Figure 4-1 A model of Ithe inflammatory mediators associated to ALS ALS is characterized by the degeneration and loss of motor neurons. Mutations in SOD1 were the first to be described as responsible for fALS. Currently, it is accepted that the non-cell autonomous theory where motor neuron lost is not only a result of intrinsic mechanisms that occur in neurons but also of neighbor glial cells. Toxic aggregates somehow released by motor neurons induced the activation of microglia mediated by TLR2 and CD14 receptors. Microglial cells might induce astrocytes activation by the production cytokines. Activation of both, microglia and astrocytes, amplify the motor neurons injury by induction of AP-1 y NF-κβ pathways and the secretion of pro-inflammatory cytokines and apoptosis induced molecules such as TNF-α y FASL. TNF-α e IL-1β. ATP released by dying motor neurons axacerbate microglia activation through the activation of the P2X7 purinergic receptors expressed in microglia. B) Pro-caspase-1 expression in treated cells analyzed by Western blot with cell lysates from primary microglial cells treated for 24 h with 2.5 µg/mL of TDP43 or mock. Blot is representative of 5 independent experiments (N=5). C) Quantitative determination by ELISA of IL-1b and TNF-a cytokine levels in supernatants from N13 (TNF-a) or primary microglial cells (IL-1b), 6 h or 24 h after treatment with 5, 2.5 or 1.25 μg/mL of TDP43 or mock, with or without pre-treatment with Bay11-7082. Bars are the mean of at least 3 independent experiments (N≥3), each one performed with duplicate samples. D) Quantitative determination by ELISA of secreted, activated IL-18 in culture supernatants of primary microglia 24 h after treatment with 2.5 or 5 µg/mL of TDP43 or mock. Absolute measured levels of IL-18 for TDP-43- treated cells ranged between 60 and 90 pg/mL between independent experiments. Bars are mean of six ! 84! independent experiments (N=6), each one performed with duplicate samples. E) Western blot analysis of cleaved IL-18 in supernatants from primary microglial cells collected 24 h after treatment with 5 µg/mL of TDP43 or mock. Recombinant murine IL-18 was used as a control in the same gel (right end). Bottom: band density quantification (relative values) from three independent experiments (N=3).
