimens which respond satisfactorily to the spawning induction and to select them in order to minimize the work and costs involved in the conditioning tasks in the hatchery. Likewise, the results suggested that in a hatchery (where the release of large number of eggs is expected using a spawning induction) inducing spawning on a monthly basis is not very suitable. The fertilization trial showed that, in Haliotis tuberculata coccinea , the optimal sperm concentration necessary to obtain good fertilization rates and a good percentage of normal larval development occurred with 10 4 spermatozoids / ml.

124 – Conclusiones – CONCLUSIONES 1. La reproducción de Haliotis tuberculata coccinea en la naturaleza se extendió a todo el año, aunque se observó una mayor actividad reproductora entre agosto y febrero, coincidiendo con las mayores temperaturas del agua. 2. El ovario de Haliotis tuberculata coccinea se desarrolló por grupos sincrónicos, es decir, en un ovario maduro se encontraron dos grupos de oocitos, uno de mayor tamaño que es expulsado en el acto de puesta y otro de menor tamaño que se desarrolla cuando ha sido expulsado y/o absorbido por el ovario, el de mayor tamaño. 3. Las hembras superiores a 55 mm en longitud de concha, provenientes de una población natural, contuvieron un número significativamente superior de huevos en las gónadas maduras. Este hecho sugiere el acondicionamiento de hembras de una talla igual o superior a 55 mm. 4. Dependiendo de la cantidad de fertilizante (amonio y fosfato), utilizado en el cultivo de la macroalga Gracilaria cornea , su composición proximal varió. Se logró un porcentaje de aproximadamente un 30 % de proteína con la adición de 33,3 mg / l d de amonio y 3,3 mg / l d de fosfato al cultivo. 5. La utilización del alimento Gracilaria cornea , con un alto contenido en proteína (aproximadamente un 30 %) favoreció el crecimiento (en términos de longitud de concha y ganancia en peso) de hembras reproductores de Haliotis tuberculata coccinea . Así mismo, aumentó la fecundidad relativa (mayor número de huevos por gramo de animal), y tanto el diámetro del huevo como del citoplasma del huevo fueron significativamente más grandes. Se observó además, que bajo este régimen de alimentación, el ovario maduro de Haliotis tuberculata coccinea contuvo un mayor contenido de ácido Araquidonico. 6. El uso de aceite de clavo como anestésico en ejemplares adultos de Haliotis tuberculata coccinea , fue adecuado a una concentración de 0,5 ml / l, disuelto previamente al 50% en alcohol absoluto. 7. La especie de poliqueto, que afectó al cultivo de Haliotis tuberculata coccinea en las instalaciones del Instituto Canario de Ciencias Marinas fue Polydora hoplura . 8. La utilización de baños múltiples de mebendazol (3 baños en tres días consecutivos cada mes, durante 7 meses), en una concentración de 120 mg / l, erradicó la infestación de Polydora hoplura en Haliotis tuberculata coccinea .
