Nuevas formulaciones de cementos de fosfato de calcio macroporosos para la regeneración ósea

Abstract

El objetivo del presente proyecto es la mejora de la inyectabilidad y de la cohesión de un cemento de fosfato de calcio macroporoso desarrollado en estudios anteriores. La introducción de la macroporosidad en este tipo de materiales tiene como finalidad mejorar la reabsorción y la integración del cemento en el tejido óseo. En aplicaciones quirúrgicas el cemento está en contacto con la sangre y los líquidos fisiológicos. Es necesario asegurar la cohesión de la pasta al entrar en contacto con estos líquidos para impedir toda reacción inflamatoria. Para mejorar la cohesión y la inyectabilidad del cemento se estudia la incorporación de un polímero soluble en agua (el alginato de sodio) en la fase líquida del cemento. En primer lugar se estudia la influencia del porcentaje de alginato de sodio en las características de la espuma. Se observa un gran aumento de la estabilidad de ésta pero se reduce de manera dramática la espumabilidad del líquido, lo que deja suponer una disminución de la porosidad del cemento a continuación. Posteriormente se estudia, vía un diseño factorial de experimentos (22 con un punto central), la influencia de dos variables (la relación L/P y el porcentaje de alginato de sodio) en distintas propiedades del cemento, como la cohesión, la inyectabilidad, la porosidad, la resistencia mecánica. La evaluación se lleva a cabo sobre muestras moldeadas con espátula o inyectadas con una jeringa a dos tiempos diferentes. Los cementos obtenidos se caracterizan por ensayos de cohesión y de inyectabilidad, microscopía óptica y electrónica, medidas de porosidad por picnometría de mercurio y porosimetría de mercurio, ensayos de resistencia a compresión. Comprobamos la mejora de la cohesión y de la inyectabilidad al añadir alginato de sodio pero observamos una disminución de la macroporosidad y del tamaño de los poros. Aunque no se pierda la macroporosidad, se nota un deterioro de la macroestructura del material si la pasta está inyectada a 5min, mientras que se mantiene la forma de los poros y la continuidad del material si la inyección se hace a 2min30.
